Abstract

This invention provides polydentate selective high affinity ligands (SHALs) that can be used in a variety of applications in a manner analogous to the use of antibodies. SHALs typically comprise a multiplicity of ligands that each binds different regions on the target molecule. The ligands are joined directly or through a linker thereby forming a polydentate moiety that typically binds the target molecule with high selectivity and avidity.

Claims (48)

1. A polydentate selective high affinity ligand (SHAL) comprising a first ligand and a second ligand,

wherein said first ligand and said second ligand bind to a first pocket and a second pocket, respectively, on the β-subunit of an HLA-DR10 surface antigen, said first and second pockets being two adjacent pockets flanking the R70 amino acid residue of the HLA-DR10 surface antigen, and

wherein said first ligand is linked to said second ligand by a linker comprising polyethylene glycol or a linker comprising lysine and polyethylene glycol, and

wherein said first ligand and second ligand is a peptide or a small organic molecule.

2. The SHAL of claim 1 , wherein said first ligand and said second ligand are selected from Table 4.

3. The SHAL of claim 1 , wherein said first ligand and said second ligand each is a small organic molecule.

4. The SHAL of claim 1 , wherein said first ligand is selected from Table 2, and said second ligand is selected from Table 3.

The SHAL of claim 1 , further comprising an effector attached to the SHAL, wherein the effector is selected from the group consisting of an epitope tag, an antibody, a second SHAL, a label, a cytotoxin, a liposome, a radionuclide, a drug, a prodrug, a viral particle, a cytokine, and a metal isotope.

6. The SHAL of claim 5 , wherein said epitope tag is selected from the group consisting of an avidin, and a biotin.

7. The SHAL of claim 5 , wherein said cytotoxin is selected from the group consisting of a Diphtheria toxin, a Pseudomonas exotoxin, a ricin, an abrin, and a thymidine kinase.

The SHAL of claim 5 , wherein said metal isotope is selected from the group consisting of 99 Tc, 203 Pb, 67 Ga, 68 Ga, 72 As, 111 In, 113m In, 97 Ru, 62 Cu, 64 Cu, 52 Fe, 52m Mn, 51 Cr, 186 Re, 188 Re, 77 As, 90 Y, 67 Cu, 169 Er, 121 Sn, 127 Te, 142 Pr, 143 Pr, 198 Au, 199 Au, 161 Tb, 109 Pd, 165 Dy, 149 Pm, 151 Pm, 153 Sm, 157 Gd, 159 Gd, 166 Ho, 172 Tm, 169 Yb, 175 Yb, 177 Lu, 105 Rh, and 111 Ag.

9. A pharmaceutical formulation said formulation comprising:

a pharmaceutically acceptable excipient and a SHAL of claim 1 .

10. The pharmaceutical formulation of claim 9 , wherein said formulation is a unit dosage formulation.

11. A pharmaceutical formulation said formulation comprising:

a pharmaceutically acceptable excipient and a SHAL of claim 5 .

12. The pharmaceutical formulation of claim 11 , wherein said formulation is a unit dosage formulation.

13. A method of inhibiting the growth or proliferation of a cancer cell that displays a Lym-1 epitope, said method comprising contacting said cell with a SHAL of claim 1 .

14. The method of claim 13 , wherein said cell is a metastatic cell.

15. The method of claim 13 , wherein said cell is a solid tumor cell.

16. A method of inhibiting the growth or proliferation of a cancer cell, said method comprising contacting said cell with a SHAL of claim 1 , wherein said SHAL is attached to a cytotoxin or a radionuclide.

17. The method of claim 16 , wherein said cell is a metastatic cell.

18. The method of claim 16 , wherein said cell is a solid tumor cell.

19. A method of detecting a cancer cell that displays a Lym-1 epitope, said method comprising:

contacting said cancer cell with a SHAL of claim 1 ,

wherein said SHAL is attached to a detectable label; and

detecting the presence or absence of said detectable label.

20. The method of claim 19 , wherein said detectable label is a selected from the group consisting of a gamma-emitter, a positron-emitter, an x-ray emitter, an alpha emitter, and a fluorescence-emitter.

21. A method of detecting a cell that displays a Lym-1 epitope, said method comprising:

contacting a cell with a SHAL of claim 1 , wherein said SHAL is attached to an epitope tag;

contacting said SHAL with a chelate comprising a detectable moiety whereby said chelate binds to said epitope tag thereby associating said detectable moiety with said cell; and

detecting said detectable moiety.

22. The method of claim 21 , wherein said detectable moiety is a radionuclide.

23. The method of claim 21 , wherein said detectable moiety is selected from the group consisting of a gamma-emitter, a positron-emitter, an alpha emitter, an x-ray emitter, and a fluorescence-emitter.

24. The method of claim 21 , wherein said cell is a cancer cell.

25. The method of claim 21 , wherein said detecting comprises external imaging.

26. The method of claim 21 , wherein said detecting comprises internal imaging.

The method of claim 21 , wherein said detectable moiety comprises a metal isotope selected from the group consisting of 99 Tc, 203 Pb, 67 Ga, 68 Ga, 72 As, 111 In, 113m In, 97 Ru, 62 Cu, 64l Cu, 52 Fe, 52m Mn, 51 Cr, 186 Re, 188 Re, 77 As, 90 Y, 67 Cu, 169 Er, 121 Sn, 127 Te, 142 Pr, 143 Pr, 198 Au, 199 Au, 161 Tb, 109 Pd, 165 Dy, 149 Pm, 151 Pm, 153 Sm, 157 Gd, 159 Gd, 166 Ho, 172 Tm, 169 Yb, 175 Yb, 177 Lu, 105 Rh, and 111 Ag.

28. The method of claim 21 , wherein said chelate comprises DOTA.

29. The method of claim 21 , wherein said epitope tag is an avidin or a biotin.

30. A kit comprising a SHAL of claim 1 and an effector selected from the group consisting of an epitope tag, an antibody, a second SHAL, a label, a cytotoxin, a liposome, a radionuclide, a drug, a prodrug, a viral particle, a cytokine, and a metal isotope.

31. The kit of claim 30 , wherein said SHAL is in a pharmacologically acceptable excipient.

32. A polydentate selective high affinity ligand (SHAL) comprising a first ligand and a second ligand, wherein said first ligand is linked to said second ligand by a linker comprising polyethylene glycol or a linker comprising lysine and polyethylene glycol, and wherein said first and second ligands are identified by a method comprising:

retrieving, by a computer, the three-dimensional structure of a first pocket and a second pocket on the β-subunit of an HLA-DR10 surface antigen, said first and second pockets being two adjacent pockets flanking the R70 amino acid residue of the HLA-DR10 surface antigen,

retrieving the three-dimensional structures of a plurality of candidate molecules,

determining, for each of said candidate molecules, whether the three-dimensional structure of the candidate molecule matches, spatially, to either of said pockets, and

identifying a first ligand and a second, from the candidate molecules, that match said first pocket and second pocket, respectively.

The SHAL of claim 32 , further comprising an effector attached to the SHAL, wherein the effector is selected from the group consisting of an epitope tag, an antibody, a second SHAL, a label, a cytotoxin, a liposome, a radionuclide, a drug, a prodrug, a viral particle, a cytokine, and a metal isotope.
