Abstract

The present invention relates to a neurotrophic peptide having an amino acid sequence of VGDGGLFEKKL (SEQ ID NO:1), EDQQVHFTPTEG (SEQ ID NO:2) or IPENEADGMPATV (SEQ ID NO:3), and comprising an adamantyl group at the C- and/or N-terminal end.

Claims (27)

1. A neurotrophic peptide

consisting of the sequence VGDGGLFEKKL (SEQ ID NO: 1) and

a cycloalkyl or polycycloalkyl group bound to either the C- or N-terminal end of said sequence.

2. The peptide of claim 1 , wherein the cycloalkyl or polycycloalkyl group is an adamantyl group.

3. The peptide of claim 2 , wherein the adamantyl group is adamantyl-L-glycine, adamantyl-L-alanine, an alkylated adamantane group, an isomer of an adamantine, or a di- or triadamantane group.

4. A pharmaceutical composition, comprising:

a peptide fragment consisting of a sequence selected from the group consisting of GDGGLFEK (SEQ ID NO: 5), GLFEKKLW (SEQ ID NO: 6), VGDG (SEQ ID NO: 7), DGGL (SEQ ID NO: 9), and GGLF (SEQ ID NO: 10); and

a cycloalkyl or polycycloalkyl group bound to either the C- or N-terminal end of the fragment.

5. The composition of claim 4 , wherein the cycloalkyl or polycycloalkyl group is an adamantyl group.

6. The composition of claim 5 , wherein the adamantyl group is adamantyl-L-glycine, adamantyl-L-alanine, an alkylated adamantane group, an isomer of an adamantine, or a di- or triadamantane group.

7. The composition of claim 4 , further comprising a pharmaceutically acceptable excipient and/or carrier.

8. The composition of claim 4 , wherein the composition is suitable for intravenous, intramuscular, spinal, epidural, transdermal, subcutaneous, parenteral, intranasal, mucosal, oral, enteral, and/or rectal administration.

9. The composition of claim 4 , further comprising a slow release carrier.

10. The composition of claim 4 , wherein the peptide fragment is dosed in an amount between 0.1 μg/g to 100 mg/g.

11. The composition of claim 10 , wherein the peptide fragment is dosed in an amount between 1 μg to 80 mg/g.

12. A method of treating a subject comprising:

obtaining a pharmaceutical composition comprising a peptide fragment consisting of a sequence selected from the group consisting of GDGGLFEK (SEQ ID NO: 5), GLFEKKLW (SEQ ID NO: 6), VGDG (SEQ ID NO: 7), DGGL (SEQ ID NO: 9), and GGLF (SEQ ID NO: 10) and a cycloalkyl or polycycloalkyl group bound to either the C- or N-terminal end of the fragment; and

providing the composition to a subject;

wherein learning-memory capacity is improved in the subject.

13. The method of claim 12 , wherein the composition further comprises at least one additional pharmaceutically active component.

14. The method of claim 12 , wherein the composition is provided intravenously, intramuscularly, spinally, epidurally, intranasally, mucosally, transdermally, subcutaneously, parenterally, orally, enterally, or rectally.

15. The method of claim 12 , wherein the composition a slow release formulation.

16. The method of claim 12 , wherein the composition comprises the peptide fragment in an amount between 0.1 μg/g to 100 mg/g.

17. The method of claim 16 , wherein the composition comprises the peptide fragment in an amount between preferably 1 μg to 80 mg/g.

18. The method of claim 17 , wherein the peptide fragment is administered to the subject at a dose of 0.1 μg/kg to 20 mg/kg body weight.

19. The method of claim 18 , wherein the peptide fragment is administered to the subject at a dose of 0.5 μg/kg to 10 mg/kg body weight.

20. The method of claim 12 , wherein the subject is a human.
