Abstract

A method for constructing a compound of immunologically modified nanotubes and method for using the compound to deliver immunoadjuvants to tumor cells and to produce targeted, synergistic photophysical and immunological reactions for cancer treatment. To prepare the immunologically modified nanotubes, carbon nanotubes are dissolved in a solution of glycated chitosan, an immunostimulant, hence using glycated chitosan as a surfactant for rendering the aqueous solution of nanotubes stable. The compound can be used for treatment of cancer. The method includes steps of intratumorally administering immunologically modified nanotubes and administering laser irradiation of the target tumor. The nanotube serves as a carrier to deliver immunoadjuvants to the tumor cells and serves as a light-absorbing agent in a cell body of a tumor in a host. Upon laser irradiation of target tumor cells, immunologically modified nanotubes inside the tumor cells can produce spatially and temporally synchronized photothermal and immunological reactions for cancer treatment.

Claims (19)

1. A method of photoimmunological therapy for inducing tumor cell destruction and anti-tumor immune responses comprising:

internalizing immunologically modified nanostructures in a body of the tumor cell in a host, said immunologically modified nanostructures comprised of single-walled carbon nanotubes bound to glycated chitosan;

exposing said tumor cell to either continuous near-infrared light or radiofrequency radiation for providing thermal damage or stress, thereby leading to death of the tumor cell and exposure of tumor antigens of said tumor cell to an immune system of said host for promoting anti-tumor immune responses;

wherein said glycated chitosan solubilizes and stabilizes said nanotubes;

wherein said steps of internalizing and exposing induce a synergistic, spatially and temporally synchronized photothermal-immunological anti-tumor response using the immunologically modified nanostructures as an exogenous light-absorbing agent and the immunological stimulant for immune stimulation and activation.

2. The method according to claim 1 wherein:

said single-walled carbon nanotubes are comprised of gold.

3. The method according to claim 1 wherein:

said nanostructures have an absorption peak in a range of 700 to 1500 nm.

4. The method according to claim 3 wherein:

said nanostructures have an absorption peak with a wavelength having a range of 800 to 1200 nm.

5. The method according to claim 1 wherein:

said near-infrared light is produced by a 980-nm laser.

6. The method according to claim 5 wherein:

said near-infrared light is produced by a laser having a laser power density in the ranges of 0.1 to 5 W/cm 2 , and administered with an irradiating duration of 5 to 60 minutes.

7. The method according to claim 6 wherein:

said near-infrared light is produced by a laser with a power density of 0.75 to 2 W/cm 2 .

8. The method according to claim 6 wherein:

said near-infrared light is administered with an irradiating duration of 10 to 30 minutes.
