Abstract

Silk is purified to eliminate immunogenic components (particularly sericin) and is used to form fabric that is used to form tissue-supporting prosthetic devices for implantation. The fabrics can carry functional groups, drugs, and other biological reagents. Applications include hernia repair, tissue wall reconstruction, and organ support, such as bladder slings. The silk fibers are arranged in parallel and, optionally, intertwined (e.g., twisted) to form a construct; sericin may be extracted at any point during the formation of the fabric, leaving a construct of silk fibroin fibers having excellent tensile strength and other mechanical properties.

Claims (18)

1. A method for forming a biocompatible, implantable, substantially sericin free biodegradable fabric from not previously dissolved silk, the method comprising the steps of:

(a) aligning not previously dissolved silk fibroin fibers in parallel or intertwined with other not previously dissolved silk fibroin fibers to form a yarn,

(b) before and/or after aligning the not previously dissolved silk fibroin fibers substantially removing sericin from the not previously dissolved silk fibroin fibers without dissolving the silk fibroin fibers and without substantially altering the native structure of the silk fibroin fibers, by extracting the sericin from the silk fibrin fibers using a Na 2 CO 3 and soap solution at a temperature between 37° C. and 94° C. for about 30 minutes to an hour and

(c) organizing a plurality of the yarns thereby forming the biocompatible, implantable, substantially sericin free, biodegradable fabric from not previously dissolved silk fibroin fibers,

wherein cells for proliferating and differentiating into a tissue have not been added to or absorbed into the fabric.

2. The method of claim 1 , wherein sericin is substantially removed by extraction using a temperature at or below 90° C.

3. The method of claim 1 , further comprising rinsing the sericin-extracted fibroin fibers with water.

4. The method of claim 1 , wherein the sericin-extracted fibroin fibers comprise fibroin fibers obtained from Bombyx mori silkworm.

5. The method of claim 1 , further comprising in step (c) organizing the plurality of yarns non-randomly to form a knit or woven fabric.

6. The method of claim 1 , further comprising in step (c) organizing the plurality of yarns randomly to form a non-woven fabric.

7. A method for making a biocompatible, implantable, substantially sericin free silk fabric from not previously dissolved silk, the method comprising the steps of:

(a) aligning not previously dissolved silk fibers in parallel or intertwined with other also not previously dissolved silk fibers to thereby form a yarn;

(b) repeating step (a) to obtain a plurality of the yarns;

(c) by a knit, woven or non-woven process organizing the plurality of the yarns to thereby make the biocompatible, implantable substantially sericin free silk fabric from not previously dissolved silk, and;

(d) before, during and/or after any of the steps (a) to (c) contacting the not previously dissolved silk fibers with a Na 2 CO 3 and soap solution at a temperature between 37° C. and 94° C. for about 30 minutes to an hour to thereby extract substantially all the sericin from the not previously dissolved silk fibers without substantially dissolving the silk fibers,

wherein cells for proliferating and differentiating into a tissue have not been added to or absorbed into the silk fabric.

8. The method of claim 7 , further comprising rinsing the sericin extracted silk fibers with water.

9. The method of claim 7 , wherein the silk fibers for the aligning step are obtained from a Bombyx mori silkworm.
