Abstract

The invention provides delivery systems comprised of stabilized multilamellar vesicles, as well as compositions, methods of synthesis, and methods of use thereof. The stabilized multilamellar vesicles may comprise prophylactic, therapeutic and/or diagnostic agents.

Claims (15)

1. A multilamellar lipid vesicle having covalent crosslinks between lipid bilayers, wherein at least two lipid bilayers in the multilamellar lipid vesicle are covalently crosslinked to each other through headgroups that react with covalent crosslinkers to form the covalent crosslinks between lipid bilayers.

2. The multilamellar lipid vesicle of claim 1 , wherein the vesicle comprises phosphocholine.

3. The multilamellar lipid vesicle of claim 1 , wherein the vesicle comprises phosphoglycerol.

4. The multilamellar lipid vesicle of claim 1 , wherein the headgroups are maleimide headgroups.

5. The multilamellar lipid vesicle of claim 1 , wherein the vesicle comprises an antigen.

6. The multilamellar lipid vesicle of claim 1 , wherein the vesicle comprises an adjuvant.

7. The multilamellar lipid vesicle of claim 1 , wherein the vesicle comprises a protein.

8. The multilamellar lipid vesicle of claim 1 , wherein the vesicle comprises an agent encapsulated within the vesicle.

9. The multilamellar lipid vesicle of claim 1 , wherein the vesicle comprises an agent encapsulated between the lipid bilayers.

10. The multilamellar lipid vesicle of claim 1 , wherein the vesicle is conjugated to polyethylene glycol (PEG).

11. The multilamellar lipid vesicle of claim 1 , wherein the vesicle comprises DOPC, DOPG and a maleimide functionalized lipid.

12. The multilamellar lipid vesicle of claim 11 , wherein the maleimide functionalized lipid is a maleimide functionalized phosphoethanolamine.

13. A composition comprising the multilamellar lipid vesicle of claim 1 and a pharmaceutically acceptable carrier.

14. A composition comprising the multilamellar lipid vesicle of claim 1 and an excipient suitable for lyophilization.

15. A composition comprising the multilamellar lipid vesicle of claim 14 , wherein the excipient suitable for lyophilization comprises sucrose.
