Abstract

A transmission electron microscope includes an electron beam source to generate an electron beam. Beam optics are provided to converge the electron beam. An aberration corrector corrects the electron beam for at least a spherical aberration. A specimen holder is provided to hold a specimen in the path of the electron beam. A detector is used to detect the electron beam transmitted through the specimen. The transmission electron microscope may operate in an incoherent mode and may be used to locate a sequence of objects on a molecule.

Claims (29)

1. A transmission electron microscope comprising:

an electron beam source to generate an electron beam;

beam optics to converge the electron beam;

a specimen holder to hold a specimen in the path of the electron beam; and

a detector to detect the electron beam transmitted through the specimen,

wherein the transmission electron microscope is adapted to generate electron beams that are incoherent in relation to one another at different times to operate in an incoherent mode.

2. The transmission electron microscope of claim 1 , wherein the transmission electron microscope is adapted to alter the energy of the electron beam source.

3. The transmission electron microscope of claim 2 , wherein the transmission electron microscope is adapted to scan a focus of the electron beam simultaneously that the energy of the electron beam source is altered.

The transmission electron microscope of claim 1 , wherein the beam optics comprise an objective lens having an aperture, and wherein the transmission electron microscope is adapted to rock or scan the electron beam within the angular range that is collected by the aperture of the objective lens.

5. The transmission electron microscope of claim 1 , further comprising a tilting prism to alternately flip the electron beam between two mirror angles.

6. A transmission electron microscope comprising:

an electron beam source to generate an electron beam;

beam optics to converge the electron beam, the beam optics defining an optic axis of the microscope along which there is substantial cylindrical symmetry of the beam optics;

a specimen holder to hold a specimen in the path of the electron beam; and

a detector to detect the electron beam transmitted through the specimen,

wherein the transmission electron microscope is adapted to generate electron beams that are incoherent in relation to one another at different times to operate in an incoherent mode.

7. The transmission electron microscope of claim 6 , wherein the transmission electron microscope is adapted to alter the energy of the electron beam source.

8. The transmission electron microscope of claim 7 , wherein the transmission electron microscope is adapted to scan a focus of the electron beam simultaneously that the energy of the electron beam source is altered.

9. The transmission electron microscope of claim 6 , wherein the beam optics comprise an objective lens having an aperture, and wherein the transmission electron microscope is adapted to rock or scan the electron beam within the angular range that is collected by the aperture of the objective lens.

10. The transmission electron microscope of claim 6 , further comprising a tilting prism to alternately flip the electron beam between two mirror angles.

11. A method of locating a sequence of objects on a molecule, the method comprising:

generating electron beams that are incoherent in relation to one another at different times to operate the electron beams in an incoherent mode;

converging the electron beams;

holding a specimen in the path of the electron beams; and

detecting the electron beams transmitted through the specimen to locate a sequence of objects on a molecule.

12. The method of claim 11 , comprising altering the energy of the electron beam.

13. The method of claim 12 , comprising scanning a focus of the electron beam simultaneously with altering the energy of the electron beam.

14. The method of claim 11 , comprising rocking or scanning the electron beam within the angular range that is collected by an aperture of an objective lens.

15. The transmission electron microscope of claim 11 , comprising alternately flipping the electron beam between two mirror angles.
