Method and probe set for detecting cancer
Methods for detecting cancer that include hybridizing a set of chromosomal probes to a biological sample obtained from a patient, and identifying if aneusomic cells are present in a selected subset of cells obtained from the biological sample are described. A set of chromosomal probes and kits for detecting cancer that include sets of chromosomal probes, are also described.
1. A method of detecting aneusomic cells, said method comprising:
a) obtaining a kit comprising a set of chromosomal probes consisting of centromeric chromosomal probes to chromosomes 3, 7, and 17 or centromeric chromosomal probes to chromosomes 3, 7, and 17 and a locus-specific probe to 9p21;
b) hybridizing said set of chromosomal probes of said kit to a biological sample from a subject; and
c) detecting aneusomic cells by examining the hybridization pattern of said set of chromosomal probes in said biological sample.
2. The method of claim 1 , comprising selecting a plurality of cytologically abnormal cells from said biological sample and detecting aneusomic cells in said selected cells by examining the hybridization pattern of said set of chromosomal probes in each of said selected cells.
3. The method of claim 2 , wherein cells are selected by nuclear morphology.
4. The method of claim 2 , wherein cells are selected by nuclear size.
5. The method of claim 2 , wherein cells are selected by nuclear shape.
6. The method of claim 1 , wherein said chromosomal probes are fluorescently labeled.
7. The method of claim 1 , wherein said biological sample is selected from the group consisting of urine, blood, cerebrospinal fluid, pleural fluid, sputum, peritoneal fluid, a bladder washing, oral washing, tissue sample, touch prep, and fine-needle aspirate.
8. A method of performing fluorescence in situ hybridization, said method comprising:
a) hybridizing a set of chromosomal probes consisting of centromeric probes to chromosomes 3, 7, and 17 to a biological sample from a subject; and
b) detecting aneusomic cells by examining the hybridization pattern of said set of chromosomal probes in said biological sample.
9. The method of claim 8 , comprising selecting a plurality of cytologically abnormal cells from said biological sample and detecting aneusomic cells in said selected cells by examining the hybridization pattern of said set of chromosomal probes in each of said selected cells.
10. The method of claim 9 , wherein cells are selected by nuclear morphology.
11. The method of claim 9 , wherein cells are selected by nuclear size.
12. The method of claim 9 , wherein cells are selected by nuclear shape.
13. The method of claim 8 , wherein said chromosomal probes are fluorescently labeled.
14. The method of claim 8 , wherein said biological sample is selected from the group consisting of urine, blood, cerebrospinal fluid, pleural fluid, sputum, peritoneal fluid, a bladder washing, oral washing, tissue sample, touch prep, and fine-needle aspirate.
