Abstract

Methods for introgressing an allele of interest of a locus associated with a yield trait into Zea mays germplasm are provided. In some embodiments, the methods include providing a Zea mays plant that contains an allele of interest of a locus associated with a yield trait, wherein the locus associated with the yield trait is identifiable by PCR amplification of a Zea mays nucleic acid with a pair of oligonucleotides primers as disclosed herein, and introgressing the allele of interest into Zea mays germplasm that lacks the allele. Also provided are methods for identifying Zea mays plants that contain at least one allele associated with improved yield, improved maize plants, elite Zea mays plants, biomass produced from improved Zea mays plants, isolated and purified genetic markers, and compositions that include an amplification primer pair capable of amplifying a Zea mays nucleic acid to generate a Zea mays marker amplicon.

Claims (12)

1. A method of introgressing an allele of interest associated with increased digestibility into a corn germplasm that lacks the allele, the method comprising:

(a) providing a population of corn plants;

(b) genotyping at least one corn plant in the population;

(c) selecting one or more genotyped corn plants from (b) based on the presence of an allele of interest associated with increased digestibility comprising a genomic sequence consisting of either an A at nucleotide position 116 of SEQ ID NO: 16 or an A at nucleotide position 33 of SEQ ID NO: 116; and

(d) introgressing the allele of interest of (c) into corn germplasm that lacks the allele.

The method of claim 1 , wherein the allele of interest associated with increased digestibility comprising a genomic sequence consisting of either SEQ ID NO: 16 or SEQ ID NO: 116 is identified by PCR amplification of corn genomic DNA with a pair of oligonucleotide primers consisting of primer pair 6 represented by a primer comprising SEQ ID NO: 17 and a primer comprising SEQ ID NO: 18.

3. The method of claim 1 , wherein the allele of interest, associated with increased digestibility, positively correlates with improved starch digestibility or negatively correlates with an improved protein associated trait.

4. A method of selecting a corn plant with increased digestibility in a plant breeding process, the method comprising:

(a) providing a population of corn plants;

(b) genotyping at least one corn plant in the population; and

(c) selecting one or more genotyped corn plants from (b) based on the presence of an allele of interest associated with increased digestibility comprising a genomic sequence consisting of either an A at nucleotide position 116 of SEQ ID NO: 16 or an A at nucleotide position 33 of SEQ ID NO: 116.

5. The method of claim 4 , wherein the allele of interest, associated with increased digestibility, positively correlates with improved starch digestibility or negatively correlates with an improved protein associated trait.
