View Patent ↗In accordance with certain embodiments of the present disclosure, a kit is described. The kit includes primed living cells joined to and at least partially within a three-dimensional hydrogel structure and an isolated polypeptide having the carboxy-terminal amino acid sequence of an alpha Connexin, or a conservative variant thereof, wherein the polypeptide does not include the full length alpha Connexin protein.
1. A kit comprising: primed living bone-marrow stromal cells joined to and at least partially within a three-dimensional flexible hydrogel structure, the hydrogel structure comprising collagen and having a toroid shape; and an isolated polypeptide comprising the carboxy-terminal amino acid sequence of an alpha Connexin protein, or a conservative variant thereof, wherein the polypeptide does not comprise the full length alpha Connexin protein and the carboxy-terminal amino acid sequence is capable of inhibiting gap junction formation.
2. A method of forming a toroid, hydrogel structure comprising placing primed living bone marrow stromal cells on a three-dimensional, flexible hydrogel structure, the hydrogel structure comprising collagen, and permitting epithelialization of the cells in which the cells attach to the surface of the hydrogel structure, undergo epithelial mesenchymal transition and penetrate the surface of the hydrogel structure to form a toroid shape.
