Transgene delivering retrovirus targeting collagen exposed at site of tissue injury
A viral or non-viral vector particle having a modified viral surface protein wherein the viral surface protein is modified to include a targeting polypeptide including a binding region which binds to an extracellular matrix component. Such vector particles are useful in delivering genes encoding therapeutic agents to cells located at the site of an exposed extracellular matrix component.
1. A method of treating a disease associated with exposition of collagen of a host, comprising: directly or intravenously delivering to the host a retroviral particle comprising:
(a) a modified viral envelope protein wherein the viral envelope protein includes a receptor binding region, wherein the receptor binding region is modified to include a collagen binding domain, and
(b) at least one polynucleotide encoding a therapeutic polypeptide;
wherein the viral particle binds to exposed collagen and expresses the therapeutic polypeptide, whereby said disease is treated.
2. The method of claim 1 , wherein said disease is a vascular disorder or a lesion.
3. The method of claim 2 , wherein said vascular disorder is a cardiovascular disease, thrombosis, or neointima formation.
4. The method of claim 2 , wherein said lesion is a vascular lesion or an ulcerative lesion.
5. The method of claim 1 , wherein the modified viral envelope protein is a modified retroviral envelope protein.
6. The method of claim 1 , wherein the collagen binding domain is a von Willebrand Factor functional domain.
7. The method of claim 1 , wherein the collagen is collagen Type I or collagen Type IV.
8. A method of treating a disease in a host, wherein the disease is an inflammation or a connective tissue disorder, comprising: directly or intravenously delivering to the host a retroviral particle comprising:
(a) a modified viral envelope protein wherein the viral envelope protein includes a receptor binding region, wherein the receptor binding region is modified to include a collagen binding domain; and
(b) at least one polynucleotide encoding a therapeutic polypeptide;
wherein the viral particle binds to collagen exposed at the site of the disease and expresses the therapeutic polypeptide, whereby said disease is treated.
9. The method of claim 8 , wherein the connective tissue disorder is a disorder of a ligament, tendon, or cartilage.
10. The method of claim 8 , wherein the modified viral envelope protein is a modified retroviral envelope protein.
11. The method of claim 8 , wherein the collagen binding domain is a von Willebrand Factor functional domain.
12. The method of claim 8 , wherein the collagen is collagen Type I or collagen Type IV.
13. A retroviral particle for treating a disease in a host, comprising a modified viral envelope protein, wherein the modified viral envelope protein includes a targeting polypeptide comprising a binding region which binds to collagen and at least one therapeutic polypeptide for treating the disease in the host.
14. The retroviral particle of claim 13 , wherein the collagen binding domain is a von Willebrand Factor functional domain.
15. The retroviral particle of claim 13 , wherein the collagen is collagen Type I or collagen Type IV.
16. The retroviral particle of claim 13 , wherein the therapeutic polypeptide expresses a cell cycle control agent, a growth factor, a cytokine, an anticoagulant, an anti-platelet agent, an anti-inflammatory agent, a tumor suppressor protein, a clotting factor, a cystic fibrosis transmembrane conductance regulator (CFTR) protein, or a negative selective marker.
17. The retroviral particle of claim 13 , wherein the disease is a vascular disorder, a cardiovascular disease, cirrhosis of the liver, a connective tissue disorder, a tumor, a vascular lesion, an ulcerative lesion, an inflammation, thrombosis or neointima formation.
18. The retroviral particle of claim 13 , wherein the disease is a tumor or a vascular disorder.
