Abstract

Amylin derivatives, pharmaceutical compositions containing such derivatives, as well as methods of treating diabetes and hyperglycaemia are disclosed.

Claims (27)

1. A derivative of amylin, which is a human amylin analogue, wherein the human amylin analogue comprises an amino acid sequence of formula 1:

Xaa 1 -Cys- Xaa 3 -Thr-Ala-Thr-Cys-Ala-Thr-Gln-Arg-Leu-Ala- Xaa 14 -Phe-Leu- Xaa 17 -Xaa 18 -Ser-Ser-Xaa 21 -Asn- Phe- Xaa 24 - Xaa 25 - Xaa 26 -Leu-Xaa 28 -Xaa 29 -Thr- Xaa 31 -Val-Gly-Ser-Asn-Thr-Tyr  Formula (1) (SEQ ID No:2)

wherein

Xaa 1 is deleted or independently selected from Lys, Arg, His and Glu;

Xaa 3 is independently selected from Asn and Lys;

Xaa 14 is independently selected from Glu, Asn, Gln and Asp;

Xaa 17 is independently selected from His, Ser, Gly, Arg and Pro;

Xaa 18 is independently selected from His or Arg;

Xaa 21 is independently selected from Asp, Asn and Gln;

Xaa 24 is independently selected from Glu and Gly;

Xaa 25 is independently selected from Ala and Pro;

Xaa 26 is independently selected from Pro and Ile;

Xaa 28 is independently selected from Ser and Pro;

Xaa 29 is independently selected from Ser and Pro;

Xaa 31 is independently selected from Glu and Asn;

the C-terminal may optionally be derivatized as an amide;

wherein a substituent is connected to the amino acid residue Xaa 1 or to Cys in position 2, which substituent comprises an albumin binding moiety.

2. A pharmaceutical composition comprising a derivative according to claim 1 , and a pharmaceutically acceptable excipient.

3. A process for preparing a pharmaceutical composition comprising mixing a derivative according to claim 1 with at least one pharmaceutically acceptable excipient.

4. The derivative according to claim 1 , wherein the amino acids in position 25, 28 and 29 are Pro.

5. The derivative according to claim 1 , wherein the substituent comprises a linker.

The derivative according to claim 5 , wherein the linker comprises amino acids selected from the group consisting of γGlu, Arg, γGlu-Arg, γGlu-His, Trx-γGlu, γGlu-His, Glu-Lys, Glu-Glu, Glu-Arg, γGlu-His-His, γGlu-Arg-His, γGlu-His-Arg, γGlu-Glu-Arg, Glu-Glu-Arg, Glu-Lys-Arg, γGlu-Glu-His-His (SEQ ID: 36), γGlu-Glu-His-Arg (SEQ ID: 37), Glu-Glu-Arg-Glu (SEQ ID: 38) and Glu-Glu-Glu-Glu (SEQ ID: 39).

7. The derivative according to 6 , wherein the linker comprises —C(O)—(CH 2 ) l —O—[CH 2 CH 2 —O] m —(CH 2 ) p -[NHC(O)—(CH 2 ) l —O—[(CH 2 ) n —O] m —(CH 2 ) p ] q —NH— wherein l, m, n, and p independently are 1-7, and q is 0-5.

8. The derivative according to claim 1 , wherein the albumin binding moiety is an acyl group selected from the group comprising HOOC(CH 2 ) s CO—, wherein s is an integer from 17 to 21.

9. The derivative according to claim 8 , wherein s is 18.

10. The derivative according to claim 8 , wherein s is 19.

11. The derivative according to claim 8 , wherein s is 20.
