Abstract

The present invention provides novel stabilized crosslinked compounds having secondary structure motifs, libraries of these novel compounds, and methods for the synthesis of these compounds libraries thereof. The synthesis of these novel stabilized compounds involves (1) synthesizing a peptide from a selected number of natural or non-natural amino acids, wherein the peptide comprises at least two moieties capable of undergoing reaction to promote carbon-carbon bond formation; and (2) contacting the peptide with a reagent to generate at least one crosslinker and to effect stabilization of a secondary structure motif. The present invention, in a preferred embodiment, provides stabilized p53 donor helical peptides. Additionally, the present invention provides methods for disrupting the p53/MDM2 binding interaction comprising (1) providing a crosslinked stabilized a-helical structure; and (2) contacting the crosslinked stabilized a-helical structure with MDM2.

Claims (25)

1. A method for synthesizing a stabilized helical peptide comprising:

a) synthesizing a peptide from a selected number of amino acids, wherein the peptide comprises at least two alpha, alpha-disubstituted amino acids, further wherein each alpha, alpha-disubstituted amino acid comprises a moiety capable of undergoing reaction to form a carbon-carbon bond;

wherein each of the alpha, alpha-disubstituted amino acids comprises at the alpha position a methyl group; and

b) contacting the peptide with a reagent to generate at least one cross-linker connecting at least two moieties to result in a stabilized helical peptide,

wherein the at least two alpha, alpha disubstituted amino acids are cross-linked.

2. A method for synthesizing a stabilized peptide comprising:

a) synthesizing a peptide from a selected number of amino acids, wherein the peptide comprises at least two alpha, alpha-disubstituted amino acids, further wherein each alpha, alpha-disubstituted amino acid comprises a moiety capable of undergoing reaction to form a carbon-carbon bond;

wherein each of the alpha, alpha-disubstituted amino acids is an alpha-methyl, alpha-alkenyl amino acid; and

b) contacting the peptide with a reagent to generate at least one cross-linker connecting at least two moieties to result in a stabilized peptide.

3. The method of claim 2 , wherein the alpha-methyl, alpha-alkenyl amino acid comprises a pentenyl or octenyl side chain.

4. The method of claim 1 , wherein the length of the cross-linked moieties between the two alpha positions is 7 or 8 carbon atoms.

5. The method of claim 1 , wherein the length of the cross-linked moieties between the two alpha positions is 9 to 12 carbon atoms.

6. The method of claim 1 or 2 , wherein the reaction is olefin metathesis.

7. The method of claim 1 or 2 , wherein the reagent is a metathesis catalyst.

8. The method of claim 1 or 2 , wherein the reagent is a Group VIII transition metal carbene catalyst.

9. The method of claim 1 or 2 , wherein the reagent is a Group VIII transition metal carbene catalyst that is a ruthenium metathesis catalyst.

10. The method of claim 1 or 2 , wherein the peptide has exactly two alpha, alpha-disubstituted amino acids.

11. The method of claim 1 or 2 , wherein two of the at least two alpha, alpha-disubstituted amino acids have an S configuration.

12. The method of claim 1 or 2 , wherein one of the at least two alpha, alpha-disubstituted amino acid has an S configuration and one of the at least two alpha, alpha-disubstituted amino acid has an R configuration.

13. The method of claim 1 or 2 , wherein the peptide comprises three alpha, alpha-disubstituted amino acids.

14. The method of claim 1 or 2 , wherein the cross-linkers is an alkene cross-linker.

15. The method of claim 1 or 2 , wherein the cross-linker is an alkene cross-linker; and further comprising reducing the alkene cross-linker, to result in an alkane cross-linker.

16. The method of claim 2 , wherein the stabilized peptide is helical.

17. The method of claim 2 , wherein the length of the cross-linked moieties between the two alpha positions is 7 or 8 carbon atoms.

18. The method of claim 2 , wherein the length of the cross-linked moieties between the two alpha positions is 9 to 12 carbon atoms.
