Abstract

The present invention relates to the field of surface water monitoring. More specifically, the present invention provides low-cost, real-time bio-electrochemical sensors for surface water monitoring based on the metabolism of one or more electrogenic microbes.

Claims (81)

1. A bio-electrochemical system for detecting the presence or absence of at least one targeted chemical in a medium comprising:

a working electrode comprising a cathode or an anode; and

at least a first electrically active micro-organism disposed in proximity to the working electrode, wherein said micro-organism selectively interacts with the working electrode based upon the presence or absence of the targeted chemical to produce an electric current; said at least first electrically active micro-organism uses the targeted chemical as an electron donor or electron acceptor instead of the working electrode; wherein the electric current produced by the system in the presence of the targeted chemical is less than the electric current produced in the absence of the target chemical.

2. The system of claim 1 , further comprising an opening by which the medium enters the system.

3. The system of claim 1 , wherein the working electrode is connected to a counter-electrode with which at least one electrically active micro-organism interacts.

4. The system of claim 1 , wherein the working electrode is connected to a counter-electrode with which no electrically active micro-organism interacts.

5. The system of claim 1 , further comprising at least one selectively permeable barrier which allows passage of the targeted chemical to the working electrode and excludes passage of the at least first electrically active micro-organism.

6. The system of claim 5 , wherein the barrier is part of a membrane-electrode assembly.

7. The system of claim 5 , wherein the barrier is a sterile filter.

8. The system of claim 1 , further comprising at least one selectively permeable barrier which allows passage of the targeted chemical to the working electrode and excludes passage of any micro-organism.

9. The system of claim 1 , further comprising a second electrically active micro-organism.

10. The system of claim 9 , wherein each of the first and second electrically active micro-organisms is substantially unable to use, or is prevented from using, at least one targeted chemical in its metabolic processes.

11. The system of claim 10 , wherein each of the first and second electrically active micro-organisms is able to use a different targeted chemical in its metabolic processes.

12. The system of claim 9 , comprising at least a first and a second sensor regions, separated by a selectively permeable membrane which allows passage of the targeted chemicals and excludes passage of the first and second electrically active micro-organisms.

13. The system of claim 12 , wherein

the first sensor region comprises the working electrode, a counter-electrode, and the first electrically active micro-organism disposed in proximity to the working electrode;

the second sensor region comprises an additional working electrode, an additional counter-electrode, and the second electrically active micro-organism disposed in proximity to the additional working electrode; and

the first and second sensor regions are separated by the selectively permeable membrane.

14. The system of claim 1 , wherein the at least first electrically active micro-organism uses the targeted chemical as an electron donor instead of the electrode.

15. The system of claim 1 , wherein the at least first electrically active micro-organism uses the targeted chemical as an electron acceptor instead of the electrode.

16. The system of claim 1 , wherein the at least first electrically active micro-organism is a strain selected from the group consisting of Shewanella, Psuedemonas, Geobacter, Cyanobacteria , and mutants thereof.

17. The system of claim 16 , wherein the at least first electrically active micro-organism is a strain of Shewanella oneidensis or a mutant thereof.

18. The system of claim 1 , further comprising a second electrically active micro-organisms, wherein the first and second electrically active micro-organisms are capable of detecting two or more targeted chemicals.

19. The system of claim 18 , where wherein at least one of the first and second electrically active micro-organisms is capable of using each of the two or more targeted chemicals.

20. The system of claim 1 , further comprising a flow controller for modulating the volume and rate at which the medium contacts the system.

21. The system of claim 1 , further comprising a consumable substrate.

The system of claim 1 , in which the targeted chemical is selected from the group consisting of oxygen, nitrite (NO 2 − ), nitrate (NO 3 − ), fumarate, dimethyl sulfoxide, arsenate, (AsO 4 3− ), uranium (VI), molybdenum (VI), vanadium (V or IV), hydrogen (H 2 ), methane (CH 4 ), ammonium (NH 4 + ), ammonia (NH 3 ), sulfide, sulfite, iron (II), arsenite, elemental sulfur (S 0 ), chromium (III), uranium (III), molybdenum (V), vanadium (IV or III), vanadate, reduced phosphate, carbohydrates, sugars, glucose, selenate, selenite, sulfate (SO 4 2− ), chromium (VI), phosphate, phosphite, iron (III), and combinations thereof.

A method of measuring the presence or absence of targeted chemicals in agricultural process water comprising monitoring the water with a bio-electrochemical system comprising a working electrode comprising a cathode or an anode, and at least one electrically active micro-organism disposed in proximity to the working electrode, wherein said micro-organism selectively interacts with the working electrode based upon the presence or absence of the targeted chemical to produce an electric current; wherein said at least one electrically active micro-organism uses the targeted chemical as an electron donor or electron acceptor instead of the working electrode; and the electric current produced by the system in the presence of the targeted chemical is less than the electric current produced in the absence of the target chemical.

24. A bio-electrochemical system for detecting the presence or absence of at least one targeted chemical in a medium comprising:

a working electrode comprising a cathode or an anode; and

at least a first electrically active micro-organism disposed in proximity to the working electrode, wherein said micro-organism selectively interacts with the working electrode based upon the presence or absence of the targeted chemical to produce an electric current; said at least first electrically active micro-organism preferentially uses the targeted chemical as an electron acceptor instead of the working electrode.

25. The system of claim 24 , further comprising an opening by which the medium enters the system.

26. The system of claim 24 , wherein the working electrode is connected to a counter-electrode with which at least one electrically active micro-organism interacts.

27. The system of claim 24 , wherein the working electrode is connected to a counter-electrode with which no electrically active micro-organism interacts.

28. The system of claim 24 , further comprising at least one selectively permeable barrier which allows passage of the targeted chemical to the working electrode and excludes passage of the at least first electrically active micro-organism.

29. The system of claim 28 , wherein the barrier is part of a membrane-electrode assembly.

30. The system of claim 28 , wherein the barrier is a sterile filter.

31. The system of claim 24 , further comprising at least one selectively permeable barrier which allows passage of the targeted chemical to the working electrode and excludes passage of any micro-organism.

32. The system of claim 24 , wherein the at least first electrically active micro-organism is a strain selected from the group consisting of Shewanella, Psuedemonas, Geobacter, Cyanobacteria , and mutants thereof.

33. The system of claim 32 , wherein the at least first electrically active micro-organism is a strain of Shewanella oneidensis or a mutant thereof.

34. The system of claim 24 , further comprising a second electrically active micro-organism.

35. The system of claim 34 , wherein the first and second electrically active micro-organisms are capable of detecting two or more targeted chemicals.

36. The system of claim 34 , wherein at least one of the first and second electrically active micro-organisms is capable of using each of the two or more targeted chemicals.

37. The system of claim 34 , wherein each of the first and second electrically active micro-organisms is substantially unable to use, or is prevented from using, at least one targeted chemical in its metabolic processes.

38. The system of claim 37 , wherein each of the first and second electrically active micro-organisms is able to use a different targeted chemical in its metabolic processes.

39. The system of claim 34 , comprising at least a first and second sensor regions, separated by a selectively permeable membrane which allows passage of the targeted chemicals and excludes passage of the first and second electrically active micro-organisms.

40. The system of claim 39 , wherein

the first sensor region comprises the working electrode, a counter-electrode, and the first electrically active micro-organism disposed in proximity to the working electrode;

the second sensor region comprises an additional working electrode, an additional counter-electrode, and the second electrically active micro-organism disposed in proximity to the additional working electrode; and

the first and second sensor regions are separated by the selectively permeable membrane.

41. The system of claim 24 , wherein the electric current produced by the system in the presence of the targeted chemical is less than the electric current produced in the absence of the target chemical.

42. The system of claim 24 , further comprising a flow controller for modulating the volume and rate at which the medium contacts the system.

43. The system of claim 24 , further comprising a consumable substrate.

44. The system of claim 24 , in which the targeted chemical is selected from the group consisting of oxygen, nitrite (NO 2 − ), nitrate (NO 3 − ), fumarate, dimethyl sulfoxide, arsenate, (AsO 4 3− ), uranium (VI), molybdenum (VI), vanadium (V or IV), hydrogen (H 2 ), methane (CH 4 ), ammonium (NH 4 − ), ammonia (NH 3 ), sulfide, sulfite, iron (II), arsenite, elemental sulfur (S 0 ), chromium (III), uranium (III), molybdenum (V), vanadium (IV or III), vanadate, reduced phosphate, carbohydrates, sugars, glucose, selenate, selenite, sulfate (SO 4 2− ), chromium (VI), phosphate, phosphite, iron (III), and combinations thereof.

45. A bio-electrochemical system for detecting the presence or absence of at least one targeted chemical in a medium comprising:

a working electrode comprising a cathode or an anode; and

at least a first electrically active micro-organism disposed in proximity to the working electrode, wherein said at least first micro-organism selectively interacts with the working electrode based upon the presence or absence of the targeted chemical to produce an electric current; and the at least first electrically active micro-organism comprises Shewanella oneidensis or a mutant thereof.

46. The system of claim 45 , further comprising an opening by which the medium enters the system.

47. The system of claim 45 , wherein the working electrode is connected to a counter-electrode with which at least one electrically active micro-organism interacts.

48. The system of claim 45 , wherein the working electrode is connected to a counter-electrode with which no electrically active micro-organism interacts.

49. The system of claim 45 , further comprising at least one selectively permeable barrier which allows passage of the targeted chemical to the working electrode and excludes passage of the at least one electrically active micro-organism.

50. The system of claim 49 , further comprising at least one selectively permeable barrier which allows passage of the targeted chemical to the working electrode and excludes passage of any micro-organism.

51. The system of claim 49 , wherein the barrier is part of a membrane-electrode assembly.

52. The system of claim 49 , wherein the barrier is a sterile filter.

53. The system of claim 45 , wherein the at least first electrically active micro-organism is configured to preferentially use the targeted chemical over the working electrode as an electron donor or an electron acceptor.

54. The system of claim 45 , wherein the at least first electrically active micro-organism uses the targeted chemical as an electron donor instead of the electrode.

55. The system of claim 45 , wherein the at least first electrically active micro-organism uses the targeted chemical as an electron acceptor instead of the electrode.

56. The system of claim 45 , further comprising a second electrically active micro-organism.

57. The system of claim 56 , wherein the first and second electrically active micro-organisms are capable of detecting two or more targeted chemicals.

58. The system of claim 56 , wherein at least one of the first and second electrically active micro-organisms is capable of using each of the two or more targeted chemicals.

59. The system of claim 56 , wherein each of the first and second electrically active micro-organisms is substantially unable to use, or is prevented from using, at least one targeted chemical in its metabolic processes.

60. The system of claim 59 , wherein each of the first and second electrically active micro-organisms is able to use a different targeted chemical in its metabolic processes.

61. The system of claim 56 , comprising at least a first and second sensor regions, separated by a selectively permeable membrane which allows passage of the targeted chemicals and excludes passage of the first and second electrically active micro-organisms.

62. The system of claim 61 , wherein

the first sensor region comprises the working electrode, a counter-electrode, and the first electrically active micro-organism disposed in proximity to the working electrode;

the second sensor region comprises an additional working electrode, an additional counter-electrode, and the second electrically active micro-organism disposed in proximity to the additional working electrode; and

the first and second sensor regions are separated by the selectively permeable membrane.

63. The system of claim 45 , wherein the electric current produced by the system in the presence of the targeted chemical is less than the electric current produced in the absence of the target chemical.

64. The system of claim 45 , further comprising a flow controller for modulating the volume and rate at which the medium contacts the system.

65. The system of claim 45 , further comprising a consumable substrate.

66. The system of claim 45 , in which the targeted chemical is selected from the group consisting of oxygen, nitrite (NO 2 − ), nitrate (NO 3 − ), fumarate, dimethyl sulfoxide, arsenate, (AsO 4 3− ), uranium (VI), molybdenum (VI), vanadium (V or IV), hydrogen (H 2 ), methane (CH 4 ), ammonium (NH 4 + ), ammonia (NH 3 ), sulfide, sulfite, iron (II), arsenite, elemental sulfur (S 0 ), chromium (III), uranium (III), molybdenum (V), vanadium (IV or III), vanadate, reduced phosphate, carbohydrates, sugars, glucose, selenate, selenite, sulfate (SO 4 2− ), chromium (VI), phosphate, phosphite, iron (III), and combinations thereof.
