Abstract

The present invention relates to the use of LNA antisense apoB oligonucleotides for the treatment of acute coronary syndrome.

Claims (11)

1. A method of treatment of acute coronary syndrome, comprising administering to a patient an oligonucleotide comprising GsTstsgsascsascstsgsTsmC (SEQ ID NO:41), wherein upper case letters indicate LNA, mC indicates methyl C-LNA, and the s represents phosphorothioate linkage to a patient suffering from acute coronary syndrome.

2. The method of claim 1 , wherein the oligonucleotide is administered in a dosage to achieve at least a 60% reduction in PK/PD within the first week of treatment.

3. The method of claim 1 , further comprising monitoring wherein potential side effects on kidney and liver is monitored by measuring ALT, AST, bilirubin and creatinine in the patient, and wherein the dosage of the oligonucleotide accordingly is adjusted to avoid side effects on these organs.

4. The method of claim 1 , wherein the oligonucleotide is administered to the patient in a dosage to achieve a reduction in PK/PD of between 60% and 80% within the first week of treatment.

5. The method of claim 1 , wherein the oligonucleotide is used in combination with a statin.

6. The method of claim 1 wherein the oligonucleotide is administered intravenously or subcutaneously.

7. The method of claim 1 , wherein the oligonucleotide has a fast onset to reach maximum effect on total cholesterol within 7 days after onset of treatment.

8. The method of claim 1 comprising administering a loading dosage of up to 50 mg/kg followed by administering a maintenance dosages of up to 10 mg/kg.

9. The method of claim 1 further comprising administrating a statin.

10. The method of claim 1 , wherein the patient is newly diagnosed as being at risk of developing acute coronary syndrome.

11. The method of claims 1 - 5 and 6 - 10 wherein the oligonucleotide is conjugated to one or more sugar residues or cholesterol.
