Polypeptides having cellobiohydrolase activity and polynucleotides encoding same
The present invention relates to isolated polypeptides having cellobiohydrolase activity and polynucleotides encoding the polypeptides. The invention also relates to nucleic acid constructs, vectors, and host cells comprising the polynucleotides as well as methods of producing and using the polypeptides.
1. A nucleic acid construct comprising a polynucleotide encoding a polypeptide having cellobiohydrolase activity, wherein the polynucleotide is operably linked to one or more heterologous control sequences that direct the production of the polypeptide in an expression host, and wherein the polypeptide having cellobiohydrolase activity comprises:
(a) the sequence of amino acids 19-456 of SEQ ID NO: 2; or a sequence that differs by no more than four amino acids from the sequence of amino acids 19-456 of SEQ ID NO: 2;
(b) the sequence of amino acids 19-456 of SEQ ID NO: 6; or a sequence that differs by no more than eight amino acids from the sequence of amino acids 19-456 of SEQ ID NO: 6; or
(c) the sequence of amino acids 19-457 of SEQ ID NO: 8; or a sequence that differs by no more than four amino acids from the sequence of amino acids 19-457 of SEQ ID NO: 8.
2. A recombinant host cell comprising the nucleic acid construct of claim 1 .
3. A method of producing a polypeptide having cellobiohydrolase activity, comprising:
(a) cultivating the host cell of claim 2 under conditions conducive for production of the polypeptide; and
(b) recovering the polypeptide.
4. A recombinant expression vector comprising the nucleic acid construct of claim 1 .
5. The nucleic acid construct of claim 1 , wherein the polypeptide having cellobiohydrolase activity further comprises a cellulose binding domain.
6. The nucleic acid construct of claim 1 , wherein the polypeptide having cellobiohydrolase activity comprises a sequence that differs by no more than four amino acids from the sequence of amino acids 19-456 of SEQ ID NO: 2.
7. The nucleic acid construct of claim 1 , wherein the polypeptide having cellobiohydrolase activity comprises a sequence that differs by no more than three amino acids from the sequence of amino acids 19-456 of SEQ ID NO: 2.
8. The nucleic acid construct of claim 1 , wherein the polypeptide having cellobiohydrolase activity comprises a sequence that differs by no more than two amino acids from the sequence of amino acids 19-456 of SEQ ID NO: 2.
9. The nucleic acid construct of claim 1 , wherein the polypeptide having cellobiohydrolase activity comprises a sequence that differs by one amino acid from the sequence of amino acids 19-456 of SEQ ID NO: 2.
10. The nucleic acid construct of claim 1 , wherein the polypeptide having cellobiohydrolase activity comprises the sequence of amino acids 19-456 of SEQ ID NO: 2.
11. The nucleic acid construct of claim 1 , wherein the polypeptide having cellobiohydrolase activity comprises the sequence of amino acids of SEQ ID NO: 2.
12. The nucleic acid construct of claim 1 , wherein the polypeptide having cellobiohydrolase activity comprises a sequence that differs by no more than eight amino acids from the sequence of amino acids 19-456 of SEQ ID NO: 6.
13. The nucleic acid construct of claim 1 , wherein the polypeptide having cellobiohydrolase activity comprises a sequence that differs by no more than six amino acids from the sequence of amino acids 19-456 of SEQ ID NO: 6.
14. The nucleic acid construct of claim 1 , wherein the polypeptide having cellobiohydrolase activity comprises a sequence that differs by no more than four amino acids from the sequence of amino acids 19-456 of SEQ ID NO: 6.
15. The nucleic acid construct of claim 1 , wherein the polypeptide having cellobiohydrolase activity comprises a sequence that differs by no more than three amino acids from the sequence of amino acids 19-456 of SEQ ID NO: 6.
16. The nucleic acid construct of claim 1 , wherein the polypeptide having cellobiohydrolase activity comprises a sequence that differs by no more than two amino acids from the sequence of amino acids 19-456 of SEQ ID NO: 6.
17. The nucleic acid construct of claim 1 , wherein the polypeptide having cellobiohydrolase activity comprises a sequence that differs by one amino acid from the sequence of amino acids 19-456 of SEQ ID NO: 6.
18. The nucleic acid construct of claim 1 , wherein the polypeptide having cellobiohydrolase activity comprises the sequence of amino acids 19-456 of SEQ ID NO: 6.
19. The nucleic acid construct of claim 1 , wherein the polypeptide having cellobiohydrolase activity comprises the sequence of amino acids of SEQ ID NO: 6.
20. The nucleic acid construct of claim 1 , wherein the polypeptide having cellobiohydrolase activity comprises a sequence that differs by no more than four amino acids from the sequence of amino acids 19-457 of SEQ ID NO: 8.
21. The nucleic acid construct of claim 1 , wherein the polypeptide having cellobiohydrolase activity comprises a sequence that differs by no more than three amino acids from the sequence of amino acids 19-457 of SEQ ID NO: 8.
22. The nucleic acid construct of claim 1 , wherein the polypeptide having cellobiohydrolase activity comprises a sequence that differs by no more than two amino acids from the sequence of amino acids 19-457 of SEQ ID NO: 8.
23. The nucleic acid construct of claim 1 , wherein the polypeptide having cellobiohydrolase activity comprises a sequence that differs by one amino acid from the sequence of amino acids 19-457 of SEQ ID NO: 8.
24. The nucleic acid construct of claim 1 , wherein the polypeptide having cellobiohydrolase activity comprises the sequence of amino acids 19-457 of SEQ ID NO: 8.
25. The nucleic acid construct of claim 1 , wherein the polypeptide having cellobiohydrolase activity comprises the sequence of amino acids of SEQ ID NO: 8.
26. An isolated recombinant host cell transformed with a polynucleotide encoding a polypeptide having cellobiohydrolase activity, wherein the polynucleotide is heterologous to the recombinant host cell, and wherein the polypeptide having cellobiohydrolase activity comprises:
(a) the sequence of amino acids 19-456 of SEQ ID NO: 2; or a sequence that differs by no more than four amino acids from the sequence of amino acids 19-456 of SEQ ID NO: 2;
(b) the sequence of amino acids 19-456 of SEQ ID NO: 6; or a sequence that differs by no more than eight amino acids from the sequence of amino acids 19-456 of SEQ ID NO: 6; or
(c) the sequence of amino acids 19-457 of SEQ ID NO: 8; or a sequence that differs by no more than four amino acids from the sequence of amino acids 19-457 of SEQ ID NO: 8.
