Abstract

Methods for regulating T cell function in a subject, particularly regulatory T cell activity are provided. Methods of the invention include administering to a subject a therapeutically effective amount of an Interleukin 35-specific binding agent, such as an antibody or small molecule inhibitor. The invention further provides methods for enhancing the immunogenicity of a vaccine or overcoming a suppressed immune response to a vaccine in a subject, including administering to the subject a therapeutically effective amount of an IL35-specific binding agent and administering to the subject a vaccine. In one embodiment the vaccine is a cancer vaccine.

Claims (10)

1. A cell cultured by the method of

a. isolating a population of T effector cells;

b. activating said population of T effector cells; and,

c. culturing said activated population of T effector cells with ectopically expressed Interleukin 35 (IL35), wherein said ectopically expressed IL35 is present in an amount sufficient to suppress T effector cell proliferation.

2. The cell of claim 1 , wherein the IL35 comprises an Epstein-Barr virus-induced gene 3 (EBI3) subunit and a P35 subunit which are not covalently linked to one another.

3. A method of culturing a population of activated T effector cells comprising:

a. isolating a population of T effector cells;

b. activating said population of T effector cells; and,

c. culturing said activated population of T effector cells with ectopically expressed Interleukin 35 (IL35), wherein said ectopically expressed IL35 is present in an amount sufficient to suppress T effector cell proliferation.

4. The method of claim 3 , wherein the IL35 comprises an Epstein-Barr virus-induced gene 3 (EBI3) subunit and a P35 subunit which are not covalently linked to one another.
