Abstract

The disclosure relates to compounds of formula (I): wherein , A, W, X, and Ra are as defined in the disclosure, and salts thereof, and to pharmaceutical compositions comprising said compounds, to processes for preparing them, and to their use as medicaments, in particular as MET inhibitors.

Claims (48)

1. A compound of formulae E′, M1, M2, or M3:

wherein:

R 6 represents alkyl, wherein the alkyl is optionally substituted with NR 3 R 4 , —(CH 2 ) n —NR 3 R 4 , alkoxy, hydroxyl, heterocycloalkyl, phenyl or —(CH 2 ) n -phenyl, wherein the phenyl is optionally substituted and n represents an integer from 1 to 4;

R 7 represents cycloalkyl or alkyl, wherein the cycloalkyl or alkyl is optionally substituted with NR 3 R 4 , alkoxy, hydroxyl, phenyl, heteroaryl or heterocycloalkyl, wherein the phenyl, heteroaryl or heterocycloalkyl is optionally substituted;

R a represents hydrogen; halo; alkoxy optionally substituted with chloro, hydroxyl or heterocycloalkyl; O-cycloalkyl; heteroaryl; phenyl; NHC(═O)alkyl; NHC(═O)cyclo alkyl; or NR 1 R 2 ;

one of R 1 and R 2 represents hydrogen or alkyl, and the other of R 1 and R 2 represents hydrogen, cycloalkyl or alkyl, wherein the cycloalkyl or alkyl is optionally substituted with one or more substituents independently selected from the group consisting of hydroxyl, alkoxy, heteroaryl, heterocycloalkyl, NR 3 R 4 and optionally substituted phenyl, or else R 1 and R 2 form, with the nitrogen atom to which they are attached, a cyclic moiety containing from 3 to 10 members and optionally one or more other heteroatoms selected from the group consisting of O, S, N and NH;

R 3 and R 4 independently represent hydrogen, alkyl, cycloalkyl, heteroaryl or phenyl, wherein the alkyl, cycloalkyl, heteroaryl or phenyl is optionally substituted, or else R 3 and R 4 form, with the nitrogen atom to which they are attached, a cyclic moiety containing from 3 to 10 members and optionally one or more other heteroatoms selected from the group consisting of O, S, N and NH;

wherein the heterocycloalkyl, the heteroaryl and the phenyl and the cyclic moieties that R 1 and R 2 or R 3 and R 4 can form with the nitrogen atom to which they are attached, are optionally substituted with one or more substituents selected from the group consisting of halo, hydroxyl, oxo, alkoxy, NH 2 , NHalkyl, N(alkyl) 2 , alkyl, cycloalkyl, heterocycloalkyl, CH 2 -heterocycloalkyl, phenyl, CH 2 -phenyl, heteroaryl, CO-phenyl and S-heteroaryl;

wherein the alkyl, heterocycloalkyl, phenyl and heteroaryl substituents are optionally further substituted with one or more substituents selected from the group consisting of halo, hydroxyl, oxo, alkyl, alkoxy, NH 2 , NHalkyl and N(alkyl) 2 ; and

wherein:

alkyl is C 1 to C 6 alkyl;

alkoxy is C 1 to C 6 alkoxy;

cycloalkyl is a saturated carbocyclic radical containing 3 to 10 carbon atoms;

heterocycloalkyl is a mono or bicyclic carbocyclic moiety containing from 3 to 10 members, with one or more heteroatoms independently selected from the group consisting of oxygen, nitrogen or sulphur; and

heteroaryl is a monocyclic or bicyclic, unsaturated or partially unsaturated, heterocyclic moiety containing at most 12 members, which optionally contains a —C(═O)— member and containing one or more heteroatoms which may be the same or different chosen from oxygen, nitrogen or sulphur atoms,

or the addition salts with pharmaceutically acceptable mineral and organic acids or with pharmaceutically acceptable mineral and organic bases of said compound.

2. The compound of claim 1 , wherein R a represents alkoxy optionally substituted with chloro, hydroxyl or heterocycloalkyl; O-cycloalkyl; NHC(═O)alkyl; or NR 1 R 2 ;

or the addition salts with pharmaceutically acceptable mineral and organic acids or with pharmaceutically acceptable mineral and organic bases of said compound.

3. The compound of claim 1 , wherein R 3 and R 4 independently represent hydrogen or alkyl;

or the addition salts with pharmaceutically acceptable mineral and organic acids or with pharmaceutically acceptable mineral and organic bases of said compound.

4. The compound of claim 1 , wherein R a represents hydrogen, halo or phenyl;

or the addition salts with pharmaceutically acceptable mineral and organic acids or with pharmaceutically acceptable mineral and organic bases of said compound.

5. The compound of claim 1 , wherein R 3 and R 4 form, with the nitrogen atom to which they are attached, a cyclic moiety containing from 3 to 10 members and optionally one or more other heteroatoms chosen from O, S, N and NH;

or the addition salts with pharmaceutically acceptable mineral and organic acids or with pharmaceutically acceptable mineral and organic bases of said compound.

6. The compound of claim 1 , wherein R a represents hydrogen, chloro or

wherein R b represents halo; or S-heteroaryl optionally substituted with a substituent selected from the group consisting of halo, hydroxyl, alkyl, alkoxy, NH 2 , NHalkyl and N(alkyl) 2 ;

or the addition salts with pharmaceutically acceptable mineral and organic acids or with pharmaceutically acceptable mineral and organic bases of said compound.

7. The compound of claim 1 , wherein the compound is a compound of Formula E′, or the addition salts with pharmaceutically acceptable mineral and organic acids or with pharmaceutically acceptable mineral and organic bases of said compound.

8. The compound of claim 7 , wherein the compound of Formula E′ is:

3-chloro-6-(4-fluorophenyl)-7,8-dihydro[1,2,4]triazolo[4,3-b]pyridazine,

or the addition salts with pharmaceutically acceptable mineral and organic acids or with pharmaceutically acceptable mineral and organic bases of said compound.

9. The compound of claim 1 , wherein the compound is a compound of Formula M1, or the addition salts with pharmaceutically acceptable mineral and organic acids or with pharmaceutically acceptable mineral and organic bases of said compound.

10. The compound of claim 9 , wherein the compound of Formula M1 is:

methyl (6-sulphanyl-1,3-benzothiazol-2-yl)carbamate;

1,1-dimethylethyl (6-sulphanyl-1,3-benzothiazol-2-yl)carbamate; or

2-morpholin-4-ylethyl (6-sulphanyl-1,3-benzothiazol-2-yl)carbamate;

or the addition salts with pharmaceutically acceptable mineral and organic acids or with pharmaceutically acceptable mineral and organic bases of said compound.

11. The compound of claim 1 , wherein the compound is a compound of Formula M2, or the addition salts with pharmaceutically acceptable mineral and organic acids or with pharmaceutically acceptable mineral and organic bases of said compound.

12. The compound of claim 11 , wherein the compound of Formula M2 is:

1-(2-morpholin-4-ylethyl)-3-(6-sulphanyl-1,3-benzothiazol-2-yl)urea;

1-{2-[(2R,6S)-2,6-dimethylmorpholin-4-yl]ethyl}-3-(6-sulphanyl-1,3-benzothiazol-2-yl)urea;

1-[2-(4-methylpiperazin-1-yl)ethyl]-3-(6-sulphanyl-1,3-benzothiazol-2-yl)urea; or

1-[2-(pyrrolidin-1-yl)ethyl]-3-(6-sulphanyl-1,3-benzothiazol-2-yl)urea;

or the addition salts with pharmaceutically acceptable mineral and organic acids or with pharmaceutically acceptable mineral and organic bases of said compound.

13. The compound of claim 1 , wherein the compound is a compound of Formula M3, or the addition salts with pharmaceutically acceptable mineral and organic acids or with pharmaceutically acceptable mineral and organic bases of said compound.

14. The compound of claim 13 , wherein the compound of Formula M3 is:

4-morpholin-4-yl-N-(6-sulphanyl-1,3-benzothiazol-2-yl)butanamide;

or the addition salts with pharmaceutically acceptable mineral and organic acids or with pharmaceutically acceptable mineral and organic bases of said compound.
