Vaccine antigens that direct immunity to conserved epitopes
A method of identification and elimination of immunodominant epitopes to elicit a response to secondary epitopes, especially conserved structures, is described, and applied to influenza haemagglutinin (HA). Identification of the primary epitopes in (HA), and replacement of amino acids having high LODrps with corresponding low LODrps amino acids produces an HA molecule which induces antibody responses to conserved HA residues. Modified HA molecules induce a broadly neutralizing vaccine.
1. A method of making a vaccine that elicits an immune response against non-immunodominant conserved epitopes on an influenza H5 haemagglutinin, comprising:
(a) identifying one or more primary antigenic determinant residues of a primary immunodominant epitope of a native influenza H5 haemagglutinin, wherein the primary immunodominant epitope comprises amino acids 138-209 of SEQ ID NO:1, and wherein the identification of the primary antigenic determinant residues comprises;
comparing the amino acid sequence of the native influenza H5 haemagglutinin to the corresponding amino acid sequence of an escape mutant and/or a genetic drift isolate; and
identifying one or more amino acids that differ between the amino acid sequence of the native influenza H5 haemagglutinin and the amino acid sequence of the escape mutant and/or the genetic drift isolate;
(b) determining the log odds relative propensity scale (LODrps) value of one or more of the primary antigenic determinant residues;
(c) modifying at least one of the primary antigenic determinant residues having a high LODrps value by replacing the at least one of the primary antigenic determinant residues having a high LODrps value with an amino acid having a low LODrps value thereby creating a modified influenza H5 haemagglutinin with reduced antigenicity, wherein the at least one modified primary antigenic determinant residue of amino acids 138-209 of SEQ ID NO:1 is selected from the group consisting of Ser141Ala, Ser141Val, Ser141Leu, Ser141Ile, Ser144Ala, Ser144Val, Ser144Leu, Ser144Ile, Glu146Ser, Glu146Thr, Glu146Val, Glu146Leu, Glu146Ile, Pro156Ala, Pro156Val, Pro156Ile, Gln158Thr, Gln158Val, Lys160Leu, Ser161Val, Ser161Ile, Lys172Ser, Lys172Ala, Lys172Val, Lys172Leu, Thr176Leu, Arg182Ala, Arg182Leu, Lys209Ala, and/or Lys209Val; and
(d) utilizing the modified influenza H5 haemagglutinin to induce antibodies against the non-immunodominant conserved epitopes.
2. The method of claim 1 , wherein each of the primary antigenic determinant residues having a high LODrps value is replaced with an amino acid having a low LODrps value.
3. A vaccine made by the method of claim 1 .
4. A modified influenza H5 haemagglutinin in which at least one primary antigenic determinant residue of a primary immunodominant epitope in a native influenza H5 haemagglutinin is modified by replacement of at least one high LODrps amino acid with a low LODrps amino acid, thereby reducing the antigenicity of the primary immunodominant epitope, wherein the primary immunodominant epitope comprises amino acids 138-209 of SEQ ID NO:1, and wherein the at least one modified primary antigenic residue of amino acids 138-209 of SEQ ID NO:1 is selected from the group consisting of Ser141Ala, Ser141Val, Ser141Leu, Ser141Ile, Ser144Ala, Ser144Val, Ser144Leu, Ser144Ile, Glu146Ser, Glu146Thr, Glu146Val, Glu146Leu, Glu146Ile, Pro156Ala, Pro156Val, Pro156Ile, Gln158Thr, Gln158Val, Lys160Leu, Ser161Val, Ser161Ile, Lys172Ser, Lys172Ala, Lys172Val, Lys172Leu, Thr176Leu, Arg182Ala, Arg182Leu, Lys209Ala, and/or Lys209Val.
5. A vaccine, comprising one or more of the modified influenza H5 haemagglutinins of claim 4 , and a pharmaceutically acceptable carrier.
6. A method of immunizing a subject against a H5 influenza virus infection, comprising administering to the subject at least one therapeutically effective dose of the vaccine of claim 5 .
7. A method of immunizing a subject against a H5 influenza virus infection, wherein at least a first and second therapeutically effective dose of a vaccine comprising one or more of the influenza H5 haemagglutinins of claim 4 and a pharmaceutically acceptable carrier are administered to a subject; wherein the first dose comprises a first modified influenza H5 haemagglutinin according to claim 4 , and the second dose comprises a second modified influenza H5 haemagglutinin according to claim 4 , and wherein the first and second influenza haemagglutinins are different.
8. The method of claim 6 , further comprising administration of an adjuvant.
9. A method of producing therapeutic antibodies against conserved epitopes of influenza H5 haemagglutinin comprising:
(a) administration of one or more modified influenza H5 haemagglutinins of claim 4 to a subject; and
(b) isolation of anti-H5 haemagglutinin antibodies.

The modified influenza H5 haemagglutinin of claim 4 , in which at least three primary antigenic determinant residues of the primary immunodominant epitope are modified by replacement of at least one high LODrps amino acid with a low LODrps amino acid, wherein the at least three modified primary antigenic residues of amino acids 138-209 of SEQ ID NO:1 are selected from the group consisting of Ser141Thr, Ser141Ala, Ser141Val, Ser141Leu, Ser141Ile, Ser144Thr, Ser144Ala, Ser144Val, Ser144Leu, Ser144Ile, Glu146Ser, Glu146Thr, Glu146Ala, Glu146Val, Glu146Leu, Glu146Ile, Pro156Ala, Pro156Val, Pro156Leu, Pro156Ile, Gln158Ser, Gln158Thr, Gln158Ala, Gln158Val, Lys160Leu, Lys160Ile, Ser161Thr, Ser161Ala, Ser161Val, Ser161Leu, Ser161Ile, Lys172Ser, Lys172Thr, Lys172Ala, Lys172Val, Lys172Leu, Lys172Ile, Thr176Leu, Thr176Ile, Arg182Thr, Arg182Ala, Arg182Leu, Lys209Ser, Lys209Thr, Lys209Ala, Lys209Val, Lys209Leu, and/or Lys209Ile.
11. A modified influenza H5 haemagglutinin, comprising amino acids 138-209 of SEQ ID NO:9.

The method of claim 1 , further comprising modifying at least three of the primary antigenic determinant residues having a high LODrps value by replacing the at least three of the primary antigenic determinant residues having a high LODrps value with an amino acid having a low LODrps value thereby creating a modified influenza H5 haemagglutinin with reduced antigenicity, wherein the at least three modified primary antigenic determinant residues of amino acids 138-209 of SEQ ID NO:1 are selected from the group consisting of Ser141Thr, Ser141Ala, Ser141Val, Ser141Leu, Ser141Ile, Ser144Thr, Ser144Ala, Ser144Val, Ser144Leu, Ser144Ile, Glu146Ser, Glu146Thr, Glu146Ala, Glu146Val, Glu146Leu, Glu146Ile, Pro156Ala, Pro156Val, Pro156Leu, Pro156Ile, Gln158Ser, Gln158Thr, Gln158Ala, Gln158Val, Lys160Leu, Lys160Ile, Ser161Thr, Ser161Ala, Ser161Val, Ser161Leu, Ser161Ile, Lys172Ser, Lys172Thr, Lys172Ala, Lys172Val, Lys172Leu, Lys172Ile, Thr176Leu, Thr176Ile, Arg182Thr, Arg182Ala, Arg182Leu, Lys209Ser, Lys209Thr, Lys209Ala, Lys209Val, Lys209Leu, and/or Lys209Ile.
