Abstract

There are provided inter alia multisubstituted aromatic compounds useful for the inhibition of thrombin, which compounds include substituted pyrazolyl or substituted triazolyl. There are additionally provided pharmaceutical compositions. There are additionally provided methods of treating and preventing a disease or disorder, which disease or disorder is amenable to treatment or prevention by the inhibition of thrombin.

Claims (180)

1. A compound with structure of the following formula:

or pharmaceutically acceptable salt, ester, solvate, or prodrug thereof;

wherein:

L 1 is —NR 5 —;

L 2 is a bond;

L 3 is a bond, substituted or unsubstituted alkylene, substituted or unsubstituted heteroalkylene, —S—, —SO—, —SO 2 —, —O—, —NHSO 2 —, or —NR 5 —;

L 4 is a bond, substituted or unsubstituted alkylene, substituted or unsubstituted heteroalkylene, —S—, —SO—, —SO 2 —, —O—, —NHSO 2 —, or —NR 5 —;

R 1 is substituted alkyl, wherein the substituted alkyl has one or more substituent group selected from the group consisting of —OH, —NH 2 , —SH, —CN, —CF 3 , —NO 2 , oxo, halogen, —COOH, substituted or unsubstituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted heterocycloalkyl, substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl;

R 2 is substituted or unsubstituted cycloalkyl, substituted or unsubstituted cycloalkenyl, substituted or unsubstituted heterocycloalkyl, substituted or unsubstituted heterocycloalkenyl, substituted or unsubstituted fused ring aryl, or substituted or unsubstituted heteroaryl, wherein the substituted cycloalkyl, substituted cycloalkenyl, substituted heterocycloalkyl, substituted heterocycloalkenyl, substituted fused ring aryl, or substituted heteroaryl has one or more substituent group selected from the group consisting of oxo, —OH, —NH 2 , —SH, —CN, —CF 3 , —NO 2 , halogen, —COOH, substituted or unsubstituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, or substituted or unsubstituted heterocycloalkyl;

R 3 is substituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted cycloalkenyl, substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl, wherein the substituted heteroaryl group has one or more substituent group selected from the group consisting of —OH, —NH 2 , —SH, —CN, —CF 3 , —NO 2 , halogen, —COOH, substituted or unsubstituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted heterocycloalkyl, substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl;

R 4 is hydrogen, substituted or unsubstituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted heterocycloalkyl, substituted aryl, or substituted or unsubstituted heteroaryl; and

R 5 is hydrogen, or substituted or unsubstituted alkyl.

2. The compound according to claim 1 , wherein L 3 is a bond, substituted or unsubstituted alkylene, or substituted or unsubstituted heteroalkylene, and R 3 is substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl.

3. The compound according to claim 1 , wherein L 3 is a bond, —NH—, —NHCH 2 —or —NH(CH 2 ) 2 —.

4. The compound according to claim 1 , wherein L 4 is a bond; and R 4 is hydrogen, substituted or unsubstituted alkyl, or substituted or unsubstituted heteroalkyl.

5. A compound with structure of the following formula:

or pharmaceutically acceptable salt, ester, solvate, or prodrug thereof;

wherein:

L 1 is a bond, substituted or unsubstituted alkylene, substituted or unsubstituted heteroalkylene, —S—, —SO—, —SO 2 —, —NHSO 2 —, or —NR 5 —;

L 2 is a bond;

L 3 is a bond, substituted or unsubstituted alkylene, substituted or unsubstituted heteroalkylene, —S—, —SO—, —SO 2 —, —O—, —NHSO 2 —, or —NR 5 —;

L 4 is a bond;

R 1 is substituted or unsubstituted alkyl, substituted or unsubstituted heteroalkyl having at least one heteroatom selected from the group consisting of N, P, Si, and S, substituted or unsubstituted cycloalkyl, substituted or unsubstituted heterocycloalkyl, substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl, wherein the substituted alkyl, substituted heteroalkyl, substituted cycloalkyl, substituted heterocycloalkyl, substituted aryl, or substituted heteroaryl has one or more substituent group selected from the group consisting of —OH, —NH 2 , —SH, —CN, —CF 3 , —NO 2 , oxo, halogen, —COOH, substituted or unsubstituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted heterocycloalkyl, substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl;

R 2 is substituted or unsubstituted cycloalkyl, substituted or unsubstituted cycloalkenyl, substituted or unsubstituted heterocycloalkyl, substituted or unsubstituted heterocycloalkenyl, substituted or unsubstituted fused ring aryl, or substituted or unsubstituted heteroaryl, wherein the substituted cycloalkyl, substituted cycloalkenyl, substituted heterocycloalkyl, substituted heterocycloalkenyl, substituted fused ring aryl, or substituted heteroaryl has one or more substituent group selected from the group consisting of oxo, —OH, —NH 2 , —SH, —CN, —CF 3 , —NO 2 , halogen, —COOH, substituted or unsubstituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, or substituted or unsubstituted heterocycloalkyl;

R 3 is substituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted cycloalkenyl, substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl, wherein the substituted heteroaryl group has one or more substituent selected from the group consisting of —OH, —NH 2 , —SH, —CN, —CF 3 , —NO 2 , halogen, —COOH, substituted or unsubstituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted heterocycloalkyl, substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl;

R 4 is unsubstituted alkyl; and

R 5 is hydrogen, or substituted or unsubstituted alkyl;

provided that:

L 1 is a bond, R 1 is substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted heterocycloalkyl,

when L 1 is a substituted or unsubstituted alkylene, R 1 is substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, unsubstituted heterocycloalkyl, substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl; and

when L 1 is —S—or substituted or unsubstituted heteroalkylene wherein the heteroatom is S, R 1 is substituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted heterocycloalkyl, substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl.

6. A pharmaceutical composition comprising a compound according to claim 1 , and a pharmaceutically acceptable excipient.

7. A compound with structure of the following formula:

or pharmaceutically acceptable salt, ester, solvate, or prodrug thereof;

wherein:

L 1 is a bond, substituted or unsubstituted alkylene, substituted or unsubstituted heteroalkylene, —S—, —SO—, —SO 2 —, —NHSO 2 —, or —NR 5 —;

L 2 is a bond;

L 3 is a bond, substituted or unsubstituted alkylene, substituted or unsubstituted heteroalkylene, —S—, —SO—, —SO 2 —, —O—, —NHSO 2 —, or —NR 5 —;

L 4 is a bond; substituted or unsubstituted alkylene, substituted or unsubstituted heteroalkylene, —S—, —SO—, —SO 2 —, —O—, —NHSO 2 —, or —NR 5 —;

R 1 is substituted or unsubstituted alkyl, substituted or unsubstituted heteroalkyl having at least one heteroatom selected from the group consisting of N, P, Si, and S, substituted or unsubstituted cycloalkyl, substituted or unsubstituted heterocycloalkyl, substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl, wherein the substituted alkyl, substituted heteroalkyl, substituted cycloalkyl, substituted heterocycloalkyl, substituted aryl, or substituted heteroaryl has one or more substituent group selected from the group consisting of —OH, —NH 2 , —SH, —CN, —CF 3 , —NO 2 , oxo, halogen, —COOH, substituted or unsubstituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted heterocycloalkyl, substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl;

R 2 is substituted or unsubstituted cycloalkyl, substituted or unsubstituted cycloalkenyl, substituted or unsubstituted heterocycloalkyl, substituted or unsubstituted heterocycloalkenyl, substituted or unsubstituted fused ring aryl, or substituted or unsubstituted heteroaryl, wherein the substituted cycloalkyl, substituted cycloalkenyl, substituted heterocycloalkyl, substituted heterocycloalkenyl, substituted fused ring aryl, or substituted heteroaryl has one or more substituent group selected from the group consisting of oxo, —OH, —NH 2 , —SH, —CN, —CF 3 , —NO 2 , halogen, —COOH, substituted or unsubstituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, or substituted or unsubstituted heterocycloalkyl;

R 3 is substituted or unsubstituted heteroaryl, wherein the substituted heteroaryl group has one or more substituent selected from the group consisting of —OH, —NH 2 , —SH, —CN, —CF 3 , —NO 2 , halogen, —COOH, substituted or unsubstituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted heterocycloalkyl, substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl;

R 4 is hydrogen, substituted or unsubstituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted heterocycloalkyl, substituted aryl, or substituted or unsubstituted heteroaryl; and

R 5 is hydrogen, or substituted or unsubstituted alkyl;

provided that:

L 1 is a bond, R 1 is substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, or substituted or unsubstituted heterocycloalkyl,

when L 1 is a substituted or unsubstituted alkylene, R 1 is substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, unsubstituted heterocycloalkyl, or substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl; and

when L 1 is —S—or substituted or unsubstituted heteroalkylene wherein the heteroatom is S, R 1 is substituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted heterocycloalkyl, substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl.

8. The compound according to claim 7 , wherein said R 3 substituted or unsubstituted heteroaryl contains nitrogen as a heteroatom.

The compound according to claim 8 , wherein R 3 is substituted or unsubstituted pyrrolyl, substituted or unsubstituted pyrazolyl, substitute or unsubstituted pyridyl, substituted or unsubstituted pyrimidinyl, substituted or unsubstituted imidazolyl, substituted or unsubstituted pyrazinyl, substituted or unsubstituted pyridazinyl, substituted or unsubstituted purinyl, substituted or unsubstituted indolyl, substituted or unsubstituted quinolinyl, substituted or unsubstituted isoquinolinyl, substituted or unsubstituted triazolyl, or substituted or unsubstituted tetrazolyl.

10. The compound according to claim 1 , wherein said R 1 substituted alkyl group is substituted with one or more aryl or heteroaryl groups.

11. The compound according to claim 10 , wherein said R 1 substituted alkyl group is substituted with one or more aryl or heteroaryl groups substituted with one or more halogens.

12. The compound according to claim 1 , wherein L 3 is a bond, or substituted or unsubstituted alkylene, and R 3 is substituted or unsubstituted aryl or substituted or unsubstituted heteroaryl.

13. A compound with structure of the following formula:

or pharmaceutically acceptable salt, ester, solvate, or prodrug thereof;

wherein:

L 1 is a bond, substituted or unsubstituted alkylene, substituted or unsubstituted heteroalkylene, —S—, —SO—, —SO 2 —, —NHSO 2 —, or —NR 5 —;

L 2 is a bond;

L 3 is —C(O)O—;

L 4 is a bond, substituted or unsubstituted alkylene, substituted or unsubstituted heteroalkylene, —S—, —SO—, —SO 2 —, —O—, —NHSO 2 —, or —NR 5 —;

R 1 is substituted or unsubstituted alkyl, substituted or unsubstituted heteroalkyl having at least one heteroatom selected from the group consisting of N, P, Si, and S, substituted or unsubstituted cycloalkyl, substituted or unsubstituted heterocycloalkyl, substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl, wherein the substituted alkyl, substituted heteroalkyl, substituted cycloalkyl, substituted heterocycloalkyl, substituted aryl, or substituted heteroaryl has one or more substituent group selected from the group consisting of —OH, —NH 2 , —SH, —CN, —CF 3 , —NO 2 , oxo, halogen, —COOH, substituted or unsubstituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted heterocycloalkyl, substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl;

when L 1 is a substituted or unsubstituted alkylene, R 1 is substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, unsubstituted heterocycloalkyl, or substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl; and

14. A compound with structure of the following formula:

or pharmaceutically acceptable salt, ester, solvate, or prodrug thereof;

wherein:

L 1 is a bond, substituted or unsubstituted alkylene, substituted or unsubstituted heteroalkylene, —S—, —SO—, —SO 2 —, —NHSO 2 —, or —NR 5 —;

L 2 is a bond;

L 3 —C(O)NR 6 , where R 6 is hydrogen or akyl;

L 4 is a bond; substituted or unsubstituted alkylene, substituted or unsubstituted heteroalkylene, —S—, —SO—, —SO 2 —, —O—, —NHSO 2 —, or —NR 5 —;

R 5 is hydrogen, or substituted or unsubstituted alkyl;

provided that:

L 1 is a bond, R 1 is substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, or substituted or unsubstituted heterocycloalkyl,

when L 1 is a substituted or unsubstituted alkylene, R 1 is substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, unsubstituted heterocycloalkyl, or substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl; and

when L 1 is —S—or substituted or unsubstituted heteroalkylene wherein the heteroatom is S, R 1 is substituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted heterocycloalkyl, substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl.

15. The compound according to claim 1 , wherein R 3 is substituted or unsubstituted phenyl or substituted or unsubstituted pyridyl.

16. The compound according to claim 7 , wherein L 1 is —S—, —NR 5 , substituted or unsubstituted alkylene, or substituted or unsubstituted heteroalkylene, and R 1 is substituted or unsubstituted aryl, substituted or unsubstituted heteroaryl, or unsubstituted heterocycloalkyl.

17. The compound according to claim 1 , wherein R 2 is substituted or unsubstituted cycloalkyl, substituted or unsubstituted cycloalkenyl, substituted or unsubstituted heterocycloalkyl, or substituted or unsubstituted heterocycloalkenyl.

18. The compound according to claim 1 , wherein R 2 is substituted or unsubstituted heteroaryl.

19. The compound according to claim 18 , wherein R 2 is substituted or unsubstituted furyl, substituted or unsubstituted thienyl, or substituted or unsubstituted pyridyl.

20. A compound with structure of the following formula:

or pharmaceutically acceptable salt, ester, solvate, or prodrug thereof;

wherein:

L 1 is a bond, substituted or unsubstituted alkylene, substituted or unsubstituted heteroalkylene, —S—, —SO—, —SO 2 —, —NHSO 2 —, or —NR 5 —;

L 2 is a bond;

L 3 is a bond, substituted or unsubstituted alkylene, substituted or unsubstituted heteroalkylene, —S—, —SO—, —SO 2 —, —O—, —NHSO 2 —, or —NR 5 —;

L 4 is a bond; substituted or unsubstituted alkylene, substituted or unsubstituted heteroalkylene, —S—, —SO—, —SO 2 —, —O—, —NHSO 2 —, or —NR 5 —;
