Abstract

The present invention relates to compositions and methods for preventing, treating or delaying various cardiovascular diseases or disorders in mammals, particularly in humans. More particularly, the present invention provides for compositions and methods for preventing, treating or delaying various cardiovascular diseases or disorders using, inter alia, a neuregulin protein, or a functional fragment thereof, or a nucleic acid encoding a neuregulin protein, or a functional fragment thereof, or an agent that enhances production and/or function of said neuregulin.

Claims (17)

1. A method for preventing, treating or delaying cardiac toxicity in a human who has been administered an anti-neoplasm agent, the method comprising administering to the human a neuregulin 1 protein in an amount from about 10 U/kg to about 400 U/kg of body weight of the human once a day for a period of between 3 days and 21 days.

2. The method of claim 1 , wherein the cardiac toxicity to be prevented, treated or delayed comprises a decrease in left ventricular ejection fraction (LVEF), a decrease in stroke volume (SV), a decrease in cardiac output (CO), or a decrease in cardiac index (CI).

3. The method of claim 1 , wherein the cardiac toxicity to be prevented, treated or delayed comprises inhibition of the contracting and pumping ability of left ventricle.

4. The method of claim 1 , wherein the anti-neoplasm agent is an anthracycline anti-neoplasm agent.

5. The method of claim 4 , wherein the anthracycline anti-neoplasm agent comprises doxorubicin, daunorubicin, epirubicin, or idarubicin.

6. The method of claim 5 , wherein the anthracycline anti-neoplasm agent is doxorubicin.

7. The method of claim 1 , wherein the anti-neoplasm agent comprises mitoxantrone, mitomycin, bleomycin, cyclophosphamide, fluorouracil, actinomycin D, or vincristine.

8. The method of claim 1 , wherein the neuregulin 1 protein comprises neuregulin α2 or neuregulin β2.

9. The method of claim 8 , wherein the neuregulin 1 protein is a neuregulin β2 comprising an amino acid sequence set forth in SEQ ID NO:4.

10. The method of claim 1 , wherein the neuregulin 1 protein is administered as a pharmaceutical composition.

11. The method of claim 10 , where said pharmaceutical composition comprises a pharmaceutically acceptable carrier or excipient.

12. The method of claim 1 , wherein the neuregulin 1 protein is administered prior to the administration of the anti-neoplasm agent.

13. The method of claim 1 , wherein the neuregulin 1 protein is administered concurrently with the administration of the anti-neoplasm agent.

14. The method of claim 1 , wherein the neuregulin 1 protein is administered subsequent to the administration of the anti-neoplasm agent.

15. The method of claim 1 , wherein the anti-neoplasm agent is administered in an amount that is greater than a maximally allowed amount compared to its administration in the absence of the neuregulin 1 protein.

16. The method of claim 1 , wherein the human has a malignant tumor.

17. The method of claim 16 , wherein the malignant tumor comprises lung cancer, breast cancer, bladder carcinoma, testis carcinoma, thyroid cancer, soft tissue carcinoma, osteosarcoma, neuroblastoma, acute leukemia, malignant lymphoma, gastric carcinoma, liver cancer, esophageal carcinoma, or cervical carcinoma.
