Abstract

Novel adeno-associated virus (AAV) vectors in nucleotide and amino acid forms and uses thereof are provided. The isolates show specific tropism for certain target tissues, such as blood stem cells, liver, heart and joint tissue, and may be used to transduce stem cells for introduction of genes of interest into the target tissues. Certain of the vectors are able to cross tightly controlled biological junctions, such as the blood-brain barrier, which open up additional novel uses and target organs for the vectors, providing for additional methods of gene therapy and drug delivery.

Claims (18)

1. A recombinant adeno-associated virus (rAAV) vector comprising a capsid protein comprising an amino acid sequence having at least 95% sequence identity with the amino acid sequence of amino acids 203-736 of SEQ ID NO:2, wherein the amino acid sequence of the capsid protein comprises: C at position 206; H at position 296; Q at position 312; A at position 346; N at position 464; S at position 468; I at position 501; R at position 505; R at position 590; G at position 626; Y at position 626; M at position 681; R at position 687; K at position 690; C at position 706; or G at position 718.

2. The rAAV vector of claim 1 , wherein the amino acid sequence of the capsid protein comprises: G at position 626 and G at position 718; H at position 296, N at position 464, R at position 505, and M at position 681; R at position 505 and R at position 687; A at position 346 and R at position 505; or I at position 501, R at position 505, and C at position 706.

3. The rAAV vector of claim 1 , wherein the capsid protein comprises the amino acid sequence of amino acids 203-736 of SEQ ID NO: 2, 3, 4, 6, 7, 10, 11, 12, 13, 14, 15, 16, or 17.

A recombinant adeno-associated virus (rAAV) vector comprising a capsid protein comprising an amino acid sequence having at least 95% sequence identity with the amino acid sequence of amino acids 138-736 of SEQ ID NO:2, wherein the amino acid sequence of the capsid protein comprises: R at position 151; D at position 160; C at position 206; H at position 296; Q at position 312; A at position 346; N at position 464; S at position 468; I at position 501; Rat position 505; Rat position 590; G at position 626; Y at position 626; Mat position 681; R at position 687; K at position 690; C at position 706; or G at position 718.

The rAAV vector of claim 4 , wherein the amino acid sequence of the capsid protein comprises: G at position 626 and G at position 718; H at position 296, N at position 464, R at position 505, and M at position 681; R at position 505 and R at position 687; A at position 346 and R at position 505; or I at position 501, R at position 505, and C at position 706.

6. The rAAV vector of claim 4 , wherein the capsid protein comprises the amino acid sequence of amino acids 138-736 of SEQ ID NO: 2, 3, 4, 5, 6, 7, 9, 10, 11, 12, 13, 14, 15, 16, or 17.

A recombinant adeno-associated virus (rAAV) vector comprising a capsid protein comprising an amino acid sequence having at least 95% sequence identity with the amino acid sequence of amino acids 1-736 of SEQ ID NO:2, wherein the amino acid sequence of the capsid protein comprises: T at position 2; I at position 65; V at position 68; Rat position 77; L at position 119; R at position 151; D at position 160; C at position 206; H at position 296; Q at position 312; A at position 346; N at position 464; S at position 468; I at position 501; R at position 505; R at position 590; G at position 626; Y at position 626; M at position 681; R at position 687; K at position 690; C at position 706; or G at position 718.

The rAAV vector of claim 7 , wherein the amino acid sequence of the capsid protein comprises: T at position 2 and Q at position 312; I at position 65 and Y at position 626; R at position 77 and K at position 690; L at position 119 and S at position 468; G at position 626 and G at position 718; H at position 296, N at position 464, R at position 505, and M at position 681; R at position 505 and R at position 687; A at position 346 and R at position 505; or I at position 501, R at position 505, and C at position 706.

9. The rAAV vector of claim 7 , wherein the capsid protein comprises the amino acid sequence of amino acids 1-736 of SEQ ID NO: 2, 3, 4, 5, 6, 7, 8, 9, 10, 11, 12, 13, 14, 15, 16, or 17.

10. The rAAV vector of claim 1 , further comprising a therapeutic nucleotide sequence.

11. The rAAV vector of claim 2 , further comprising a therapeutic nucleotide sequence.

12. The rAAV vector of claim 3 , further comprising a therapeutic nucleotide sequence.

13. The rAAV vector of claim 4 , further comprising a therapeutic nucleotide sequence.

14. The rAAV vector of claim 5 , further comprising a therapeutic nucleotide sequence.

15. The rAAV vector of claim 6 , further comprising a therapeutic nucleotide sequence.

16. The rAAV vector of claim 7 , further comprising a therapeutic nucleotide sequence.

17. The rAAV vector of claim 8 , further comprising a therapeutic nucleotide sequence.

18. The rAAV vector of claim 9 , further comprising a therapeutic nucleotide sequence.
