Abstract

Provided herein are integrated continuous biomanufacturing processes for producing a therapeutic protein drug substance. Also provided are systems that are capable of continuously producing a therapeutic protein drug substance.

Claims (39)

1. A biological manufacturing system for obtaining a recombinant therapeutic protein in a fluid, comprising:

a first periodic counter current chromatography system (PCCCS1) comprising an inlet; and

a second periodic counter current chromatography system (PCCCS2) comprising an outlet, wherein

the PCCCS1 and the PCCCS2 are in fluid communication with each other;

the biological manufacturing system is configured such that the fluid is passed into the inlet, through the PCCCS1 and the PCCCS2, and exits the biological manufacturing system through the outlet;

the PCCCS1 is configured to perform at least the unit operation of capturing the recombinant therapeutic protein by a resin; and

the PCCCS2 is configured to perform at least two unit operations of purifying and polishing the recombinant therapeutic protein by resins, wherein the resin used in purifying is different in chemical structure compared to the resin used in the polishing.

2. The biological manufacturing system of claim 1 , further comprising a bioreactor, wherein the bioreactor and the inlet are in fluid communication with each other, and wherein the manufacturing system is configured such that fluid present in the bioreactor can be passed into the inlet.

3. The biological manufacturing system of claim 1 , wherein the PCCCS1 is configured to perform at least two separate unit operations.

4. The biological manufacturing system of claim 3 , wherein the PCCCS1 is configured to perform two unit operations of capturing the recombinant therapeutic protein and inactivating viruses.

5. The biological manufacturing system of claim 1 , wherein the use of the PCCCS1 or the PCCCS2 involves column switching, or the use of each of the PCCCS1 and the PCCCS2 involves column switching.

6. The biological manufacturing system of claim 4 , wherein the PCCCS1 comprises four columns.

7. The biological manufacturing system of claim 6 , wherein the three columns in the four columns are capable of capturing the recombinant therapeutic protein from a liquid culture medium.

8. The biological manufacturing system of claim 7 , wherein the fourth column of the four columns is a reservoir that is capable of holding eluate from the three columns containing the recombinant therapeutic protein at a pH of about 3.0 to 5.0.

9. The biological manufacturing system of claim 8 , wherein the PCCCS2 comprises three chromatography columns and a chromatographic membrane.

10. The biological manufacturing system of claim 9 , further comprising a fluid conduit disposed between the three chromatography columns in the PCCCS2 and the chromatographic membrane in the PCCCS2.

The biological manufacturing system of claim 10 , further comprising an in-line buffer adjustment reservoir in fluid communication with the fluid conduit disposed between the three chromatography columns in the PCCCS2 and the chromatographic membrane in the PCCCS2, and configured such that buffer contained within the in-line buffer adjustment reservoir is introduced into the fluid present in the fluid conduit disposed between the three chromatography columns in the PCCCS2 and the chromatographic membrane in the PCCCS2.

12. The biological manufacturing system of claim 9 , wherein the three chromatography columns in the PCCCS2 are capable of purifying the recombinant therapeutic protein through cation or anion exchange chromatography.

13. The biological manufacturing system of claim 9 , wherein the chromatographic membrane in the PCCCS2 is a cation exchange chromatographic membrane.

14. The biological manufacturing system of claim 9 , further comprising a fluid conduit between the chromatographic membrane in the PCCCS2 and the outlet.

The biological manufacturing system of claim 14 , further comprising a filter disposed in the fluid conduit between the chromatographic membrane in the PCCCS2 and the outlet, and configured such that the filter is capable of removing particulate matter from the fluid present in the fluid conduit between the chromatographic membrane in the PCCCS2 and the outlet.

16. The biological manufacturing system of claim 1 , further comprising a pump system that is in fluid communication with the inlet.

17. The biological manufacturing system of claim 16 , wherein the pump system comprises a pump that is capable of pushing the fluid into the inlet.

18. The biological manufacturing system of claim 16 , further comprising a fluid conduit disposed between the pump and the inlet.

19. The biological manufacturing system of claim 18 , further comprising a filter disposed in the fluid conduit between the pump and the inlet, and configured such that the filter is capable of removing particulate matter from the fluid present in the fluid conduit between the pump and the inlet.

20. The biological manufacturing system of claim 1 , further comprising a fluid conduit disposed between the PCCCS1 and the PCCCS2.

The biological manufacturing system of claim 20 , further comprising an in-line buffer adjustment reservoir in fluid communication with the fluid conduit disposed between the PCCCS1 and the PCCCS2, and configured such that buffer contained within the in-line buffer adjustment reservoir is introduced into the fluid present in the fluid conduit disposed between the PCCCS1 and the PCCCS2.

The biological manufacturing system of claim 20 , further comprising a filter disposed in the fluid conduit between the PCCCS1 and the PCCCS2, and configured such that the filter is capable of removing particulate matter from the fluid present in the fluid conduit between the PCCCS1 and the PCCCS2.

23. The biological manufacturing system of claim 1 , wherein the PCCCS1 is configured to perform at least two separate unit operations.

24. A biological manufacturing system for obtaining a recombinant therapeutic protein in a fluid, comprising two or more subsystems, wherein each of the two or more subsystems comprises:

(i) a first periodic counter current chromatography system (PCCCS1) comprising an inlet; and

(ii) a second periodic counter current chromatography system (PCCCS2) comprising an outlet, wherein the PCCCS1 and the PCCCS2 are in fluid communication with each other, and wherein the biological manufacturing system is configured such that fluid can be passed into the inlet, through the PCCCS1 and the PCCCS2, and exit the biological manufacturing system through the outlet;

wherein:

the two or more subsystems are configured such that they are each in fluid communication with a single reservoir containing the fluid;

the fluid from the single reservoir passes into the inlet of the two or more subsystems;

the PCCCS1 in each of the two or more subsystems is configured to perform at least the unit operation of capturing the recombinant therapeutic protein by a resin; and

the PCCCS2 in each of the two or more subsystems is configured to perform at least two unit operations of purifying and polishing the recombinant therapeutic protein by resins, wherein the resin used in purifying is different in chemical structure compared to the resin used in the polishing.

25. The biological manufacturing system of claim 24 , wherein each of the two or more subsystems is placed on its own skid.

26. The biological manufacturing system of claim 24 , wherein the entire system is placed on a skid.
