Abstract

This invention provide novel compositions, methods and devices for sustained drug delivery. The compositions can include a fluid carrier; a plurality of first polymeric microparticles having a bioactive agent dispersed in the fluid carrier; and a plurality of second polymeric microparticles having a visualization agent dispersed in the fluid carrier with the first microparticles. Alternative compositions can include: a fluid carrier; and a plurality of polymeric microparticles having a bioactive agent encapsulated therein and a visualization agent on a surface of the polymeric microparticles, the plurality of polymeric microparticles being dispersed in the fluid carrier. The microencapsulated sustained drug delivery compositions are deposited using oscillating needle apparatus oscillating at 10-12000 minutes per minutes at an amount of 1.0E-03 mL to 1.0E-16 mL per injection.

Claims (49)

1. An injectable composition comprising:

a fluid carrier;

a plurality of first polymeric microparticles having a bioactive agent dispersed in the fluid carrier; and

a plurality of second polymeric microparticles having a visualization agent dispersed in the fluid carrier with the first microparticles.

2. The injectable composition of claim 1 , wherein:

the first polymeric microparticles are devoid of a visualization agent; and

the second polymeric microparticles are devoid of a bioactive agent.

3. The injectable composition of claim 1 , wherein the plurality of second polymeric microparticles have the visualization agent coated, stained or covalently linked to a biodegradable polymer that forms the second polymeric microparticles.

4. The injectable composition of claim 1 , wherein at least one of the first polymeric microparticles or second polymeric microparticles are prepared from a biodegradable polymer.

5. The injectable composition of claim 4 , wherein the biodegradable polymer is:

selected from a group consisting of polymers, dendramers, copolymers or oligomers of glycolide, dl-lactide, d-lactide, l-lactide, caprolactone, dioxanone and trimethylene carbonate; degradable polyurethanes; polyamides; polyesters; polypeptides; polyhydroxyacids; polyorthocarbonates, polylactic acid; polyglycolic acid; polyanhydrides; and polylactones; or

a copolymer of polyethylene glycol and polylactone or copolymer of PEO-PPO-PEO and polylactone.

6. The injectable composition of claim 4 , wherein the biodegradable polymer is a biodegradable hydrogel.

7. The injectable composition of claim 6 , wherein the biodegradable hydrogel is a biodegradable crosslinked hydrogel or a biodegradable hydrogel having polyethylene glycol.

8. The injectable composition of claim 1 , wherein at least one of the first polymeric microparticles or second polymeric microparticles have:

an average particle size in the range of 500 nm to 300 microns;

substantially spherical, semispherical, elliptical, circular disk, or irregular shape;

the bioactive agent is selected from the group consisting of: antiinfectives, antibiotics; antiviral agents, antifungal agents, antibacterial agents, antipruritics; anticancer agents, antipsychotics; cholesterol- or lipid-reducing agents; cell cycle inhibitors; anticancer agents; antiparkinsonism drugs; HMG-CoA inhibitors; antirestenosis agents; antiinflammatory agents; antiasthmatic agents; anthelmintic; immunosuppressives; muscle relaxants; antidiuretic agents; vasodilators; nitric oxide; nitric oxide-releasing compounds; beta-blockers; hormones; antidepressants; decongestants; calcium channel blockers; growth factors, bone growth factors or bone morphogenic proteins; wound healing agents; analgesics and analgesic combinations; local anesthetic agents; antihistamines; sedatives; angiogenesis-promoting agents; angiogenesis-inhibiting agents; tranquilizers;

the bioactive agent at about 1% to 50% relative to weight of polymer; or

the first polymeric microparticles or second polymeric microparticles are present from 1% to 99% of total microparticles.

9. The injectable composition of claim 1 , wherein the visualization agent is a coloring composition, a fluorescent composition, a radio opaque contrast agent, or an NMR contrast agent.

10. The injectable composition of claim 1 , wherein the fluid carrier is a buffered aqueous solution.

11. An injectable composition comprising:

a fluid carrier; and

a plurality of polymeric microparticles having a bioactive agent encapsulated therein and a visualization agent on a surface of the polymeric microparticles, the plurality of polymeric microparticles being dispersed in the fluid carrier.

12. The injectable composition of claim 11 , wherein the plurality of second polymeric microparticles have the visualization agent coated, stained or covalently linked to a biodegradable polymer that forms the second polymeric microparticles.

13. The injectable composition of claim 11 , wherein the polymeric mi crop articles are prepared from a biodegradable polymer.

14. The injectable composition of claim 13 , wherein the biodegradable polymer is:

selected from a group consisting of polymers, dendramers, copolymers or oligomers of glycolide, dl-lactide, d-lactide, 1-lactide, caprolactone, dioxanone and trimethylene carbonate; degradable polyurethanes; polyamides; polyesters; polypeptides; polyhydroxy acids; polyorthocarbonates, polylactic acid; polyglycolic acid; polyanhydrides; and polylactones; or

a copolymer of polyethylene glycol and polylactone or copolymer of PEO-PPO-PEO and polylactone.

15. The injectable composition of claim 13 , wherein the biodegradable polymer is a biodegradable hydrogel.

16. The injectable composition of claim 15 , wherein the biodegradable hydrogel is a biodegradable crosslinked hydrogel or a biodegradable hydrogel having polyethylene glycol.

17. The injectable composition of claim 11 , wherein the polymeric mi crop articles have:

an average particle size in the range of 500 nm to 300 microns;

substantially spherical, semi spherical, elliptical, circular disk, or irregular shape;

the bioactive agent is selected from the group consisting of: antiinfectives, antibiotics; antiviral agents, antifungal agents, antibacterial agents, antipruritics; anticancer agents, antipsychotics; cholesterol- or lipid-reducing agents; cell cycle inhibitors; anticancer agents; antiparkinsonism drugs; HMG-CoA inhibitors; antirestenosis agents; antiinflammatory agents; antiasthmatic agents; anthelmintic; immunosuppressives; muscle relaxants; antidiuretic agents; vasodilators; nitric oxide; nitric oxide-releasing compounds; beta-blockers; hormones; antidepressants; decongestants; calcium channel blockers; growth factors, bone growth factors or bone morphogenic proteins; wound healing agents; analgesics and analgesic combinations; local anesthetic agents; antihistamines; sedatives; angiogenesis-promoting agents; angiogenesis-inhibiting agents; tranquilizers; or

the bioactive agent at about 1% to 50% relative to weight of polymer.

18. The injectable composition of claim 1 , wherein the visualization agent is a coloring composition, a fluorescent composition, a radio opaque contrast agent, or an NMR contrast agent.

19. The injectable composition of claim 1 , wherein the fluid carrier is a buffered aqueous solution.

20. A method of delivering an injectable composition at a local site of a tissue, the method comprising:

providing the injectable composition of claim 1 ; and

injecting the injectable composition into the tissue.

21. The method of claim 20 , wherein the injecting is performed at an amount of 1.0E-03 mL to 1.0E-16 mL per injection.

22. The method of claim 20 , wherein the tissue is live tissue or bioprosthesis tissue.

23. A method of delivering an injectable composition at a local site of a tissue, the method comprising:

providing the injectable composition of claim 11 ; and

injecting the injectable composition into the tissue.

24. The method of claim 20 , wherein the injecting is performed at an amount of 1.0E-03 mL to 1.0E-16 mL per injection.

25. The method of claim 20 , wherein the tissue is live tissue or bioprosthesis tissue.
