Abstract

The present invention relates generally to compositions and methods for treating cancer and neoplastic diseases. Provided herein are substituted imidazole-pyridine derivative compounds and pharmaceutical compositions comprising said compounds. The subject compounds and compositions are useful for inhibition of histone demethylase enzymes. Furthermore, the subject compounds and compositions are useful for the treatment of cancer, such as pancreatic cancer, prostate cancer, breast cancer, bladder cancer, lung cancer, gastric cancer, leukemia and/or melanoma and the like.

Claims (12)

1. A compound having the structure of Formula (Ic),

or a pharmaceutically acceptable salt thereof, wherein

R 1 is aralkyl, or aryl optionally substituted by one of more of the following substituents: halogen, hydroxy, —CN, alkyl, alkoxy, —O-(cycloalkylalkyl), alkylamino, aryl, carbocyclyl, or carbocyclylalkyl;

R 2 is C 1 alkyl;

R 3 is hydrogen; and

R 4 is halogen, or C 1 -C 4 alkyl, optionally substituted by halogen.

2. The compound of claim 1 , or a pharmaceutically acceptable salt thereof, wherein R 1 is aryl.

3. The compound of claim 2 , or a pharmaceutically acceptable salt thereof, wherein R 1 is an aryl, and wherein the aryl is a phenyl optionally substituted with at least one of halogen, hydroxy, —CN, alkyl, alkoxy, —O-(cycloalkylalkyl), alkylamino, aryl, carbocyclyl, or carbocyclylalkyl.

4. The compound of claim 1 , or a pharmaceutically acceptable salt thereof, wherein R 4 is C 1 -C 4 alkyl, and the alkyl is substituted with at least one fluoro.

5. The compound of claim 4 , or a pharmaceutically acceptable salt thereof, wherein R 4 is CH 2 F, CHF 2 , or CF 3 .

The compound of claim 1 , or a pharmaceutically acceptable salt thereof, wherein said compound is selected from the group consisting of 4-(5-chloro-1H-triazol-4-yl)-2-[5-[2-(cyclopropylmethoxy)-4-fluorophenyl]-1-methylimidazol-4-yl]pyridine, and 2-[5-(4-fluorophenyl)-1-methyl-1H-imidazol-4-yl]-4-[5-(trifluoromethyl)-1H-1,2,3-triazol-4-yl]pyridine.

7. A pharmaceutical composition comprising the compound of claim 1 , or a pharmaceutically acceptable salt thereof.
