Abstract

Disclosed herein are compounds, compositions, and methods for preventing and treating proliferative diseases associated with aberrant receptor tyrosine kinase (RTK) activity. The therapeutic indications described herein more specifically relate to the non-selective inhibition of RTKs associated with vascular and pulmonary disorders.

Claims (27)

1. A formulation comprising:

a) a compound of the formula:

or a pharmaceutically-acceptable salt thereof; and

b) at least one excipient, wherein the excipient comprises a hydrophobic amino acid, wherein the formulation is a powder.

2. The formulation of claim 1 , wherein the powder has a moisture content below about 5% by water weight.

3. The formulation of claim 1 , wherein the hydrophobic amino acid is leucine.

4. The formulation of claim 1 , wherein the formulation further comprises a carbohydrate bulking agent.

5. The formulation of claim 4 , wherein the carbohydrate bulking agent is trehalose.

6. The formulation of claim 1 , wherein the formulation comprises two excipients.

7. The formulation of claim 6 , wherein the excipients are leucine and trehalose.

8. The formulation of claim 1 , wherein the powder has an average particle size of about 0.5 to 10 microns.

9. The formulation of claim 1 , wherein the compound is present in an amount from 12.67% to 61.73%.

10. A method of treating pulmonary arterial hypertension in a subject in need thereof, the method comprising administering to the subject a formulation comprising:

a) a therapeutically-effective amount of a compound of the formula:

or a pharmaceutically-acceptable salt thereof; and

b) at least one excipient, wherein the excipient comprises a hydrophobic amino acid, wherein the formulation is a powder.

11. The method of claim 10 , wherein the administration is by inhalation.

12. The method of claim 10 , wherein the administration is intranasal.

13. The method of claim 10 , wherein the administration is by an inhaler.

14. The method of claim 10 , wherein the administration is by a nebulizer.

15. The method of claim 10 , wherein the administration is by an atomizer.

16. The method of claim 10 , wherein the therapeutically-effective amount of the compound is from about 0.01 mg/kg to about 100 mg/kg per day.

17. The method of claim 10 , wherein the formulation provides a C max of about 1 to 5000 ng/mL of the compound in the subject's plasma after administration to the subject.

18. The method of claim 10 , wherein the formulation provides a C max of about 1 to 5000 ng/mL of the compound in the subject's plasma 24 h after administration to the subject.

19. The method of claim 10 , wherein the hydrophobic amino acid is leucine.

20. The method of claim 10 , wherein the formulation further comprises a carbohydrate bulking agent.

21. The method of claim 20 , wherein the carbohydrate bulking agent is trehalose.
