Abstract

This disclosure relates generally to a sampling device, and more particularly, a sampling device that facilitates spectroscopic measurements with a variable path length and the necessary software controlled algorithms and methods for such a device.

Claims (26)

1. A method of determining the concentration of a compound in a solution comprising:

(a) taking a first absorbance reading at a predetermined wavelength of the compound in the solution at a first pathlength;

(b) changing the first pathlength by an increment to provide a second pathlength and taking a second absorbance reading at the predetermined wavelength;

(c) repeating steps (a) and (b) one or more times;

(d) generating a regression line from the absorbance values such that a slope of the regression line is obtained; and

(e) determining the concentration of the sample by dividing the slope of the regression line by the extinction coefficient of the compound in the solution.

2. The method of claim 1 wherein the increment is the same for each iteration.

3. The method of claim 1 wherein the increment is from about 0.005 mm to about 50 mm.

4. The method of claim 1 wherein the increment is from about 0.0002 mm to about 10 mm.

5. The method of claim 1 wherein the regression line has an R 2 value of from around 0.99950 to about 0.99999.

6. The method of claim 1 wherein the regression line has an R 2 value of from around 0.99990 to about 0.99999.

7. The method of claim 1 , wherein the pathlength is changed by moving a probe relative to a detector.

8. A method of determining the extinction coefficient of a compound in a solution at a wavelength where the extinction coefficient is not known comprising:

(a) taking a first absorbance reading of the solution at a first predetermined wavelength where the extinction coefficient is known and a second predetermined wavelength where the extinction coefficient is not known at a given pathlength;

(b) repeating step (c) one or more times to determine the ratio of the absorbance to the path length at the first wavelength and the second wavelength;

(c) calculate the extinction coefficient at the second wavelength from the ratios of the absorbance to path length at the first and second wavelengths and the extinction coefficient for the first wavelength.

9. A method of determining the concentration of a compound in a solution comprising:

(a) taking an first absorbance spectrum of the compound in the solution at a first pathlength;

(b) changing the first pathlength by an increment to provide a second pathlength and taking a second absorbance spectrum reading at the predetermined wavelength;

(c) repeating steps (a) and (b) one or more times;

(d) generating a regression line from the absorbance values at a given wavelength such that a slope of the regression line is obtained; and

(e) determining the concentration of the sample by dividing the slope of the regression line by the extinction coefficient of the compound in the solution.

10. The method of claim 9 further comprising baseline correction.

11. The method of claim 10 wherein the baseline correction comprises subtracting a first baseline spectrum from the first absorbance spectrum and subtracting a second baseline spectrum from the second absorbance spectrum.

12. The method of claim 9 further comprising scatter correction.

13. The method of claim 12 wherein the scatter correction comprises applying a scatter correction algorithm to the first absorbance spectrum and to the second absorbance spectrum.
