Abstract

The present invention relates to anti-PHF-tau antibodies and methods of making and using them.

Claims (18)

1. An isolated antibody that binds paired helical filament tau (PHF-tau) comprising a heavy chain complementarity determining region (HCDR) 1, a heavy chain complementarity determining region (HCDR) 2 and a heavy chain complementarity determining region (HCDR) 3 of a heavy chain variable region (VH) of SEQ ID NO: 35, and a light chain complementarity determining region (LCDR) 1, a light chain complementarity determining region (LCDR) 2 and a light chain complementarity determining region (LCDR) 3 of a light chain variable region (VL) of SEQ ID NO: 36.

2. A pharmaceutical composition comprising the antibody of claim 1 and a pharmaceutically acceptable carrier.

3. The isolated antibody of claim 1 , wherein the antibody is humanized.

4. A vector comprising a polynucleotide encoding an antibody of claim 1 .

5. A host cell comprising the vector of claim 4 .

6. The isolated antibody of claim 1 , wherein the HCDR1, the HCDR2 and the HCDR3 comprise amino acid sequences of SEQ ID NOs:19, 20 and 21, respectively, and the LCDR1, the LCDR2 and the LCDR3 comprise amino acid sequences of SEQ ID NOs: 22, 23 and 24, respectively.

7. The isolated antibody of claim 6 , wherein the antibody is humanized.

8. A pharmaceutical composition comprising the antibody of 6 and a pharmaceutically acceptable carrier.

9. A vector comprising a polynucleotide encoding an antibody of claim 6 .

10. A host cell comprising the vector of claim 9 .

11. The isolated antibody of claim 1 , wherein the HCDR1, the HCDR2 and the HCDR3 comprise amino acid sequences of SEQ ID NOs:7, 8 and 9, respectively, and the LCDR1, the LCDR2 and the LCDR3 comprise amino acid sequences of SEQ ID NOs: 10, 11 and 12, respectively.

12. The isolated antibody of claim 11 , wherein the antibody is humanized.

13. A pharmaceutical composition comprising the antibody of 11 and a pharmaceutically acceptable carrier.

14. A vector comprising a polynucleotide encoding an antibody of claim 11 .

15. A host cell comprising the vector of claim 14 .

16. An isolated polynucleotide encoding an antibody heavy chain comprising a VH of the antibody of claim 11 .

17. An isolated polynucleotide encoding an antibody light chain comprising a VL of the antibody of claim 11 .

18. A host cell comprising the isolated polynucleotide of claim 16 or 17 .
