Abstract

The present invention relates to novel 6,7-dihydropyrazolo[1,5-a]pyrazin-4(5H)-one derivatives of formula (I) as negative allosteric modulators (NAMs) of the metabotropic glutamate receptor subtype 2 (“mGluR2”) useful for the treatment of central nervous system disorders in which the mGluR2 subtype of metabotropic receptors is involved (e.g. mood disorder, schizophrenia, etc.). The invention is also directed to pharmaceutical compositions comprising such compounds, to processes for preparing such compounds and compositions, and to the use of such compounds and compositions for treatment of central nervous system disorders in which the mGluR2 subtype of metabotropic receptors is involved.

Claims (114)

1. A compound of Formula (I):

or a stereoisomeric form thereof,

wherein:

R 1 is phenyl or 2-pyridinyl, each optionally substituted with one or more substituents independently selected from the group consisting of halo, —C 1-4 alkyl, monohaloC 1-4 alkyl, polyhaloC 1-4 alkyl, —O—C 1-4 alkyl, —C 1-4 alkyl-O—C 1-4 alkyl, monohaloC 1-4 alkyloxy, polyhaloC 1-4 alkyloxy, —C 1-4 alkyl-OH, —C 1-4 alkylthio, monohaloC 1-4 alkylthio, polyhaloC 1-4 alkylthio, cyano, —C 3-7 cycloalkyl optionally substituted with trifluoromethyl and —SF 5 ; or

R 1 is

R 2 is

R 3 is selected from the group consisting of hydrogen and —C 1-4 alkyl;

R 4 is selected from the group consisting of hydrogen, —C 1-4 alkyl, monohaloC 1-4 alkyl, polyhaloC 1-4 alkyl, —C 1-4 alkyl-O—C 1-4 alkyl and —C 1-4 alkyl-OH;

R 5 and R 6 are each independently selected from the group consisting of hydrogen, halo, cyano, —C 1-4 alkyl, —C 1-4 alkyl-OH, —C 3-7 cycloalkyl, monohaloC 1-4 alkyl, polyhaloC 1-4 alkyl, —C 1-4 alkyl-O—C 1-4 alkyl, —O—C 1-4 alkyl, monohaloC 1-4 alkyloxy, polyhaloC 1-4 alkyloxy, 1-acetylazetidin-3-yl and —NR′R″; and

R′ and R″ are each independently selected from the group consisting of hydrogen and —C 1-4 alkyl; or

R′ and R″, together with the nitrogen atom to which they are attached, form a heterocyclic group selected from the group consisting of 1-azetidinyl, 1-pyrrolidinyl, 1-piperidinyl, 1-piperazinyl and 4-morpholinyl, wherein 1-azetidinyl, 1-pyrrolidinyl, 1-piperidinyl, 1-piperazinyl and 4-morpholinyl are optionally substituted with a substituent selected from the group consisting of halo, hydroxy, —C 1-4 alkyl, monohaloC 1-4 alkyl, polyhaloC 1-4 alkyl and —C(O)—C 1-4 alkyl;

or a N-oxide or a pharmaceutically acceptable salt thereof.

2. The compound of claim 1 , wherein:

R 1 is phenyl or 2-pyridinyl, each optionally substituted with one or more substituents independently selected from the group consisting of halo, —C 1-4 alkyl, monohaloC 1-4 alkyl, polyhaloC 1-4 alkyl, —O—C 1-4 alkyl, —C 1-4 alkyl-O—C 1-4 alkyl, monohaloC 1-4 alkyloxy, polyhaloC 1-4 alkyloxy, —C 1-4 alkyl-OH, monohaloC 1-4 alkylthio, polyhaloC 1-4 alkylthio, cyano and —SF 5 ; or

R 1 is

R 2 is

R 5 and R 6 are each independently selected from the group consisting of hydrogen, halo, cyano, —C 1-4 alkyl, —C 1-4 alkyl-OH, —C 3-7 cycloalkyl, monohaloC 1-4 alkyl, polyhaloC 1-4 alkyl, —C 1-4 alkyl-O—C 1-4 alkyl, —O—C 1-4 alkyl, monohaloC 1-4 alkyloxy, polyhaloC 1-4 alkyloxy and —NR′R″; and

R′ and R″ are each independently selected from the group consisting of hydrogen and C 1-4 alkyl; or

R′ and R″, together with the nitrogen atom to which they are attached, form a heterocyclic group selected from the group consisting of 1-azetidinyl, 1-pyrrolidinyl and 1-piperidinyl, wherein 1-azetidinyl, 1-pyrrolidinyl and 1-piperidinyl are optionally substituted with a halo substituent;

or a N-oxide or a pharmaceutically acceptable salt thereof.

3. The compound of claim 1 , wherein:

R 1 is phenyl, optionally substituted with one or more substituents independently selected from the group consisting of halo, —C 1-4 alkyl, monohaloC 1-4 alkyl, polyhaloC 1-4 alkyl, —O—C 1-4 alkyl, monohaloC 1-4 alkylthio, polyhaloC 1-4 alkylthio, cyano and —SF 5 ; or

R 1

R 2 is

R 5 and R 6 are each independently selected from the group consisting of hydrogen, —C 1-4 alkyl, monohaloC 1-4 alkyl, polyhaloC 1-4 alkyl, —C 1-4 alkyl-O—C 1-4 alkyl, —O—C 1-4 alkyl and —NR′R″; and

R′ and R″ are each independently selected from the group consisting of hydrogen and C 1-4 alkyl;

or a pharmaceutically acceptable salt thereof.

4. The compound of claim 1 , wherein:

R 1 is phenyl, optionally substituted with one or more substituents independently selected from the group consisting of halo, —C 1-4 alkyl, monohaloC 1-4 alkyl, polyhaloC 1-4 alkyl, —O—C 1-4 alkyl, monohaloC 1-4 alkyloxy, polyhaloC 1-4 alkyloxy, cyano and —SF 5 ; or

R 1 is

R 2 is

R 4 is selected from the group consisting of hydrogen and —C 1-4 alkyl;

R 5 and R 6 are each independently selected from the group consisting of hydrogen, —C 1-4 alkyl, —C 1-4 alkyl-O—C 1-4 alkyl and —NR′R″; and

R′ and R″ are each hydrogen;

or a pharmaceutically acceptable salt thereof.

5. The compound of claim 1 , wherein:

R 1 is phenyl, optionally substituted with one or more substituents independently selected from the group consisting of halo, —C 1-4 alkyl, polyhaloC 1-4 alkyl and —SF 5 ;

R 2 is

R 4 is hydrogen; and

R 5 and R 6 are each independently selected from the group consisting of hydrogen, —C 1-4 alkyl and —O—C 1-4 alkyl;

or a pharmaceutically acceptable salt thereof.

6. The compound of claim 1 , wherein R 1 is selected from the group consisting of

9. The compound of claim 7 , wherein the compound is the free base or a pharmaceutically acceptable salt selected from the group consisting of the hydrochloride salt, the sulfate salt, the methanesulfonate salt and the maleate salt.

10. The compound of claim 1 , wherein the compound is:

or a pharmaceutically acceptable salt thereof.

11. A pharmaceutical composition comprising a therapeutically effective amount of a compound of claim 1 and a pharmaceutically acceptable carrier or excipient.

12. A method for modulating metabotropic glutamate receptor 2 activity in a subject, comprising administering to a subject in need thereof a therapeutically effective amount of a compound of claim 1 .

13. The method of claim 12 , wherein the subject suffers from a central nervous system disorder or condition selected from the group consisting of a depressive disorder, a neurocognitive disorder, a neurodevelopmental disorder, a substance-related disorder, an addictive disorder, a psychotic disorder, a somatic symptom disorder and a hypersomnolence disorder.

14. The method of claim 13 , wherein the central nervous system disorder or condition is selected from the group consisting of depression, attention-deficit/hyperactivity disorder, schizophrenia and a neurocognitive disorder.

15. The method of claim 14 , wherein the depression is selected from the group consisting of major depressive disorder and treatment resistant depression.

16. The method of claim 12 , wherein the subject suffers from a central nervous system disorder or condition selected from the group consisting of a mood disorder, a cognitive disorder, a substance-related disorder, a psychotic disorder, a somatoform disorder and a hypersomnic sleep disorder.

17. The method of claim 12 wherein the central nervous system disorder or condition is selected from the group consisting of major depressive disorder, depression and treatment resistant depression.

18. A method for modulating metabotropic glutamate receptor 2 activity in a subject, comprising administering to a subject in need thereof a therapeutically effective amount of a compound of claim 7 .

19. The method of claim 18 , wherein the subject suffers from a central nervous system disorder or condition selected from the group consisting of major depressive disorder and treatment resistant depression.

20. A process for the preparation of a compound of Formula (I):

wherein:

R 1 is phenyl or 2-pyridinyl, each optionally substituted with one or more substituents independently selected from the group consisting of halo, —C 1-4 alkyl, monohaloC 1-4 alkyl, polyhaloC 1-4 alkyl, —O—C 1-4 alkyl, —C 1-4 alkyl-O—C 1-4 alkyl, monohaloC 1-4 alkyloxy, polyhaloC 1-4 alkyloxy, —C 1-4 alkyl-OH, —C 1-4 alkylthio, monohaloC 1-4 alkylthio, polyhaloC 1-4 alkylthio, cyano, —C 3-7 cycloalkyl optionally substituted with trifluoromethyl and —SF 5 ; or

R 1 is

R 2

R 3 is selected from the group consisting of hydrogen and —C 1-4 alkyl;

R 4 is selected from the group consisting of hydrogen, —C 1-4 alkyl, monohaloC 1-4 alkyl, polyhaloC 1-4 alkyl, —C 1-4 alkyl-O—C 1-4 alkyl and —C 1-4 alkyl-OH;

R 5 and R 6 are each independently selected from the group consisting of hydrogen, halo, cyano, —C 1-4 alkyl, —C 1-4 alkyl-OH, —C 3-7 cycloalkyl, monohaloC 1-4 alkyl, polyhaloC 1-4 alkyl, —C 1-4 alkyl-O—C 1-4 alkyl, —O—C 1-4 alkyl, monohaloC 1-4 alkyloxy, polyhaloC 1-4 alkyloxy, 1-acetylazetidin-3-yl and —NR′R″; and

R′ and R″ are each independently selected from the group consisting of hydrogen and —C 1-4 alkyl; or

R′ and R″, together with the nitrogen atom to which they are attached, form a heterocyclic group selected from the group consisting of 1-azetidinyl, 1-pyrrolidinyl, 1-piperidinyl, 1-piperazinyl and 4-morpholinyl, wherein 1-azetidinyl, 1-pyrrolidinyl, 1-piperidinyl, 1-piperazinyl and 4-morpholinyl are optionally substituted with a substituent selected from the group consisting of halo, hydroxy, —C 1-4 alkyl, monohaloC 1-4 alkyl, polyhaloC 1-4 alkyl and —C(O)—C 1-4 alkyl;

comprising:

a) reacting a compound of Formula (V):

wherein:

R 2a is halo;

R 3 is selected from the group consisting of hydrogen and —C 1-4 alkyl; and

R 4 is selected from the group consisting of hydrogen, —C 1-4 alkyl, monohaloC 1-4 alkyl, polyhaloC 1-4 alkyl, —C 1-4 alkyl-O—C 1-4 alkyl and —C 1-4 alkyl-OH;

with a compound of Formula (IIIa):

wherein:

R 5 and R 6 are each independently selected from the group consisting of hydrogen, halo, cyano, —C 1-4 alkyl, —C 1-4 alkyl-OH, —C 3-7 cycloalkyl, monohaloC 1-4 alkyl, polyhaloC 1-4 alkyl, —C 1-4 alkyl-O—C 1-4 alkyl, —O—C 1-4 alkyl, monohaloC 1-4 alkyloxy, polyhaloC 1-4 alkyloxy, 1-acetylazetidin-3-yl and —NR′R″;

R′ and R″ are each independently selected from the group consisting of hydrogen and —C 1-4 alkyl; or

R′ and R″, together with the nitrogen atom to which they are attached, form a heterocyclic group selected from the group consisting of 1-azetidinyl, 1-pyrrolidinyl, 1-piperidinyl, 1-piperazinyl and 4-morpholinyl, wherein 1-azetidinyl, 1-pyrrolidinyl, 1-piperidinyl, 1-piperazinyl and 4-morpholinyl are optionally substituted with a substituent selected from the group consisting of halo, hydroxy, —C 1-4 alkyl, monohaloC 1-4 alkyl, polyhaloC 1-4 alkyl and —C(O)—C 1-4 alkyl; R 3 is selected from the group consisting of hydrogen and —C 1-4 alkyl;

R 7a and R 8a are each independently selected from the group consisting of hydrogen and —C 1-4 alkyl; or

R 7a and R 8a are taken together to form a bivalent radical of the formula —(CH 2 ) 2 —, —(CH 2 ) 3 — or —C(CH 3 ) 2 —C(CH 3 ) 2 —;

in the presence of a palladium catalyst, a base and solvent, to form a compound of Formula (II):

wherein:

R 2 is

R 3 is selected from the group consisting of hydrogen and —C 1-4 alkyl;

R 4 is selected from the group consisting of hydrogen, —C 1-4 alkyl, monohaloC 1-4 alkyl, polyhaloC 1-4 alkyl, —C 1-4 alkyl-O—C 1-4 alkyl and —C 1-4 alkyl-OH;

R 5 and R 6 are each independently selected from the group consisting of hydrogen, halo, cyano, —C 1-4 alkyl, —C 1-4 alkyl-OH, —C 3-7 cycloalkyl, monohaloC 1-4 alkyl, polyhaloC 1-4 alkyl, —C 1-4 alkyl-O—C 1-4 alkyl, —O—C 1-4 alkyl, monohaloC 1-4 alkyloxy, polyhaloC 1-4 alkyloxy, 1-acetylazetidin-3-yl and —NR′R″; and

R′ and R″ are each independently selected from the group consisting of hydrogen and C 1-4 alkyl; or

R′ and R″, together with the nitrogen atom to which they are attached, form a heterocyclic group selected from the group consisting of 1-azetidinyl, 1-pyrrolidinyl, 1-piperidinyl, 1-piperazinyl and 4-morpholinyl, wherein 1-azetidinyl, 1-pyrrolidinyl, 1-piperidinyl, 1-piperazinyl and 4-morpholinyl are optionally substituted with a substituent selected from the group consisting of halo, hydroxy, C 1-4 alkyl, monohaloC 1-4 alkyl, polyhaloC 1-4 alkyl and —C(O)—C 1-4 alkyl; and

b) reacting the compound of Formula (II) above with a compound of Formula (III):

R 1 is phenyl or 2-pyridinyl, each optionally substituted with one or more substituents independently selected from the group consisting of halo, —C 1-4 alkyl, monohaloC 1-4 alkyl, polyhaloC 1-4 alkyl, —O—C 1-4 alkyl, —C 1-4 alkyl-O—C 1-4 alkyl, monohaloC 1-4 alkyloxy, polyhaloC 1-4 alkyloxy, —C 1-4 alkyl-OH, —C 1-4 alkylthio, monohaloC 1-4 alkylthio, polyhaloC 1-4 alkylthio, cyano, —C 3-7 cycloalkyl optionally substituted with trifluoromethyl and —SF 5 ; or

R 1 is

 and

X is halo;

in the presence of (i) 2-(7-aza-1H-benzotriazol-1-yl)-1,1,3,3-tetramethyluronium hexafluorophosphate, or (ii) a catalyst, a ligand, a base and solvent, to provide the compound of Formula (I) above.

21. A process for preparing a pharmaceutical composition comprising admixing a pharmaceutically acceptable carrier or excipient with a therapeutically effective amount of a compound of claim 1 .

22. A compound of Formula (II):

wherein:

R 2 is

R 3 is selected from the group consisting of hydrogen and —C 1-4 alkyl;

R 4 is selected from the group consisting of hydrogen, —C 1-4 alkyl, monohaloC 1-4 alkyl, polyhaloC 1-4 alkyl, —C 1-4 alkyl-O—C 1-4 alkyl and —C 1-4 alkyl-OH;

R 5 and R 6 are each independently selected from the group consisting of hydrogen, halo, cyano, —C 1-4 alkyl, —C 1-4 alkyl-OH, —C 3-7 cycloalkyl, monohaloC 1-4 alkyl, polyhaloC 1-4 alkyl, —C 1-4 alkyl-O—C 1-4 alkyl, —O—C 1-4 alkyl, monohaloC 1-4 alkyloxy, polyhaloC 1-4 alkyloxy, 1-acetylazetidin-3-yl and —NR′R″; and

R′ and R″ are each independently selected from the group consisting of hydrogen and C 1-4 alkyl; or

R′ and R″, together with the nitrogen atom to which they are attached, form a heterocyclic group selected from the group consisting of 1-azetidinyl, 1-pyrrolidinyl, 1-piperidinyl, 1-piperazinyl and 4-morpholinyl, wherein 1-azetidinyl, 1-pyrrolidinyl, 1-piperidinyl, 1-piperazinyl and 4-morpholinyl are optionally substituted with a substituent selected from the group consisting of halo, hydroxy, C 1-4 alkyl, monohaloC 1-4 alkyl, polyhaloC 1-4 alkyl and —C(O)—C 1-4 alkyl.

23. A compound of Formula (V):

wherein:

R 2a is halo;

R 3 is selected from the group consisting of hydrogen and —C 1-4 alkyl; and

R 4 is selected from the group consisting of hydrogen, —C 1-4 alkyl, monohaloC 1-4 alkyl, polyhaloC 1-4 alkyl, —C 1-4 alkyl-O—C 1-4 alkyl and —C 1-4 alkyl-OH.

24. The compound of claim 23 , wherein the compound has the Formula (V′):
