Abstract

Platelets or blood cells are detected in a fluid sample by adjusting a focal depth of a microscope through a range of values, the microscope having a mounted sample and an objective lens adapted with one or both of (a) a spherical aberration correction unmatched to a utilized cover plate for the sample, or (2) a numerical aperture unmatched to a utilized illumination source for the sample. Images are recorded at different specific focal depths and in multiple z planes of a fluid bearing the platelets, where the position of platelets may overlap on different of the multiple z planes that are recorded, the images recorded through the cover plate, thus causing the generation of a specific light-dark pattern indicative of platelets at particular positions and at multiple depths in the fluid media. The images are analyzed for the specific light-dark pattern.

Claims (16)

1. An optical platelet counting method, comprising:

adjusting a focal depth of a microscope through a range of values, the microscope having an objective lens adapted with one or both of (a) a spherical aberration correction unmatched to a utilized cover plate for a sample of platelet-bearing fluid contained by a slide having a depth, or (b) a numerical aperture unmatched to a utilized illumination source for imaging the sample; recording out of focus images caused by one or both of the unmatched spherical aberration correction and the unmatched numerical aperture at different specific focal depths and in multiple z planes of the platelet-bearing fluid, where the inter-plane coordinates of platelets may overlap on different ones of the multiple z planes that are recorded, the images recorded through the cover plate, thus causing the generation of a specific light-dark pattern across a sequence of a plurality of the out of focus images indicative of and distinguishing platelets at particular inter-plane coordinates and at multiple depths in the fluid media; and analyzing the sequence of the out of focus images and identifying the presence of a platelet upon detecting the specific light-dark pattern.

2. The method of claim 1 , further comprising: a z range for the z planes of approximately 320 micrometers and a slide depth of approximately 100 micrometers.

3. The method of claim 1 , further comprising: recording forty images at different depths of the fluid at three images per second.

4. The method of claim 1 , further comprising: the illumination source is a blue LED source having a wavelength of approximately 470 nm placed at a distance of approximately 140 mm under the sample.

5. The method of claim 1 , further comprising: the light dark pattern comprises, in forward or reverse order (c) a dark ring around a bright center, (d) a dark circular shape, and (e) a white center with dark ring.

6. An optical blood cell counting method, comprising:

adjusting a focal depth of a microscope through a range of values, the microscope having an objective lens adapted with one or both of (a) a spherical aberration correction unmatched to a utilized cover plate for a sample of blood-cell-bearing fluid contained by a slide having a depth, or (b) a numerical aperture unmatched to a utilized illumination source for imaging the sample; recording out of focus images caused by one or both of the unmatched spherical aberration correction and the unmatched numerical aperture at different specific focal depths and in multiple z planes of the blood-cell-bearing fluid, where the inter-plane coordinates of blood cells may overlap on different ones of the multiple z planes that are recorded, the images recorded through the cover plate, thus causing the generation of a specific light-dark pattern across a sequence of a plurality of the out of focus images indicative of and distinguishing blood cells at particular inter-plane coordinates and at multiple depths in the fluid media; and analyzing the sequence of the out of focus images and identifying the presence of a blood cell upon detecting the specific light-dark pattern.

7. The method of claim 6 , further comprising: a z range for the z planes of approximately 320 micrometers and a slide depth of approximately 100 micrometers.

8. The method of claim 6 , further comprising: recording forty images at different depths of the fluid at three images per second.

9. The method of claim 6 , further comprising: the illumination source is a blue LED source having a wavelength of approximately 470 nm placed at a distance of approximately 140 mm under the sample.

10. The method of claim 6 , further comprising: the light dark pattern comprises, in forward or reverse order (c) a black ring surrounding a white core, and (d) the white core becomes smaller and completely black with a white ring surrounding it.

11. The method of claim 6 , further comprising: the light dark pattern comprises, in forward or reverse order (c) a black ring surrounding a greyish core, and (d) the white core becomes smaller and completely black with a white ring surrounding it.

12. The method of claim 6 , further comprising: the light dark pattern comprises, in forward or reverse order (c) a completely grey circle (d) a black circle, and (e) a white core surrounded by a grey circle.

13. The method of claim 6 , further comprising:

the light dark pattern comprises, in forward or reverse order (c) a grey center grey and an outside ring of a darker grey, (d) a smaller white center surrounded by a dark grey or black ring, (e) a dark grey center surrounded by a large grey outside ring.
