Abstract

A method for enhancing infectivity of HCMV virus particles is provided.

Claims (12)

1. An in vitro method for production of virus particles in a virus-infected host cell comprising the step of contacting the cell with a compound that decreases activity of LRP1, wherein the virus particles produced in the cell in the presence of the compound display increased infectivity compared to virus particles produced in the cell in the absence of the compound, wherein the virus is an enveloped virus, and wherein the compound binds LRP1 and blocks LRP1 binding activity or decreases transcription and/or translation of a polynucleotide encoding LRP1, wherein the virus is a cytomegalovirus (CMV).

An in vitro method for production of virus particles in a virus-infected host cell comprising the step of contacting the cell with a compound that increases intracellular cholesterol, wherein the virus particles produced in the cell in the presence of the compound display increased infectivity compared to virus particles produced in the cell in the absence of the compound, wherein the virus is an enveloped virus, and wherein the compound binds LRP1 and blocks LRP1 binding activity or decreases transcription and/or translation of a polynucleotide encoding LRP1, wherein the virus is a cytomegalovirus (CMV).

3. The method of claim 2 wherein the compound:

decreases LRP1 activity;

decreases LRP1 binding activity;

binds LRP1 and blocks LRP1 binding activity;

is a blocking antibody;

is an extracellular fragment of LRP1;

decreases transcription and/or translation of a polynucleotide encoding LRP1; or

decreases surface expression of LRP1.

4. The method of claim 1 wherein the compound is selected from the group consisting of an anti-LRP1 antibody and a compound that decreases transcription and/or translation of a polynucleotide encoding LRP1.

5. The method of claim 2 wherein the compound is selected from the group consisting of an anti-LRP1 antibody and a compound that decreases transcription and/or translation of a polynucleotide encoding LRP1.
