Abstract

The methods and assays described herein relate to detection, diagnosis, and treatment of lung cancer, e.g., by detecting the level of expression of certain miRNAs described herein and/or by therapeutically increasing the level of those miRNAs.

Claims (33)

1. A method of treating lung cancer in a subject in need thereof, the method comprising

administering a treatment for lung cancer to a subject determined to have a level of expression of at least 1 miRNAs selected from the group consisting of:

miR-146a-5p, miR-324-5p, miR-223-3p, miR-223-5p; miR-450b-5p, miR-221-3p-miR-505-3p, and miR-582-5p;

in a bronchial brushing or nose epithelial sample obtained from the subject which is decreased relative to a reference level,

wherein the treatment is resection, lobesctomy, pneumonectomy, radiation therapy, stereotactic body radiotherapy, chemotherapy, or immunotherapy.

2. The method of claim 1 , further comprising the first steps of obtaining a sample from a subject; and determining the level of expression of the at least 1 miRNA; and

administering a treatment for lung cancer if the level of expression of the at least 1 miRNA is decreased relative to a reference level.

3. The method of claim 1 , wherein the level of expression is detected for at least two miRNAs selected from the group consisting of:

miR-146a-5p, miR-324-5p, miR-223-3p, miR-223-5p; miR-450b-5p, miR-221-3p-miR-505-3p, and miR-582-5p.

4. The method of claim 1 , wherein the level of expression is detected for at least three miRNAs selected from the group consisting of:

miR-146a-5p, miR-324-5p, miR-223-3p, miR-223-5p; miR-450b-5p, miR-221-3p-miR-505-3p, and miR-582-5p.

5. The method of claim 1 , wherein the level of expression is detected for at least four miRNAs selected from the group consisting of:

miR-146a-5p, miR-324-5p, miR-223-3p, miR-223-5p; miR-450b-5p, miR-221-3p-miR-505-3p, and miR-582-5p.

6. The method of claim 1 , wherein the level of expression is detected for at least five miRNAs selected from the group consisting of:

miR-146a-5p, miR-324-5p, miR-223-3p, miR-223-5p; miR-450b-5p, miR-221-3p-miR-505-3p, and miR-582-5p.

7. The method of claim 1 , wherein the level of expression is detected for at least six miRNAs selected from the group consisting of:

miR-146a-5p, miR-324-5p, miR-223-3p, miR-223-5p; miR-450b-5p, miR-221-3p-miR-505-3p, and miR-582-5p.

8. The method of claim 1 , wherein the level of expression is detected for at least seven miRNAs selected from the group consisting of:

miR-146a-5p, miR-324-5p, miR-223-3p, miR-223-5p; miR-450b-5p, miR-221-3p-miR-505-3p, and miR-582-5p.

9. The method of claim 1 , wherein the level of expression is detected for miR-146a-5p, miR-324-5p, miR-223-3p, miR-223-5p; miR-450b-5p, miR-221-3p-miR-505-3p, and miR-582-5p.

10. The method of claim 1 , wherein the level of expression is detected for at least miR-146a-5p, miR-324-5p, miR-223-3p, and miR-223-5p.

11. The method of claim 1 , wherein the level of expression is detected for at least miR-146a-5p.

12. The method of claim 1 , wherein the method further comprises detecting the expression level of one or more mRNAs.

13. The method of claim 1 , wherein the expression level of no more than 100 miRNAs and/or mRNAs is detected.

14. A method of treating lung cancer in a subject in need thereof, the method comprising

administering a treatment for lung cancer to a subject determined to have a level of expression of at least 1 miRNAs selected from the group consisting of:

miR-146a-5p, miR-324-5p, miR-223-3p, miR-223-5p; miR-450b-5p, miR-221-3p-miR-505-3p, and miR-582-5p;

in a bronchial brushing or nose epithelial sample obtained from the subject which is decreased relative to a reference level, wherein the treatment for lung cancer comprises an agonist of at least 1 miRNA selected from the group consisting of:

miR-146a-5p, miR-324-5p, miR-223-3p, miR-223-5p; miR-450b-5p, miR-221-3p-miR-505-3p, and miR-582-5p.

15. The method of claim 1 , wherein the treatment is administration of afatinib, bevacizumab, certinib, crizotinib, erlotinib, nivolumab, or ramucirumab.

16. The method of claim 1 , wherein the treatment is resection, lobesctomy, or pneumonectomy.

17. The method of claim 1 , wherein the treatment comprises a first step of performing resection, lobesctomy, or pneumonectomy and a second step of administering radiation therapy, stereotactic body radiotherapy, chemotherapy, or immunotherapy.

18. The method of claim 1 , wherein the lung cancer is non-small cell lung cancer.
