Abstract

Disclosed is a lateral flow capillary device and uses thereof comprising a unipath bibulous capillary flow matrix and at least two reservoirs each in fluid communication with the capillary flow matrix wherein a reservoir contacts the capillary flow matrix through a passage having a rim pressing the matrix. The pressure that the rim applies on the matrix prevents leakage of liquids out of the capillary flow matrix at the reservoir/matrix interface, allowing accurate sequential draining of liquid from the reservoirs. During use of the disclosed lateral flow capillary device a static interface is formed between the first liquid and the second liquid in an interface creation zone inside the capillary flow matrix wherein the first amount and second amount are such that first liquid substantially remains in the first reservoir and the second liquid substantially remains in the second reservoir subsequent to the formation of the static interface and wherein the interface begins to move only subsequent to exhaustion of a liquid from a reservoir.

Claims (30)

1. A method of sequentially transferring liquids through a capillary flow matrix for analyte detection or analysis, the method comprising:

a) providing a lateral flow capillary device comprising a capillary flow matrix having an upstream end and a downstream end defining a lateral flow direction, and at least two reservoirs positioned between the upstream end and the downstream end and spaced from each other in the lateral flow direction, each reservoir being in fluid communication with a respective liquid receiving zone of said capillary flow matrix and each reservoir comprising a hollow conduit terminating in a rim configured to press against the capillary flow matrix while the matrix is dry;

b) pressing the rim of the hollow conduit of each of the reservoirs against the capillary flow matrix;

c) adding a first liquid to a first reservoir of said at least two reservoirs so that said first liquid flows into said capillary flow matrix through said respective liquid receiving zone of the capillary flow matrix associated with the first reservoir; and

d) adding a second liquid to a second reservoir of said at least two reservoirs so that said second liquid flows into said capillary flow matrix through said respective liquid receiving zone associated with the second reservoir; wherein the pressing of the rim of the hollow conduit of each of the reservoirs against the capillary flow matrix is sufficient to constrain flow out of the liquid receiving zones in said lateral flow direction such that when liquid is simultaneously present in said reservoirs, a downstream reservoir of the at least two reservoirs is substantially emptied before liquid from an upstream reservoir of the at least two reservoirs passes the downstream reservoir.

2. The method of claim 1 , wherein said liquid receiving zones are adjacent and form an interface creation zone.

3. The method of claim 1 , wherein said adding of said first liquid and of said second liquid is simultaneous.

4. The method of claim 1 , wherein said adding of said first liquid and of said second liquid is sequential.

5. The method of claim 1 , wherein said first liquid and said second liquid are identical.

6. The method of claim 1 , wherein said first liquid and said second liquid are different.

7. The method of claim 1 , wherein said first liquid and said second liquid are added in equal amounts.

8. The method of claim 1 , wherein said first liquid and said second liquid are added in different amounts.

9. The method of claim 1 , wherein said adding of said second liquid is subsequent to said adding of said first liquid.

10. A method of performing an analyte detection or testing reaction using a capillary flow matrix, the method comprising:

a) providing a lateral flow capillary device comprising a capillary flow matrix having a first liquid receiving zone in fluid communication with a first reservoir, and a second liquid receiving zone, upstream of said first liquid receiving zone, in fluid communication with a second reservoir, and a reagent disposed at a location between said first liquid receiving zone and said second liquid receiving zone, wherein each of the first and second reservoirs comprises a hollow conduit terminating in a rim configured to press against the capillary flow matrix while the matrix is dry;

b) pressing the capillary flow matrix with the rim of the hollow conduit of each of the first reservoir and the second reservoir;

c) adding a first liquid to said first reservoir so that said first liquid flows into said capillary flow matrix through said first liquid receiving zone to contact said reagent;

d) adding a second liquid to said second reservoir so that said second liquid flows into said capillary flow matrix through said second liquid receiving zone to contact said reagent wherein the pressing of the rim of the hollow conduit of each of the first and second reservoirs causes formation of a static liquid-liquid interface between the liquid receiving zones;

wherein said first liquid and said second liquid remain in said first reservoir and in said second reservoir, respectively, subsequent to formation of said static liquid-liquid interface; and

wherein said interface begins to move only subsequent to exhaustion of said first liquid from said first reservoir.

11. The method of claim 10 , wherein said first liquid and said second liquid are identical.

12. The method of claim 10 , wherein said first liquid and said second liquid are different.

13. The method of claim 10 , wherein said first liquid and said second liquid are added in equal amounts.

14. The method of claim 10 , wherein said first liquid and said second liquid are added in different amounts.

15. The method of claim 10 , wherein said capillary flow matrix further comprises downstream of said first liquid receiving zone on said capillary flow matrix, a reaction zone comprising at least one capturing entity configured to capture a material flowing through said capillary flow matrix.

16. The method of claim 10 , wherein said adding of said second liquid and said adding of said first liquid is sequential.

17. The method of claim 10 , wherein said adding of said first liquid and of said second liquid is simultaneous.

18. The method of claim 10 , wherein the static liquid-liquid interface is formed between the first liquid and the second liquid.

19. The method of claim 1 , wherein pressing of the rim of the hollow conduit of each reservoir against the capillary flow matrix causes a decrease in a ratio of capillary conduits to solid material in a cross-section of the capillary flow matrix compared to a ratio of capillary conduits to solid material in a cross-section of the capillary flow matrix at a location not pressed by the rim of a hollow conduit of one of the reservoirs.

The method of claim 10 , wherein pressing of the rim of the hollow conduit of each of the first and second reservoirs against the capillary flow matrix causes a decrease in a ratio of capillary conduits to solid material in a cross-section of the capillary flow matrix compared to a ratio of capillary conduits to solid material in a cross-section of the capillary flow matrix at a location not pressed by the rim of a hollow conduit of one of the first or second reservoirs.
