Abstract

This disclosure relates to the isolation and sequencing of nucleic acid molecules that encode cytochrome P450 polypeptides from a Papaver somniferum cultivar; uses in the production of noscapine and identification of poppy cultivars that include genes that comprise said nucleic acid molecules.

Claims (39)

1. A vector comprising a promoter adapted for expression in a microbial host cell operably linked to a nucleic acid molecule that encodes a cytochrome P450 polypeptide, wherein said nucleic acid molecule comprises or consists of a nucleotide sequence selected from the group consisting of:

i) the nucleotide sequence of SEQ ID NO:57;

ii) a nucleotide sequence that is degenerate as a result of the genetic code to the nucleotide sequence defined in (i);

iii) a nucleic acid sequence comprising at least 90% sequence identity with the nucleotide sequence of SEQ ID NO: 57;

iv) a nucleotide sequence that encodes the protein sequence of SEQ ID NO: 9; and

v) a nucleotide sequence that encodes a polypeptide comprising at least 95% sequence identity to the protein sequence of SEQ ID NO: 9.

2. The vector according to claim 1 , wherein said nucleic acid molecule comprises or consists of the nucleotide sequence of SEQ ID NO: 57.

3. The vector according to claim 1 , wherein said nucleic acid sequence comprising a promoter confers constitutive expression on said cytochrome P450 polypeptide.

4. A transgenic cell transformed or transfected with the vector of claim 1 .

5. The transgenic cell according to claim 4 , wherein said cell is a microbial cell.

A vector comprising a nucleic acid molecule comprising a transcription cassette, wherein said cassette comprises the nucleotide sequence of SEQ ID NO: 57 or 6 and is adapted for expression by provision of at least one promoter operably linked to said nucleotide sequence such that both sense and antisense molecules are transcribed from said cassette.

7. The vector according to claim 6 , adapted for expression in a plant cell.

8. A transgenic plant cell transfected with the vector of claim 6 , wherein said cell has reduced expression of SEQ ID NO: 57 or 6.

9. The vector of claim 1 , wherein the vector is a viral vector.

A vector comprising a promoter adapted for expression in a microbial host cell operably linked to a nucleic acid molecule encoding a cytochrome P450 polypeptide, wherein the nucleic acid molecule encoding the cytochrome P450 polypeptide comprises at least 95% sequence identity to the nucleotide sequence of SEQ ID NO: 57.

11. The vector of claim 10 , wherein the promoter is a bacterial or fungal promoter.

12. The vector of claim 10 , wherein the nucleic acid molecule encoding the cytochrome P450 polypeptide comprises at least 99% sequence identity to the nucleotide sequence of SEQ ID NO: 57.

13. The vector of claim 10 , wherein the nucleic acid molecule encoding the cytochrome P450 polypeptide comprises the nucleotide sequence of SEQ ID NO: 57.

14. The vector of claim 10 , further comprising a selectable marker.

15. A transgenic cell comprising the vector of claim 10 .

16. The transgenic cell of claim 15 , wherein the cell is a microbial cell.

17. The transgenic cell of claim 16 , wherein the microbial cell is a bacterial cell or fungal cell.

18. A transgenic microbial cell comprising the vector of claim 1 , wherein the nucleic acid molecule encoding the polypeptide comprises at least 95% sequence identity to the nucleotide sequence of SEQ ID NO: 57.

19. The transgenic microbial cell of claim 18 , wherein the microbial cell is a bacterial cell or fungal cell.

20. A vector comprising

a nucleic acid molecule encoding a cytochrome P450 polypeptide, wherein the nucleic acid molecule encoding the polypeptide comprises at least 95% sequence identity to the nucleotide sequence of SEQ ID NO: 57, and

a microbial promoter operably linked to the nucleic acid molecule encoding the cytochrome P450 polypeptide.

21. The vector of claim 20 , wherein the nucleic acid molecule encoding the cytochrome P450 polypeptide comprises at least 99% sequence identity to the nucleotide sequence of SEQ ID NO: 57.

22. The vector of claim 20 , wherein the isolated nucleic acid molecule encoding the cytochrome P450 polypeptide comprises the nucleotide sequence of SEQ ID NO: 57.

23. The vector of claim 20 , wherein the isolated nucleic acid molecule encoding the cytochrome P450 polypeptide consists of the nucleotide sequence of SEQ ID NO: 57.

24. The vector of claim 20 , wherein the microbial promoter is a bacterial or fungal promoter.

25. A method of modifying an alkaloid, the method comprising:

culturing the transgenic cell of claim 15 with the alkaloid tetrahydrocolumbamine in a cell culture under conditions that modify tetrahydrocolumbamine, thereby producing canadine; and

optionally isolating canadine from the cell culture.

26. The method of claim 25 , wherein the transgenic cell is a bacterial cell or fungal cell.

27. A method of modifying an alkaloid, the method comprising:

culturing the transgenic microbial cell of claim 18 with the alkaloid tetrahydrocolumbamine in a cell culture under conditions that modify tetrahydrocolumbamine thereby producing canadine; and

optionally isolating canadine from the cell culture.

28. The method of claim 27 , wherein the transgenic microbial cell is a bacterial cell or fungal cell.
