RNA complexes that inhibit melanin production
In certain aspects, provided herein are RNA complexes (e.g., asymmetric RNA complexes, such as asiRNAs and lasiRNAs) that inhibit tyrosinase expression and are therefore useful for reducing melanin production and for treating pigmentation-related disorders associated with excessive melanin production, such as melasma and age spots.
1. A method of inhibiting tyrosinase expression in a subject, comprising: administering topically, in the absence of a transfection reagent, to the skin of the subject a cell-penetrating asymmetric RNA (asiRNA) comprising an antisense strand of 19 to 21 nucleotides (nt) in length having sequence complementarity to a tyrosinase mRNA sequence and a sense strand of 15 to 17 nt in length having sequence complementarity to the antisense strand, wherein the antisense strand and the sense strand form a complex in which the 5′ end of the antisense strand and the 3′ end of the sense strand form a blunt end, and wherein the sense strand has a cholesterol moiety attached to the 3′ end thereof.
2. The method of claim 1 , wherein the asiRNA comprises at least one phosphorothioate bond.
3. The method of claim 2 , wherein the antisense strand comprises four phosphorothioate bonds.
4. The method of claim 3 , wherein the four phosphorothioate bonds are formed at the 3′ end of the antisense strand.
5. The method of claim 1 , wherein the sense strand has a sequence of SEQ ID NO: 7 and the antisense strand has a sequence of SEQ ID NO: 8.
6. The method of claim 5 , wherein the sense strand has a sequence of SEQ ID NO: 15 and the antisense strand has a sequence of SEQ ID NO: 16.
7. The method of claim 1 , wherein the sense strand has a sequence of SEQ ID NO: 17 and the antisense strand has a sequence of SEQ ID NO: 18.
8. The method of claim 1 , wherein the sense strand has a sequence of SEQ ID NO: 19 and the antisense strand has a sequence of SEQ ID NO: 20.
9. The method of claim 1 , wherein the sense strand has a sequence of SEQ ID NO: 31 and the antisense strand has a sequence of SEQ ID NO: 32.
10. The method of claim 1 , wherein the sense strand has a sequence of SEQ ID NO: 87 and the antisense strand has a sequence of SEQ ID NO: 88.
11. The method of claim 1 , wherein the sense strand has a sequence of SEQ ID NO: 89 and the antisense strand has a sequence of SEQ ID NO: 90.
12. The method of claim 1 , wherein the sense strand has a sequence of SEQ ID NO: 119 and the antisense strand has a sequence of SEQ ID NO: 120.
13. The method of claim 1 , wherein the sense strand has a sequence of SEQ ID NO: 125 and the antisense strand has a sequence of SEQ ID NO: 126.
14. The method of claim 1 , wherein the sense strand has a sequence of SEQ ID NO: 131 and the antisense strand has a sequence of SEQ ID NO: 132.
15. The method of claim 1 , wherein the subject has a skin pigmentation disorder associated with excessive melanin production, and wherein the asiRNA is administered in an effective amount to treat the skin pigmentation disorder in the subject.
16. The method of claim 15 , wherein the skin pigmentation disorder comprises melasma or age spots.
17. The method of claim 15 , further comprising administering to the subject a skin lightening agent.
18. The method of claim 17 , wherein the skin lightening agent comprises hydroquinone, arbutin, tretinoin, kojic acid, azelaic acid, or tranexamic acid.
19. The method of claim 1 , wherein the asiRNA is formulated as part of a cream or a lotion.
