Abstract

Method for the detection of glutamine and its analogues are provided by the present disclosure. Also provided are methods for measuring the levels of glutamine and its analogues, including diagnostic methods. Further provided are methods that utilise glutamine analogues for the synthesis of coloured pigments and other useful agents. The methods comprise the use of a nonribosomal peptide synthetase (NRPS) under conditions to produce an indigoidine or indigoidine-related pigment.

Claims (23)

1. A method of measuring the concentration of L-glutamine or an L-glutamine analogue in a sample obtained from a subject or a sample obtained from a bacterial or eukaryotic cell culture, the method comprising:

incubating the sample with an activated pigment-producing NRPS (non-ribosomal peptide synthetase) under conditions to produce indigoidine pigment or an indigoidine analogue pigment; and

quantifying the amount of the indigoidine pigment or the indigoidine analogue pigment produced in the sample by colorimetric analysis of the sample, thereby measuring the concentration of the L-glutamine or the L-glutamine analogue, respectively, in the sample, wherein at least one water-miscible solvent is added to the sample in an amount effective to dissolve the indigoidine pigment or indigoidine analogue pigment prior to quantification.

2. The method of claim 1 , wherein the at least one water-miscible solvent comprises dimethylsulfoxide.

3. The method of claim 1 , wherein the at least one water-miscible solvent comprises dimethylformamide.

4. The method of claim 1 , wherein the pigment-producing NRPS is BpsA (blue pigment synthetase A), and wherein the BpsA is activated to the holo form prior to isolation of the BpsA.

5. The method of claim 1 , wherein the pigment-producing NRPS is BpsA, and wherein the BpsA is activated to the holo form following isolation of the BpsA.

6. The method of claim 1 , wherein the sample is selected from the group consisting of a blood sample, a urine sample, a saliva sample, a cerebrospinal fluid sample, a lymph fluid sample, a eukaryotic cell culture sample, a bacterial cell culture sample, and a fermentation sample.

7. The method of claim 6 , wherein the sample comprises an organic co-solvent selected from the group consisting of an alcohol, a polyalcohol, dimethylformamide, and an acetonitrile.

8. The method of claim 1 , wherein the glutamine analogue is selected from the group consisting of glycyl-glutamine, N-trifluoroacetyl-L-glutamine, alanyl-glutamine, indoleacetyl glutamine, N-acetylglutamine, phenylacetyl-L-glutamine, and glutamine-terminating peptides or polypeptides.

The method of claim 1 , wherein the sample comprises a bacterial cell that synthesises L-glutamine or the glutamine analogue, and wherein an alteration in the amount of the indigoidine pigment or the indigoidine analogue pigment relative to a control or standard amount indicates altered levels of the L-glutamine or the glutamine analogue production, respectively.

10. The method of claim 9 , wherein the at least one water-miscible solvent comprises dimethylsulfoxide.

11. The method of claim 9 , wherein the at least one water-miscible solvent comprises dimethylformamide.

12. The method of claim 9 wherein the indigoidine pigment or indigoidine analogue pigment is dissolved in the at least one water-miscible solvent prior to quantification.

13. The method of claim 9 , wherein the pigment-producing NRPS is BpsA, and wherein the BpsA is activated to the holo form prior to isolation of the BpsA.

14. The method of claim 9 , wherein the pigment-producing NRPS is BpsA, and the BpsA is activated to the holo form following isolation of the BpsA.

15. The method of claim 9 , wherein the bacterial cell is selected from the group consisting of a mutant cell, a variant cell, and a cell produced by targeted or random mutagenesis.

16. The method of claim 9 , wherein the glutamine analogue is selected from the group consisting of a glutamine-terminating peptide, a glutamine-terminating polypeptide, and a glutamine-terminating dipeptide.

17. The method of claim 16 , wherein the glutamine analogue is alanyl-glutamine.

18. The method of claim 1 , wherein colorimetric analysis of the sample is carried out at 590 nm.

19. A method of measuring the concentration of L-glutamine or an L-glutamine analogue in a sample, the method comprising:

incubating a sample obtained from a subject or a sample obtained from a bacterial or eukaryotic cell culture with an activated pigment-producing NRPS (non-ribosomal peptide synthetase) under conditions to produce indigoidine pigment or an indigoidine analogue pigment; and

quantifying the amount of the indigoidine pigment or the indigoidine analogue pigment produced in the sample by colorimetric analysis of the sample, thereby measuring the concentration of the L-glutamine or the L-glutamine analogue, respectively, in the sample, wherein at least one water-miscible solvent is added to the sample in an amount effective to dissolve the indigoidine pigment or indigoidine analogue pigment prior to quantification.
