Abstract

Provided herein are compositions and methods for the treatment of cancer by inhibition of β-catenin or a β-catenin pathway. In particular, inhibitors of β-catenin and/or the Wnt/p-catenin signaling pathway are employed prevent or reverse evasion of immune response or immunotherapy by cancers.

Claims (25)

1. A method for the treatment of solid tumor cancer in a subject comprising;

(a) testing tumor cells or tissue from the subject for one or more of:

(i) exclusion of T cell infiltration,

(ii) transcriptional repression of chemokine CCL4,

(iii) defective recruitment of CD103 + dermal dendritic cells; and

(b) administering a β-catenin inhibitor and/or a Wnt/β-catenin pathway inhibitor to the subject.

2. The method of claim 1 , wherein the subject suffers from melanoma.

3. The method of claim 1 , wherein the subject has one or more tumors exhibiting tumor-intrinsic-β-catenin-signaling.

4. The method of claim 3 , further comprising a step of testing the subject, a tumor, or a tumor cell for β-catenin signaling.

5. The method of claim 1 , wherein the β-catenin inhibitor and/or β-catenin pathway inhibitor is selected from a group consisting of a small molecule, a peptide, a polypeptide, a nucleic acid, an antibody, and an antibody fragment.

6. The method of claim 1 , further comprising co-administration of an additional therapeutic agent.

7. The method of claim 6 , wherein the additional therapeutic agent is a chemotherapeutic or an immunotherapeutic agent.

8. The method of claim 7 , wherein the additional therapeutic agent is an immunotherapeutic agent selected from the list consisting of cell-based therapies, monoclonal antibody (mAb) therapy, cytokine therapy, and adjuvant treatment.

9. The method of claim 8 , wherein the immunotherapeutic agent is a mAb therapy selected from the list consisting of anti-CTLA-4 monoclonal antibodies and/or anti-PD-L1 monoclonal antibodies.

10. The method of claim 8 , wherein the immunotherapeutic agent is a cell-based therapy selected from the list consisting of dendritic-cell therapy and T-cell therapy.

11. The method of claim 1 , further comprising:

testing tumor cells or tissue from the subject for one or more of:

(iv) tumor-intrinsic-β-catenin-signaling,

and

(v) activation of CD8 + T cells.

12. The method of claim 11 , further comprising surgical, radiation, and/or chemotherapeutic cancer intervention.

13. The method of claim 11 , further comprising:

retesting the subject for one or more of (i) through (v).

14. The method of claim 13 , wherein if the subject retests negative for one or more of (i) through (v) that were positive in an initial test, an immunotherapeutic agent is administered to the subject.

15. The method of claim 11 , further comprising testing the subject for one or more additional cancer biomarkers.
