Chimeric adeno-associated virus/ bocavirus parvovirus vector
The invention provides an isolated chimeric virus comprising bocavirus capsid protein and a recombinant adeno-associated viral (AAV) genome, an isolated rBoV comprising human bocavirus capsid protein and a recombinant BoV genome, and uses therefor. For example, the chimeric virus may be employed to deliver transgenes, such as those encoding therapeutic or prophylactic gene products, to mammalian cells.
1. A method to express a heterologous gene product in mammalian cells comprising providing isolated chimeric virus comprising human bocavirus (BoV) capsid protein and a recombinant adeno-associated viral (AAV) genome comprising an expression cassette encoding the heterologous gene product; and
infecting mammalian cells having the tropism of human bovavirus with the chimeric virus an amount effective to express the heterologous gene product.
2. The method of claim 1 wherein the gene product is a catalytic RNA, a microRNA, RNA pre-transplicing molecule (PTM-RNA), a neutralizing antibody or an antigen binding fragment thereof, a prophylactic gene product, or a polypeptide.
3. The method of claim 1 further comprising
contacting the mammalian cells with at least one agent in an amount effective to additively or synergistically enhance rAAV transduction relative to corresponding mammalian cells contacted with the chimeric virus but not the agent.
4. The method of claim 3 wherein the mammalian cells are contacted with the agent before the mammalian cells are contacted with the virus.
5. The method of claim 3 wherein the mammalian cells are cell or contacted with the virus before the mammalian cells are contacted with the agent.
6. The method of claim 1 wherein the gene product encodes a therapeutic protein.
7. The method of claim 1 wherein the rAAV genome is a rAAV-1, rAAV-2, rAAV-3, rAAV-4, rAAV-5, rAAV-6, rAAV-7, rAAV-8 or rAAV-9 genome.
8. The method of claim 1 wherein the gene product is a viral antigen, bacterial antigen, tumor antigen, parasite antigen, or fungal antigen.

The method of claim 1 wherein the gene product is cystic fibrosis transmembrane conductance regulator, b-globin, g-globin, tyrosine hydroxylase, glucocerebrosidase, aryl sulfatase A, factor VIII, dystrophin, alpha 1-antitrypsin, surfactant protein SP-D, SP-A or SP-C, erythropoietin, HBoV protein, influenza virus protein, respiratory syncytial virus (RSV) protein, a bacterial protein, a neutralizing antibody or an antigen binding fragment thereof, severe acute respiratory syndrome (SARS) virus protein, interferon (IFN)-alpha, gamma, tumor necrosis factor (TNF), interleukin (IL)-1, IL-17, or IL-6.
10. The method of claim 1 wherein the HBoV is HBoV1.
11. The of claim 1 wherein the BoV is HBoV2.
12. The method of claim 1 wherein the BoV is HBoV3.
13. The method of claim 1 wherein the BoV is HBoV4.
14. The method of claim 1 wherein the heterologous gene product comprises a cystic fibrosis transmembrane conductance regulator.
15. The method of claim 1 wherein the gene product is a therapeutic gene product.
