Abstract

The invention provides an isolated and purified nucleic acid sequence encoding a chimeric antigen receptor (CAR) directed against B-cell Maturation Antigen (BCMA). The invention also provides host cells, such as T-cells or natural killer (NK) cells, expressing the CAR and methods for destroying multiple myeloma cells.

Claims (32)

1. A chimeric antigen receptor comprising:

(a) an antigen binding domain targeting B-Cell Maturation Antigen (BCMA), wherein the antigen binding domain comprises an antibody or antigen binding fragment thereof;

(b) a transmembrane domain;

(c) a 4-1BB signaling domain; and

(d) a CD3ζ signaling domain.

2. The chimeric antigen receptor of claim 1 , wherein the antigen binding domain comprises a single chain variable fragment (scFv).

The chimeric antigen receptor of claim 2 , wherein the amino acid sequence of the scFv comprises the (i) heavy chain complementarity determining region (CDR)1, (ii) heavy chain CDR2, (iii) heavy chain CDR3, (iv) light chain CDR1, (v) light chain CDR2, and (vi) light chain CDR3 of one amino acid sequence selected from the group consisting of SEQ ID NO: 4, SEQ ID NO: 5, SEQ ID NO: 6, SEQ ID NO: 8, SEQ ID NO: 9, SEQ ID NO: 10, SEQ ID NO: 11, and SEQ ID NO: 12.

4. The chimeric antigen receptor of claim 1 , wherein the transmembrane domain is a human CD28 transmembrane domain.

5. The chimeric antigen receptor of claim 1 , wherein the transmembrane domain is a human CD8α transmembrane domain.

6. The chimeric antigen receptor of claim 1 , wherein the chimeric antigen receptor further comprises a hinge domain.

7. The chimeric antigen receptor of claim 6 , wherein the hinge domain is a human CD8α hinge domain.

8. The chimeric antigen receptor of claim 6 , wherein the hinge domain is a human CD28 hinge domain.

9. The chimeric antigen receptor of claim 6 , wherein the antigen binding domain comprises an scFv, the hinge domain is a human CD8α hinge domain, and the transmembrane domain is a human CD8α transmembrane domain.

10. The chimeric antigen receptor of claim 6 , wherein the antigen binding domain comprises an scFv, the hinge domain a human CD28 hinge domain, and the transmembrane domain is a human CD28 transmembrane domain.

11. The chimeric antigen receptor of claim 6 , wherein the antigen binding domain comprises an scFv, the hinge domain is a human CD8α hinge domain, and the transmembrane domain is a human CD28 transmembrane domain.

12. The chimeric antigen receptor of claim 6 , wherein the antigen binding domain comprises an scFv, the hinge domain is a human CD28 hinge domain, and the transmembrane domain is a human CD8α transmembrane domain.

13. The chimeric antigen receptor of claim 1 , wherein the chimeric antigen receptor further comprises a signal sequence.

14. The chimeric antigen receptor of claim 13 , wherein the signal sequence is a granulocyte-macrophage colony-stimulating factor (GM-CSF) receptor signal sequence or a CD8α signal sequence.

15. The chimeric antigen receptor of claim 13 , wherein the signal sequence is a CD8α signal sequence.

16. The chimeric antigen receptor of claim 1 , wherein the CAR comprises a CD8α signal sequence, an scFv antigen binding domain, a human CD8α hinge domain, a human CD8α transmembrane domain, a human 4-1BB signaling domain, and a human CD3 signaling domain.

17. The chimeric antigen receptor of claim 1 , wherein the CAR comprises a CD8α signal sequence, an scFv antigen binding domain, a human CD28 hinge domain, a human CD28 transmembrane domain, a human 4-1BB signaling domain, and a human CD3 signaling domain.

18. The chimeric antigen receptor of claim 1 , wherein the CAR comprises a CD8α signal sequence, an scFv antigen binding domain, a human CD8α hinge domain, a human CD28 transmembrane domain, a human 4-1BB signaling domain, and a human CD3 signaling domain.

19. The chimeric antigen receptor of claim 1 , wherein the CAR comprises a CD8α signal sequence, an scFv antigen binding domain, a human CD28 hinge domain, a human CD8α transmembrane domain, a human 4-1BB signaling domain, and a human CD3 signaling domain.

20. A nucleic acid sequence encoding the chimeric antigen receptor of claim 1 .

21. The nucleic acid sequence of claim 20 , wherein the nucleic acid sequence encodes an amino acid sequence that is more than 80% identical to the amino acid sequence of SEQ ID NO:10.

22. The nucleic acid sequence of claim 20 , wherein the nucleic acid sequence encodes an amino acid sequence that is more than 90% identical to the amino acid sequence of SEQ ID NO:10.

23. The nucleic acid sequence of claim 20 , wherein the nucleic acid sequence encodes an amino acid sequence that is more than 95% identical to the amino acid sequence of SEQ ID NO:10.

24. A vector comprising the nucleic acid sequence of claim 20 .

25. The vector of claim 24 , wherein the vector is a retroviral vector.

26. The vector of claim 24 , wherein the vector is a lentiviral vector.

27. The vector of claim 24 , wherein the vector comprises one or more of promoters, enhancers, polyadenylation signals, transcription terminators, or internal ribosome entry sites (IRES).

28. The vector of claim 24 , wherein the vector further comprises a selectable marker gene.
