1H-pyrrolo[2,3-c]pyridin-7(6H)-ones and pyrazolo[3,4-c]pyridin-7(6H)-ones as inhibitors of BET proteins
The present invention relates to substituted pyrrolopyridinones and substituted pyrazolopyridinones which are inhibitors of BET proteins such as BRD2, BRD3, BRD4, and BRD-t and are useful in the treatment of diseases such as cancer.
1. A method of treating a hematological cancer, comprising administering to a patient in need of such treatment a therapeutically effective amount of a compound of Formula IIIa:
or a pharmaceutically acceptable salt thereof, wherein:
R 1 and R 2 are each independently selected from H, methyl, ethyl, propyl, cyclopropyl, cyclopentyl, pyran-4-yl, phenyl, pyridin-2-yl, 2-chloro-4-phenyl, 2-chloro-4-fluorophenyl, and 2-hydroxyethyl;
or R 1 and R 2 together with the carbon atom to which they are attached form cyclopropyl;
R 3 is H, methyl, ethyl, or propyl, wherein said methyl is optionally substituted with cyclopropyl, pyridinyl, —C(═O)NH 2 , —C(═O)NHCH 3 , —C(═O)(4-methylpiperazin-1-yl), —C(═O)NH(4-methylpiperazin-1-yl), or —C(═O)OH;
R 5 is H, 1-methyl-1H-pyrazol-4-yl, 2-furyl, CN, NO 2 , methoxy, —C(═O)NH 2 , —C(═O)NH(CH 3 ), —C(═O)N(CH 3 ) 2 , —C(═O)-(morpholin-4-yl), —C(═O)CH 3 , —CH 2 OH, —CH 2 OCH 3 , —CH 2 NH 2 , —CH 2 NHSO 2 (CH 2 CH 3 ), —CH 2 NHC(═O)CH 3 , —CH(OH)CH 3 , —SO 2 CH 3 , —SO 2 CH 2 CH 3 , —SO 2 -(isopropyl), —SO 2 N(CH 3 ) 2 , —SO 2 NH(CH 3 ), —SO 2 —NH(isopropyl), or —SO 2 -(piperidin-1-yl); and
R 7 is methyl.
2. The method of claim 1 , wherein R 1 and R 2 are each independently selected from H, methyl, ethyl, propyl, cyclopropyl, cyclopentyl, pyran-4-yl, phenyl, pyridin-2-yl, 2-chloro-4-phenyl, and 2-hydroxyethyl.
3. The method of claim 1 , wherein one of R 1 and R 2 is H and the other is not H.
4. The method of claim 1 , wherein R 1 and R 2 are each methyl, ethyl, or propyl.
5. The method of claim 1 , wherein R 1 and R 2 are each methyl.
6. The method of claim 1 , wherein R 3 is H, methyl, ethyl, or propyl, wherein said methyl is optionally substituted with cyclopropyl, pyridinyl, —C(═O)NHCH 3 , —C(═O)NH(4-methylpiperazin-1-yl), or —C(═O)OH.
7. The method of claim 1 , wherein R 3 is methyl.
8. The method of claim 1 , wherein R 5 is —SO 2 CH 3 , —SO 2 CH 2 CH 3 , —SO 2 -(isopropyl), —SO 2 N(CH 3 ) 2 , —SO 2 NH(CH 3 ), —SO 2 —NH(isopropyl), or —SO 2 -(piperidin-1-yl).
9. The method of claim 1 , wherein R 5 is S(O) 2 CH 3 .
10. The method of claim 1 , wherein R 5 is S(O) 2 CH 2 CH 3 .
11. The method of claim 1 , wherein the compound is selected from:

or a pharmaceutically acceptable salt of any of the aforementioned.
12. The method of claim 1 , wherein the hematological cancer is leukemia.
13. The method of claim 12 , wherein the leukemia is acute eosinophilic leukemia, acute erythroid leukemia, acute lymphoblastic leukemia, acute megakaryoblastic leukemia, acute monocytic leukemia, acute promyelocytic leukemia, adult T-cell leukemia, aggressive NK-cell leukemia, B-cell chronic lymphocytic leukemia, B-cell prolymphocytic leukemia, hairy cell leukemia, acute lymphocytic leukemia, acute myelogenous leukemia, chronic lymphocytic leukemia, mast cell leukemia, or acute myeloid leukemia (AML).
14. The method of claim 1 , wherein the hematological cancer is lymphoma.
15. The method of claim 14 , wherein the lymphoma is adult T-cell lymphoma, AIDS-related lymphoma, anaplastic large cell lymphoma, angioimmunoblastic T-cell lymphoma, B-cell lymphoma, Burkitt's lymphoma, cutaneous T-cell lymphoma, diffuse large B-cell lymphoma, enteropathy-associated T-cell lymphoma, follicular lymphoma, hepatosplenic T-cell lymphoma, Hodgkin's lymphoma, non-Hodgkin's lymphoma, MALT lymphoma, mantle cell lymphoma, marginal zone B-cell lymphoma, precursor T-lymphoblastic lymphoma, primary central nervous system lymphoma, primary effusion lymphoma, splenic marginal zone lymphoma, or T-cell lymphoma.
16. The method of claim 1 , wherein the hematological cancer is multiple myeloma.
17. The method of claim 1 , wherein the hematological cancer is diffuse large B-cell lymphoma.
18. The method of claim 1 , wherein the hematological cancer is acute myeloid leukemia (AML).
19. A method of treating a hematological cancer, comprising administering to a patient in need of such treatment a therapeutically effective amount of 2,2,4-trimethyl-8-(6-methyl-7-oxo-6,7-dihydro-1H-pyrrolo[2,3-c]pyridin-4-yl)-6-(methylsulfonyl)-2H-1,4-benzoxazin-3(4H)-one or a pharmaceutically acceptable salt thereof.
20. The method of claim 19 , wherein the hematological cancer is leukemia.
21. The method of claim 20 , wherein the leukemia is acute eosinophilic leukemia, acute erythroid leukemia, acute lymphoblastic leukemia, acute megakaryoblastic leukemia, acute monocytic leukemia, acute promyelocytic leukemia, adult T-cell leukemia, aggressive NK-cell leukemia, B-cell chronic lymphocytic leukemia, B-cell prolymphocytic leukemia, hairy cell leukemia, acute lymphocytic leukemia, acute myelogenous leukemia, chronic lymphocytic leukemia, mast cell leukemia, or acute myeloid leukemia (AML).
22. The method of claim 19 , wherein the hematological cancer is lymphoma.
23. The method of claim 22 , wherein the lymphoma is adult T-cell lymphoma, AIDS-related lymphoma, anaplastic large cell lymphoma, angioimmunoblastic T-cell lymphoma, B-cell lymphoma, Burkitt's lymphoma, cutaneous T-cell lymphoma, diffuse large B-cell lymphoma, enteropathy-associated T-cell lymphoma, follicular lymphoma, hepatosplenic T-cell lymphoma, Hodgkin's lymphoma, non-Hodgkin's lymphoma, MALT lymphoma, mantle cell lymphoma, marginal zone B-cell lymphoma, precursor T-lymphoblastic lymphoma, primary central nervous system lymphoma, primary effusion lymphoma, splenic marginal zone lymphoma, or T-cell lymphoma.
24. The method of claim 19 , wherein the hematological cancer is multiple myeloma.
25. The method of claim 19 , wherein the hematological cancer is diffuse large B-cell lymphoma.
26. The method of claim 19 , wherein the hematological cancer is acute myeloid leukemia (AML).
