Ovarian cancer vaccines
Provided herein are methods, kits and compositions for the treatment and/or prevention of ovarian cancer through the induction of an immune response against Anti-Mullerian Hormone Receptor, Type II (AMHR2).
1. A method of treating or preventing an ovarian cancer tumor in a female human subject comprising administering to the subject an immunogenic composition comprising an Anti-Mullerian Hormone Receptor, Type II (AMHR2) polypeptide, wherein
the AMHR2 polypeptide comprises the extracellular domain of human AMHR2,
wherein the immunogenic composition elicits an immune response specific for an AMHR2 polypeptide comprising the extracellular domain of human AMHR2.
2. The method of claim 1 , wherein the immunogenic composition comprises an adjuvant.
3. The method of claim 2 , wherein the adjuvant is selected from: Adjuvant 65, α-GalCer, aluminum phosphate, aluminum hydroxide, calcium phosphate, β-Glucan Peptide, CpG DNA, GM-CSF, GPI-0100, IFA, IFN-γ, IL-17, lipid A, lipopolysaccharide, Lipovant, Montanide, N-acety 1-muramy 1-L-alany 1-D-isoglutamine, Pam3CSK4, poly-IC, quil A, trehalose dimycolate and zymosan.
4. The method of claim 2 , wherein the adjuvant is one that induces a mixed type I/type 17 immune response.
5. The method of claim 1 , wherein the ovarian cancer tumor is epithelial ovarian cancer (EOC) tumor.
6. The method of claim 1 , wherein the ovarian cancer tumor expresses AMHR2.
7. The method of claim 1 , further comprising a step of determining whether the ovarian cancer tumor expresses AMHR2.
8. The method of claim 1 , wherein the immune response comprises a T-cell immune response against ovarian cancer cells in the subject.
9. The method of claim 1 , wherein the immune response comprises a B-cell immune response against ovarian cancer cells in the subject.
10. The method of claim 9 , wherein the B-cell immune response comprises an AMHR2-specific immunoglobulin G (IgG) response in the subject.
11. The method of claim 2 , wherein the adjuvant is aluminum hydroxide-based.
12. The method of claim 1 , wherein the immune response further comprises induction of a B-cell response against AMHR2-expressing ovarian cancer cells in a subject.
13. The method of claim 12 , wherein the immune response comprises a B-cell response comprising an AMHR2-specific IgG antibody response in the subject.
14. A method of preventing a primary ovarian cancer tumor in a female human subject comprising vaccinating the subject with a vaccine comprising an immunogenic composition comprising an Anti-Mullerian Hormone Receptor, Type II (AMHR2) polypeptide,
the AMHR2 polypeptide comprises the extracellular domain of human AMHR2,
wherein the immunogenic composition elicits an immune response specific for an AMHR2 polypeptide comprising the extracellular domain of AMHR2.
15. The method of claim 14 , wherein the female human subject comprises a BRCA1 or BRCA2 mutation in their genome.
16. The method of claim 14 , wherein the female human subject has a family history of ovarian cancer.
17. The method of claim 14 , wherein the female human subject is a post-menopausal human female.
