Abstract

This invention relates to devices and methods for purifying, detecting and using biological cells. A variety of cell types including viable tumor, stem, immune and sperm cells can be purified from a complex biological sample using a column, including a pipette tip column. Methods of the invention can aid research, diagnosis and treatment of cancer. Purified viable cells can be detected on the column or eluted from the column and detected. Cells on a column can be used as a stationary phase for liquid chromatography. Cells may be removed, recovered and analyzed.

Claims (23)

1. A method for making and using an adhesion column, comprised of:

(a) growing cells in a suspension of beads or particles, wherein the suspension is comprised of a growth medium, and wherein the cells attach to the beads or particles to produce adherent cells;

(b) providing a column;

(c) introducing the adherent cells into the column to produce an adhesion column;

(d) providing at least one analyte;

(e) passing the analyte through the adhesion column, wherein the analyte interacts with or binds to the adherent cells in the adhesion column.

2. The method of claim 1 , wherein the analyte is passed through the adhesion column in a mobile phase.

3. The method of claim 1 , wherein following step (e), an eluent is passed through the adhesion column.

4. The method of claim 3 , wherein the eluent is directed to a detector.

5. The method of claim 1 , wherein the cells are recovered and analyzed.

6. The method of claim 1 , wherein the cells are lysed following step (c) or step (e).

7. The method of claim 6 , wherein components of the lysed cells are recovered and analyzed.

8. The method of claim 1 , wherein the column is a pipette tip, syringe or cartridge.

9. The method of claim 2 , wherein the mobile phase is passed through column using back and forth flow.

10. The method of claim 3 , wherein the eluent is passed through the column using back and forth flow.

11. The method of claim 2 , wherein the mobile phase is passed through column using unidirectional flow.

12. The method of claim 1 , wherein the eluent is passed through the column using unidirectional flow.

13. The method of claim 1 , wherein the method is automated.

14. The method of claim 1 , wherein the method is performed with a plurality of columns operated in parallel.

15. The method of claim 1 , wherein the cells are stem cells, mammalian cells, human cells, cancer cells, natural killer (NK) cells, monocytes, macrophages, endothelial cells, smooth muscle cells, dendritic cells, mast cells, fibroblasts or epithelial cells.

16. The method of claim 1 , wherein following step (c), a library of compounds is passed through the column.

17. The method of claim 1 , wherein the analyte is a drug or drug candidate.

18. The method of claim 1 , wherein the sample or analyte is a tag.
