Abstract

Disclosed herein include microRNA antagonists, therapeutic compositions that include one or more of such microRNA antagonists, and methods of treating and/or ameliorating cardiac diseases and/or muscular dystrophy disorders with the microRNA antagonists. Also included are combination therapies, wherein a therapeutic composition disclosed herein and an additional therapy agent are provided to a subject having or suspected of having cardiac disease and/or muscular dystrophy disorder. In particular, some embodiments disclosed herein relate to compositions and methods for transiently administering a mixture of microRNA antagonists for promoting cardiomyocyte proliferation and cardiac regeneration.

Claims (23)

1. A cloning or expression vector, wherein the cloning or expression vector comprises:

a nucleotide sequence having at least 96% identity to one of the nucleotide sequences set forth in SEQ ID NOs: 86 and 87, or

a nucleotide sequence having at least 90% identity to one of the nucleotide sequences set forth in SEQ ID NOs: 88 and 89.

2. The cloning or expression vector of claim 1 , wherein the cloning or expression vector is a viral vector.

3. A method for promoting cardiac muscle regeneration or for treating a cardiac disease in a subject, comprising administering to a subject in need thereof a pharmaceutical composition comprising a cloning or expression vector of claim 1 .

4. The method of claim 3 , wherein the subject in need thereof is a human subject suffering from a cardiac disease.

The method of claim 3 , further comprising administering to the subject at least one additional therapeutic agent, wherein the at least one additional therapeutic agent is selected from the group consisting of Idebenone, Eplerenone, VECTTOR, AVI-4658, Ataluren/PTC124/Translarna, BMN044/PRO044, CAT-1004, MicroDystrophin AAV Gene Therapy (SGT-001), Galectin-1 Therapy (SB-002), LTBB4 (SB-001), rAAV2.5-CMV-minidystrophin, Glutamine, NFKB inhibitors, Sarcoglycan, delta (35 kDa dystrophin-associated glycoprotein), insulin like growth factor-1 (IGF-1), and combinations thereof.

6. The method of claim 5 , wherein the pharmaceutical composition and the at least one additional therapeutic agent are administered to the subject sequentially or in rotation.

7. The method of claim 3 , comprising modulating proliferation of a cardiomyocyte and/or muscle cell in the subject.

8. The method of claim 3 , comprising increasing proliferation of a heart cell and/or increasing the expression and/or activity of proteins involved in muscle structure and/or function and/or regeneration in the subject.

9. The method of claim 3 , wherein the subject is a human subject.

10. The method of claim 9 , wherein the human subject suffers from a muscular dystrophy (MD) disorder.

The method of claim 3 , further comprising administering to the subject at least one additional therapeutic agent, wherein the at least one additional therapeutic agent is selected from the group consisting of Idebenone, Eplerenone, VECTTOR, AVI-4658, Ataluren/PTC124/Translarna, BMN044/PRO044, CAT-1004, MicroDystrophin AAV Gene Therapy (SGT-001), Galectin-1 Therapy (SB-002), LTBB4 (SB-001), rAAV2.5-CMV-minidystrophin, Glutamine, NFKB inhibitors, Sarcoglycan, delta (35 kDa dystrophin-associated glycoprotein), insulin like growth factor-1 (IGF-1), and combinations thereof.

12. The cloning or expression vector of claim 1 , wherein the cloning or expression vector comprises:

a nucleotide sequence having at least 98% identity to one of the nucleotide sequences set forth in SEQ ID NOs: 86 and 87, or

a nucleotide sequence having at least 95% identity to one of the nucleotide sequences set forth in SEQ ID NOs: 88 and 89.

13. The cloning or expression vector of claim 1 , wherein the cloning or expression vector comprises:

a nucleotide sequence having one of the nucleotide sequences set forth in SEQ ID NOs: 86 and 87, or

a nucleotide sequence having one of the nucleotide sequences set forth in SEQ ID NOs: 88 and 89.

14. A method for promoting cardiac muscle regeneration or for treating a cardiac disease in a subject, comprising administering to a subject in need thereof a pharmaceutical composition comprising a cloning or expression vector of claim 12 .

15. The method of claim 14 , wherein the subject in need thereof is a human subject suffering from a cardiac disease.

16. A method for promoting cardiac muscle regeneration or for treating a cardiac disease in a subject, comprising administering to a subject in need thereof a pharmaceutical composition comprising a cloning or expression vector of claim 13 .

17. The method of claim 16 , wherein the subject in need thereof is a human subject suffering from a cardiac disease.
