Abstract

Provided herein is a stabilized oxygen transport matrix that includes a reversible oxygen binding protein, such as hemoglobin, immobilized throughout the stabilized oxygen transport matrix. The stabilized oxygen transport matrix is used to transport oxygen and can be used as an oxygen transport region and a reaction region of an analyte sensor, such as an implantable glucose sensor. The reversible binding protein can also function as an oxygen probe within the analyte sensor.

Claims (38)

1. A sensor, comprising:

a) an oxygen transport region comprising:

(i) a stabilized oxygen transport matrix;

(ii) an oxygen permeable first surface in communication with an external environment; and

(iii) an oxygen permeable second surface that is impermeable to a target analyte;

b) a target analyte reaction region in communication with the oxygen transport region at the oxygen permeable second surface, wherein the target analyte reaction region comprises a target analyte oxidase enzyme, wherein the target analyte reaction region is delineated by a target analyte-permeable surface on a single side of the target analyte reaction region, a distal boundary opposite the target analyte permeable surface, and wherein, when the target analyte enters the target analyte reaction region through the target analyte-permeable surface, the target analyte diffuses toward the distal boundary and reacts with oxygen, an analyte-derived oxygen gradient is produced within the target analyte reaction region; and

c) a sensing region comprising at least two detector probes in communication with the target analyte reaction region, wherein the two detector probes are positioned to monitor different spatial regions of the analyte-derived oxygen gradient, the sensing region is contiguous with the sensing surface.

2. The sensor of claim 1 , wherein the target analyte is selected from the group consisting of glucose, galactose, lactose, peroxide, cholesterol, amino acids, alcohol, lactic acid and mixtures thereof.

3. The sensor of claim 1 , wherein the target analyte is glucose and the sensor is a glucose sensor.

4. The sensor of claim 1 , further comprising a first reversible oxygen binding protein immobilized in the stabilized oxygen transport matrix.

5. The sensor of claim 4 , wherein the first reversible oxygen binding protein is an engineered hemeprotein or a heme derivative.

6. The sensor of claim 5 , wherein the first reversible oxygen binding protein is hemoglobin.

7. The sensor of claim 1 , wherein at least one of the oxygen transport region, the target analyte reaction region, and the sensing region comprise a stabilized matrix.

8. The sensor of claim 1 , wherein the at least two detector probes are at least two non-oxygen consuming detector probes.

9. The sensor of claim 8 , wherein the sensing region further comprises an oxygen probe.

10. The sensor of claim 9 , wherein the oxygen probe comprises a reversible oxygen binding protein.

11. The sensor of claim 1 , wherein the target analyte reaction region is parallel to the oxygen permeable second surface.

12. The sensor of claim 1 , wherein the at least two detector probes are capable of emitting and/or receiving light.

13. The sensor of claim 12 , wherein the at least two detector probes are capable of emitting and receiving light at an oxygen sensitive engineered hemeprotein, or heme derivative absorption wavelength.

14. A sensor, comprising:

a) an oxygen transport region comprising:

(i) a stabilized oxygen transport matrix;

(ii) an oxygen permeable first surface in communication with an external environment; and

(iii) an oxygen permeable second surface that is impermeable to a target analyte, wherein the stabilized oxygen transport matrix is between the oxygen permeable first surface and the oxygen permeable second surface;

b) a target analyte reaction region in communication with the oxygen transport region at the oxygen permeable second surface, wherein the target analyte reaction region comprises a target analyte oxidase enzyme, the target analyte reaction region delineated by a target analyte-permeable surface on a single side of the target analyte reaction region and a distal boundary opposite the target analyte permeable surface, and wherein, when the target analyte enters the target analyte reaction region through the target analyte-permeable surface and reacts with oxygen, an analyte-derived oxygen gradient is produced within the target analyte reaction region in a first direction from the target analyte permeable surface to the distal boundary;

c) a sensing region comprising at least two detector probes in communication with the target analyte reaction region, wherein the two detector probes are positioned along a length extending in the first direction to monitor different spatial regions of the analyte-derived oxygen gradient.

15. A sensor, comprising:

a) an oxygen transport region comprising:

(i) a stabilized oxygen transport matrix;

(ii) an oxygen permeable first surface in communication with an external environment; and

(iii) an oxygen permeable second surface that is impermeable to a target analyte, wherein the stabilized oxygen transport matrix is between the oxygen permeable first surface and the oxygen permeable second surface;

b) a target analyte reaction region in communication with the oxygen transport region at the oxygen permeable second surface, wherein the target analyte reaction region comprises a target analyte oxidase enzyme, wherein the target analyte reaction region is delineated by a target analyte-permeable surface on a single side of the target analyte reaction region, a distal boundary opposite the target analyte permeable surface, and wherein, when the target analyte enters the target analyte reaction region through the target analyte-permeable surface and reacts with oxygen, an analyte-derived oxygen gradient is produced within the target analyte reaction region; and

c) a sensing region comprising at least two detector probes in communication with the target analyte reaction region, wherein the two detector probes are positioned to monitor different spatial regions of the analyte-derived oxygen gradient.

16. The sensor of claim 1 , wherein the oxygen permeable second surface is parallel to a long axis of the of the target analyte reaction region.

17. The sensor of claim 1 , wherein the target analyte permeable surface is perpendicular to the oxygen permeable first surface.

18. The sensor of claim 1 , wherein the stabilized oxygen transport matrix comprises a polymer and a reversible oxygen binding protein, and wherein the reversible oxygen binding protein is immobilized within the stabilized oxygen transport matrix.

19. The sensor of claim 1 , wherein the stabilized oxygen transport matrix is between the oxygen permeable first surface and the oxygen permeable second surface.

20. The sensor of claim 1 , wherein the oxygen permeable first surface and the oxygen permeable second surface are at opposing ends of the stabilized oxygen transport matrix.
