Abstract

An apparatus for sorting cells includes a measurement volume that contains a cell to be measured, a light source that provides light to cause an emission by a fluorescent label attached to the cell, and an optic device that directs the light through the measurement volume. The apparatus flows the cells through the measurement volume such that as the cell flows through the measurement volume, it interacts with the light, resulting in a change in light originating from the measurement volume, the change in light is a fluorescence emission. Another optic device directs a portion of the light originating from the measurement volume to a detector, which detects the portion of the light. A processor operably coupled to the detector generates an estimate of DNA quantity in the cell based on the change in light originating from the measurement volume, and determines a characteristic of the cell from the estimate.

Claims (76)

1. An apparatus for sorting a plurality of sperm cells, the apparatus comprising:

a light source configured to provide a light, wherein the light source comprises a UV or visible laser configured to cause an emission by a fluorescent label attached to each sperm cell in the plurality of sperm cells;

a measurement volume configured to contain a sample to be measured;

a first optic device that is configured to direct the light through the measurement volume;

wherein the apparatus is configured to flow the plurality of sperm cells through the measurement volume one by one, such that as each sperm cell in the plurality of sperm cells flows through the measurement volume it interacts with the light from the light source, thereby resulting in a change in light originating from the measurement volume, wherein the change in light is the emission due to fluorescence,

a second optic device configured to direct a portion of the light originating from the measurement volume to a detector;

the detector configured to detect the portion of the light directed by the second optic device;

a signal processor operably coupled to the detector, the signal processor configured to:

generate an estimate of the DNA quantity in each sperm cell in the plurality of sperm cells based on the change in light originating from the measurement volume; and

determine a characteristic of each of the plurality of sperm cells derived from the estimate, wherein the characteristic is whether each sperm cell is x-bearing or y-bearing;

an ejection nozzle configured to:

break a fluid stream containing the plurality of sperm cells into a plurality of droplets; and

electrically charge one or more or the droplets based on the determined characteristic of the sperm cell inside the droplet; and

a sorting mechanism configured to sort the plurality of sperm cells into one of two or more output positions.

2. The apparatus of claim 1 , wherein the measurement volume is co-located with the ejection nozzle.

3. The apparatus of claim 1 , further comprising a cell destruction or immobilization facility.

4. The apparatus of claim 1 , wherein the sorting mechanism comprises a photodamage cell selection sorting mechanism.

5. The apparatus of claim 1 , wherein the plurality of sperm cells is selected from the group consisting of bovine sperm cells, ovine sperm cells, porcine sperm cells, and equine sperm cells.

6. The apparatus of claim 1 , wherein the detector is configured to perform multiple measurements with and without sperm cells in the measurement volume.

7. The apparatus of claim 1 , wherein the measurement volume is co-located with the output device.

8. The apparatus of claim 1 , further comprising a cell destruction or immobilization facility.

9. The apparatus of claim 1 , wherein the sorting mechanism comprises a photodamage cell selection sorting mechanism.

10. The apparatus of claim 1 , wherein the plurality of sperm cells is selected from the group consisting of bovine sperm cells, ovine sperm cells, porcine sperm cells, and equine sperm cells.

11. The apparatus of claim 1 , wherein the detector is configured to perform multiple measurements with and without sperm cells in the measurement volume.

12. An apparatus for sorting a plurality of sperm cells, the apparatus comprising:

a light source configured to provide a light, wherein the light source comprises a UV or visible laser configured to cause an emission by a fluorescent label attached to each sperm cell in the plurality of sperm cells;

a measurement volume configured to contain a sample to be measured;

a first optic device that is configured to direct the light through the measurement volume;

wherein the light directed to the measurement volume results in an illumination pattern in the measurement volume;

wherein the apparatus is configured to flow the plurality of sperm cells through the measurement volume one by one, such that as each sperm cell in the plurality of sperm cells flows through the measurement volume it interacts with the illumination pattern, thereby resulting in a fluorescent signal from the measurement volume;

a second optic device configured to direct a portion of the fluorescent signal from the measurement volume to a detector;

a detector configured to detect the portion of the fluorescent signal directed by the second optic device;

a signal processor operably coupled to the detector, the signal processor configured to:

generate an estimate of the DNA quantity in each sperm cell in the plurality of sperm cells based on the fluorescent signal; and

determine a characteristic of the each of the plurality of sperm cells derived from the estimate, wherein the characteristic is whether each sperm cell is x-bearing or y-bearing;

an ejection nozzle configured to:

break a fluid stream containing the plurality of sperm cells into a plurality of droplets; and

electrically charge one or more or the droplets based on the determined characteristic of the sperm cell inside the droplet; and

a sorting mechanism configured to sort the plurality of sperm cells into one of two or more output positions.

13. The apparatus of claim 12 , wherein the measurement volume is co-located with the ejection nozzle.

14. The apparatus of claim 12 , further comprising a cell destruction or immobilization facility.

15. The apparatus of claim 12 , wherein the sorting mechanism comprises a photodamage cell selection sorting mechanism.

16. The apparatus of claim 12 , wherein the plurality of sperm cells is selected from the group consisting of bovine sperm cells, ovine sperm cells, porcine sperm cells, and equine sperm cells.

17. The apparatus of claim 12 , wherein the detector is configured to perform multiple measurements with and without sperm cells in the measurement volume.

18. A method for sorting a plurality of sperm cells, the method comprising:

providing a light from a light source, wherein the light source comprises a UV or visible laser configured to cause an emission by a fluorescent label attached to each sperm cell in the plurality of sperm cells;

directing the light through a measurement volume via a first optic device, so as to form an illumination pattern in the measurement volume;

flowing the plurality of sperm cells through the measurement volume one by one, such that as each sperm cell of the plurality of sperm cells flows through the measurement volume it interacts with the light from the light source, thereby resulting in a fluorescent signal from the measurement volume;

directing a portion of the fluorescent signal from the measurement volume to a detector via a second optic device;

detecting the portion of the fluorescent signal;

generating an estimate of the DNA quantity in each sperm cell in the plurality of sperm cells based on the fluorescent signal;

determining a characteristic of each of the plurality the sperm cells derived from the estimate, wherein the characteristic is whether each sperm cell is x-bearing or y-bearing;

breaking a fluid stream containing the plurality of sperm cells into a plurality of droplets via an ejection nozzle;

electrically charging one or more or the droplets based on the determined characteristic of the sperm cell inside the droplet; and

sorting the plurality of sperm cells into one of two or more output positions based on the characteristic.

19. The method of claim 18 , further comprising cryogenically preserving a plurality of the sorted sperm cells.

20. The method of claim 18 , wherein the plurality of sperm cells is sorted via an electrostatic field.

21. The method of claim 18 , wherein the plurality of sperm cells is sorted via a cell destruction or immobilization facility.

22. The method of claim 18 , wherein the plurality of sperm cells is sorted via a photodamage cell selection sorting mechanism.

23. The method of claim 18 , wherein the plurality of sperm cells is selected from the group consisting of bovine sperm cells, ovine sperm cells, porcine sperm cells, and equine sperm cells.

24. The method of claim 18 , wherein the light directed through the measurement volume results in an illumination pattern in the measurement volume.

25. The method of claim 18 , wherein the detector performs multiple measurements with and without sperm cells in the measurement volume.

26. An apparatus for sorting a plurality of sperm cells, the apparatus comprising:

a light source configured to provide a light, wherein the light source comprises a UV or visible laser configured to cause an emission by a fluorescent label attached to each sperm cell in the plurality of sperm cells;

a measurement volume configured to contain a sample to be measured;

a first optic device that is configured to direct the light through the measurement volume;

wherein the apparatus is configured to flow the plurality of sperm cells through the measurement volume one by one, such that as each sperm cell in the plurality of sperm cells flows through the measurement volume it interacts with the light from the light source, thereby resulting in a change in light originating from the measurement volume, wherein the change in light is the emission due to fluorescence;

a second optic device configured to direct a portion of the light originating from the measurement volume to a detector;

the detector configured to detect the portion of the light directed by the second optic device;

a signal processor operably coupled to the detector, the signal processor configured to:

generate an estimate of the DNA quantity in each sperm cell in the plurality of sperm cells based on the change in light originating from the measurement volume; and

determine a characteristic of each of the plurality of sperm cells derived from the estimate, wherein the characteristic is whether each sperm cell is x-bearing or y-bearing;

an output device configured to:

break a fluid stream containing the plurality of sperm cells into a plurality of droplets; and

electrically charge one or more or the droplets based on the determined characteristic of the sperm cell inside the droplet; and

a sorting mechanism configured to sort the plurality of sperm cells into one of two or more output positions.
