Abstract

Transgenic INIR12 maize plants comprising a vip3Aa19 expression cassette linked to a secondary nopaline synthase terminator element which lack a selectable marker gene are provided. In certain embodiments such INIR12 transgenic plants comprise modifications that provide for facile excision of the INIR12 transgenic locus from the maize plant genome. Genomic DNA of INIR12 transgenic plants, detection of INIR12 plants and products thereof, methods of making INIR12 plants, and use of INIR12 plants to facilitate breeding are disclosed.

Claims (10)

1. A transgenic maize plant cell comprising a transgenic locus, wherein the transgenic locus comprises a modification of the original MIR162 transgenic locus comprising SEQ ID NO: 1 or the original MIR162 transgenic locus present in seed deposited at the ATCC under accession No. PTA-8166, wherein the transgenic locus comprises the first zmUbiInt promoter operably linked to the Vip3Aa gene, the CaMV35S terminator, and the NOS terminator of the original MIR162 transgenic locus comprising SEQ ID NO:1 or present in the seed deposited at the ATCC under accession No. PTA-8166, and wherein the modification comprises a deletion of the DNA molecule encoding the second ZmUbilnt promoter and operably linked phosphomannose isomerase coding region of the original MIR162 locus.

A transgenic maize plant seed comprising a transgenic locus, wherein the transgenic locus comprises a modification of the original_MIR162 transgenic locus comprising SEQ ID NO: 1 or the original MIR162 transgenic locus present in seed deposited at the ATCC under accession No. PTA-8166, wherein the transgenic locus comprises the first zmUbilnt promoter operably linked to the Vip3Aa gene, the CaMV35S terminator, and the NOS terminator of the original MIR162 transgenic locus comprising SEQ ID NO:1 or present in the seed deposited at the ATCC under accession No. PTA-8166, and wherein the modification comprises a deletion of the DNA molecule encoding the second ZmUbilnt promoter and operably linked phosphomannose isomerase coding region of the original MIR162 locus.

A transgenic maize plant comprising a transgenic locus, wherein the transgenic locus comprises a modification of the original MIR162 transgenic locus comprising SEQ ID NO: 1 or the original MIR162 transgenic locus present in seed deposited at the ATCC under accession No. PTA-8166, wherein the transgenic locus comprises the first zmUbilnt promoter operably linked to the Vip3Aa gene, the CaMV35S terminator, and the NOS terminator of the original MIR162 transgenic locus comprising SEQ ID NO:1 or present in the seed deposited at the ATCC under accession No. PTA-8166, and wherein the modification comprises a deletion of the DNA molecule encoding the second ZmUbilnt promoter and operably linked phosphomannose isomerase coding region of the original MIR162 locus.

A method for obtaining a bulked population of seed comprising selfing the transgenic maize plant of claim 3 and harvesting transgenic seed comprising the transgenic locus, wherein the transgenic locus comprises a modification of the original MIR162 transgenic locus comprising SEQ ID NO: 1 or the original MIR162 transgenic locus present in seed deposited at the ATCC under accession No. PTA-8166, wherein the transgenic locus comprises the first zmUbilnt promoter operably linked to the Vip3Aa gene, the CaMV35S terminator, and the NOS terminator of the original MIR162 transgenic locus comprising SEQ ID NO:1 or present in the seed deposited at the ATCC under accession No. PTA-8166, and wherein the modification comprises a deletion of a DNA molecule encoding the second ZmUbilnt promoter and operably linked phosphomannose isomerase coding region of the original MIR162 locus.

5. The transgenic maize plant cell of claim 1 , wherein the transgenic locus further comprises a deletion of a DNA segment comprising non-essential transgenic DNA in a junction polynucleotide of the original MIR162 transgenic locus.

6. The transgenic maize plant cell of claim 5 , wherein the deletion results in the 5′ junction polynucleotide of SEQ ID NO: 40.

7. The transgenic maize plant cell of claim 6 , wherein the transgenic locus comprises the DNA molecule of SEQ ID NO: 43.

8. The transgenic maize plant seed of claim 2 , wherein the transgenic locus comprises the DNA molecule of SEQ ID NO: 43.

9. The transgenic maize plant of claim 3 , wherein the transgenic locus comprises the DNA molecule of SEQ ID NO: 43.

10. The method of claim 4 , wherein the transgenic locus comprises the DNA molecule of SEQ ID NO: 43.
