Antigen-binding molecule capable of binding to two or more antigen molecules repeatedly
The present inventors discovered that antibodies having weaker antigen-binding activity at the early endosomal pH in comparison with that at the pH of plasma are capable of binding to multiple antigen molecules with a single antibody molecule, have long half-lives in plasma, and have improved durations of time in which they can bind to antigen.
1. A method of neutralizing an antigen in vivo, comprising:
administering an antibody to a human subject for a therapeutic purpose;
wherein the antibody comprises a constant region and a variable region comprising a heavy chain variable (VH) region and a light chain variable (VL) region;
wherein the variable region comprises one to nine histidine residue substitutions;
wherein the constant region comprises one or more mutations that enhance binding to neonatal Fc receptor (FcRn); and
wherein, as compared to without the one to nine histidine residue substitutions and the one or more mutations that enhance binding to FcRn, the antibody:
(1) has a KD(pH5.8)/KD(pH7.4) value that is higher; and
(2) has a prolonged retention time in plasma in vivo.
2. The method of claim 1 , wherein the KD(pH5.8)/KD(pH7.4) value of the antibody is at least two times higher.
3. The method of claim 1 , wherein the one to nine histidine residue substitutions comprises at least one histidine residue substitution that is located within a complementarity-determining region (CDR) sequence of the variable region, according to Kabat numbering.
4. The method of claim 1 , wherein one of the one to nine histidine residue substitutions is located within a CDR sequence of the VH region, according to Kabat numbering.

The method of claim 1 , wherein the one to nine histidine residue substitutions comprise a histidine residue substitution at a site selected from the group consisting of heavy chain (H) position H27, H31, H32, H33, H35, H50, H58, H59, H61, H62, H63, H64, H65, H99, H100b, and H102, and light chain (L) position L24, L27, L28, L32, L53, L54, L56, L90, L92, and L94, numbered according to Kabat numbering.
6. The method of claim 1 , wherein the one to nine histidine residue substitutions comprise a histidine residue substitution at H27, numbered according to Kabat numbering.
7. The method of claim 1 , wherein the prolonged retention time in plasma in vivo is assessed by measuring antibody concentration in plasma over time.
8. The method of claim 7 , wherein the measuring antibody concentration in plasma over time comprises determining half-life of the antibody in plasma.
9. The method of claim 7 , wherein the measuring antibody concentration in plasma over time is performed in a mouse in the presence of the antigen.
10. The method of claim 9 , wherein the mouse expresses the antigen.
11. The method of claim 9 , wherein the KD(pH5.8)/KD(pH7.4) value of the antibody is up to 164 times higher.

The method of claim 9 , wherein the one to nine histidine residue substitutions comprise one histidine residue substitution or two histidine residue substitutions, and wherein the one histidine residue substitution or one or both of the two histidine residue substitutions is located within a CDR sequence of the variable region, according to Kabat numbering.
13. The method of claim 12 , wherein one of the two histidine residue substitutions is located within a CDR sequence of the VH region, according to Kabat numbering.
14. The method of claim 12 , wherein the one histidine residue substitution is, or the two histidine residue substitutions comprise, a histidine residue substitution at a site selected from the group consisting of H27, H31, H32, H33, H35, H50, H58, H59, H61, H62, H63, H64, H65, H99, H100b, H102, L24, L27, L28, L32, L53, L54, L56, L90, L92, and L94, numbered according to Kabat numbering.
15. The method of claim 12 , wherein the two histidine residue substitutions comprise a histidine residue substitution at H27, numbered according to Kabat numbering.
16. The method of claim 12 , wherein the measuring antibody concentration in plasma over time comprises determining half-life of the antibody in plasma.
17. The method of claim 7 , wherein the measuring antibody concentration in plasma over time is performed in a monkey.
18. The method of claim 17 , wherein the KD(pH5.8)/KD(pH7.4) value of the antibody is up to 164 times higher.

The method of claim 17 , wherein the one to nine histidine residue substitutions comprise one histidine residue substitution or two histidine residue substitutions, and wherein the one histidine residue substitution or one or both of the two histidine residue substitutions is located within a CDR sequence of the variable region, according to Kabat numbering.
20. The method of claim 19 , wherein one of the two histidine residue substitutions is located within a CDR sequence of the VH region, according to Kabat numbering.
21. The method of claim 19 , wherein the one histidine residue substitution is, or the two histidine residue substitutions comprise, a histidine residue substitution at a site selected from the group consisting of H27, H31, H32, H33, H35, H50, H58, H59, H61, H62, H63, H64, H65, H99, H100b, H102, L24, L27, L28, L32, L53, L54, L56, L90, L92, and L94, numbered according to Kabat numbering.
22. The method of claim 19 , wherein the two histidine residue substitutions comprise a histidine residue substitution at H27, numbered according to Kabat numbering.
23. The method of claim 19 , wherein the measuring antibody concentration in plasma over time comprises determining half-life of the antibody in plasma.
24. The method of claim 7 , wherein the measuring antibody concentration in plasma over time is performed in a human.
25. The method of claim 24 , wherein the KD(pH5.8)/KD(pH7.4) value of the antibody is up to 164 times higher.

The method of claim 24 , wherein the one to nine histidine residue substitutions comprise one histidine residue substitution or two histidine residue substitutions, and wherein the one histidine residue substitution or one or both of the two histidine residue substitutions is located within a CDR sequence of the variable region, according to Kabat numbering.
27. The method of claim 26 , wherein one of the two histidine residue substitutions is located within a CDR sequence of the VH region, according to Kabat numbering.
28. The method of claim 26 , wherein the one histidine residue substitution is, or the two histidine residue substitutions comprise, a histidine residue substitution at a site selected from the group consisting of H27, H31, H32, H33, H35, H50, H58, H59, H61, H62, H63, H64, H65, H99, H100b, H102, L24, L27, L28, L32, L53, L54, L56, L90, L92, and L94, numbered according to Kabat numbering.
29. The method of claim 26 , wherein the two histidine residue substitutions comprise a histidine residue substitution at H27, numbered according to Kabat numbering.
