Abstract

Provided herein are compositions comprising a biocompatible microsphere, a biofilm-generating probiotic bacterium, a prebiotic, and/or a prebiofilmic. Methods for preparing and formulating the compositions and methods for treating or preventing a disease using the compositions are also provided.

Claims (23)

1. A microsphere composition comprising:

a plurality of microspheres, each microsphere comprising dextran cross-linked with epichlorohydrin;

a water-soluble carbohydrate that diffuses from within the microsphere selected from the group consisting of maltose, sucrose, and combinations thereof;

L. reuteri strain ATCC 23272, wherein the L. reuteri is adhered to the surface of each microsphere; and

a pharmaceutically acceptable carrier.

2. The microsphere composition of claim 1 , wherein each microsphere has a diameter of about 1 microns to about 75 microns.

3. The microsphere composition of claim 1 , wherein each microsphere has a diameter of about 1 microns to about 100 microns.

4. A single dose composition comprising the microsphere composition of claim 1 , wherein the single dose composition comprises between about 1×10 7 CFU/ml and 1×10 9 CFU/ml of the L. reuteri.

5. The microsphere composition of claim 1 , comprising between about 1×10 7 CFU/ml and 1×10 9 CFU/ml of L. reuteri.

6. A microsphere composition comprising:

a plurality of microspheres, each microsphere comprising dextran cross-linked with epichlorohydrin, and a water-soluble carbohydrate that diffuses from within the microsphere;

L. reuteri adhered to the surface of each microsphere; and

a pharmaceutically acceptable carrier.

7. The microsphere composition of claim 6 , wherein the L. reuteri is strain ATCC 23272.

8. The microsphere composition of claim 7 , wherein the water-soluble carbohydrate is selected from the group consisting of maltose, sucrose, and combinations thereof.

9. The microsphere composition of claim 6 , wherein the water-soluble carbohydrate is selected from the group consisting of maltose, sucrose, and combinations thereof.

10. A microsphere composition comprising:

a plurality of microspheres, each microsphere comprising a biodegradable polymer, and a water-soluble carbohydrate that diffuses from within the microsphere;

L. reuteri strain ATCC 23272 adhered to the surface of each microsphere; and

a pharmaceutically acceptable carrier.

11. A method for treating necrotizing enterocolitis in an infant in need thereof, comprising administering to the infant in need thereof the composition of claim 1 [a composition comprising:

about 1×10 7 and 1×10 9 CFU/ml of L. reuteri plurality of microspheres each comprising: cross-linked dextran and a water-soluble carbohydrate selected from the group consisting of maltose, sucrose, and combinations thereof].

12. The method of claim 11 , wherein the [ L. reuteri is strain ATCC 23272] composition comprises between about 1×10 7 and 1×10 9 CFU/ml of L. reuteri.
