Abstract

The present invention relates to an alkynyl-substituted heterocyclic compound acting as an FGFR inhibitor, a preparation method therefor and a medical use thereof. In particular, the present invention relates to a compound as shown in general formula (I) and a pharmaceutically acceptable salt thereof; a pharmaceutical composition including the compound or a pharmaceutically acceptable salt thereof; a method for treating and/or preventing FGFR-associated diseases, particularly tumors, by using the compound or a pharmaceutically acceptable salt thereof; and a preparation method for the compound or a pharmaceutically acceptable salt thereof. The present invention also relates to the use of the compound or a pharmaceutically acceptable salt thereof, or the pharmaceutical composition including the compound or a pharmaceutically acceptable salt thereof in the preparation of a drug for treating and/or preventing FGFR-associated diseases, particularly tumors, wherein the definition of each substituent group in general formula (I) is the same as that in the description.

Claims (37)

1. A compound of Formula II, a stereoisomer, a pharmaceutically acceptable salt, or a 2 H, 3 H, 13 C, 14 C, 15 N, 18 O, 17 O, 31 P, 32 P, 35 S, 18 F and 36 Cl isotope derivative thereof:

wherein,

R 1 is H, NH 2 , or —NHR 3 ;

R 3 is C 1-6 alkyl, C 3-6 cycloalkyl, or 3-6 membered heterocyclyl, wherein the alkyl, the cycloalkyl, and the heterocyclyl are optionally substituted by halogen, cyano, —OR 4 , C 1-6 alkyl, C 3-6 cycloalkyl or 3-6 membered heterocyclyl;

X is absent or is C 1-6 alkylene;

Y is absent or is C 3-8 cycloalkylene or 3-8 membered heterocyclylene;

Z is cyano,

bond a is a double bond or a triple bond;

in the case where bond a is a double bond, R a , R b and R c are each independently H, cyano, halogen, C 1-6 alkyl, C 3-6 cycloalkyl, or 3-6 membered heterocyclyl, wherein the alkyl, the cycloalkyl, and the heterocyclyl are optionally substituted by one or more substituent groups selected from the group consisting of halogen, cyano, C 1-6 alkyl, C 3-6 cycloalkyl, or 3-6 membered heterocyclyl, OR 8 , and —NR 8 R 9 ;

in the case where bond a is a triple bond, R a and R c are absent, and R b is H, cyano, halogen, C 1-6 alkyl, C 3-6 cycloalkyl, or 3-6 membered heterocyclyl, wherein alkyl, cycloalkyl and heterocyclyl are optionally substituted by one or more substituent groups selected from the group consisting of halogen, cyano, C 1-6 alkyl, C 3-6 cycloalkyl, or 3-6 membered heterocyclyl, OR 8 , and —NR 8 R 9 ;

G a , G b , G c , and G d are each independently selected from the group consisting of H, halogen, cyano, C 1-6 alkyl, C 3-8 cycloalkyl, 3-8 membered heterocyclyl, OR B , and —NR 8 R 9 , and —C(O)NR 8 R 9 , wherein the alkyl, the cycloalkyl and the heterocyclyl are optionally substituted by one or more substituent groups selected from the group consisting of halogen, cyano, C 1-6 alkyl, C 3-8 cycloalkyl, or 3-8 membered heterocyclyl; and

R 4 , R 8 , and R 9 are each independently H or C 1-6 alkyl.

2. The compound according to claim 1 , wherein

X is absent;

in which means attaching to Z, and → means attaching to pyrazole; and

3. The compound according to claim 1 , wherein

G a , G b , G b , and G d are independently H, —OC 1-2 alkyl or halogen.

4. The compound according to claim 3 , wherein G a is —OC 1-2 alkyl, G b is OC 1-2 alkyl, G c is H, G d is H or halogen, and R 1 is —NHR 3 .

5. The compound according to claim 1 , wherein

R 1 is H or —NHR 3 ; and

R 3 is C 1-6 alkyl or 3-6 membered heterocyclyl, wherein the alkyl and the heterocyclyl are optionally substituted by halogen or —OR 4 .

6. The compound of claim 1 , wherein R 1 is NH 2 or NHR 3 , R 3 is C 1-6 alkyl, and the bond a is a double bond.

7. The compound according to claim 1 , wherein

8. The compound according to claim 1 , wherein G a is —C(O)NH(C 1-2 alkyl), G b is H, G c is H, G d is halogen, and R 1 is —NHR 3 .

9. The compound according to claim 1 , wherein the isotope derivative is 2 H.

11. A pharmaceutical composition comprising the compound according to claim 1 and a pharmaceutically acceptable carrier and an excipient.

12. The pharmaceutical composition according to claim 11 , wherein the compound is

13. A method of treating a FGFR-associated disease, comprising administering to a patient in need thereof a therapeutically effective amount of the compound according to claim 1 .

The method according to claim 13 , wherein said FGFR-associated disease is liver cancer, gastric cancer, non-small cell lung cancer, bladder cancer, esophageal cancer, melanoma, rhabdomyosarcoma, renal cell carcinoma, multiple myeloma, breast cancer, ovarian cancer, endometrial cancer, cervical cancer, colon cancer, bladder cancer, pancreatic cancer, lung cancer, breast cancer, or prostate cancer.

15. The method according to claim 13 , wherein said FGFR-associated disease is liver cancer, gastric cancer, non-small cell lung cancer, or bladder cancer.
