Abstract

Methods and composition related to the engineering of a novel protein with methionine-γ-lyase enzyme activity are described. For example, in certain aspects there may be disclosed a modified cystathionine-γ-lyase (CGL) comprising one or more amino acid substitutions and capable of degrading methionine. Furthermore, certain aspects of the invention provide compositions and methods for the treatment of cancer with methionine depletion using the disclosed proteins or nucleic acids.

Claims (19)

1. A polypeptide comprising a primate cystathionine gamma-lyase variant comprising at least one amino acid substitution as compared to a native primate cystathionine gamma-lyase, wherein the cystathionine gamma-lyase variant has a catalytic specificity constant (k cat /K M ) for degrading L-methionine between about 10 M −1 s −1 and about ×10 6 M −1 s −1 and wherein the at least one amino acid substitution comprises one or more of the following substitutions relative to SEQ ID NO: 1: E59V or E59N, R119L, and/or E339V.

2. The polypeptide of claim 1 , wherein the at least one amino acid substitution comprises a R119L and E339V substitution.

3. The polypeptide of claim 2 , wherein the at least one amino acid substitution further comprises an additional substitution of E59V or E59N.

4. The polypeptide of claim 1 , wherein the cystathionine gamma-lyase variant has a k cat /K M for degrading L-Methionine at least about 10 4 M −1 s −1 .

5. The polypeptide of claim 4 , wherein the cystathionine gamma-lyase variant has a k cat /K M for degrading L-Homocystine lower than about 50 M −1 s −1 .

6. The polypeptide of claim 1 , wherein the native primate cystathionine gamma-lyase has at least 95% sequence identity to the sequence of SEQ ID NO: 1.

7. The polypeptide of claim 6 , wherein the native primate cystathionine gamma-lyase has at least 98% sequence identity to the sequence of SEQ ID NO: 1.

8. The polypeptide of claim 6 , wherein the native primate cystathionine gamma-lyase has the sequence of any of SEQ ID NOs: 18-20.

9. The polypeptide of claim 6 , wherein the native primate cystathionine gamma lyase is a human cystathionine gamma-lyase (SEQ ID NO: 1).

10. The polypeptide of claim 1 , further comprising a heterologous peptide segment.

11. The polypeptide of claim 1 , wherein the polypeptide is coupled to polyethylene glycol (PEG).

12. The polypeptide of claim 11 , wherein the polypeptide is coupled to PEG via one or more Lys or Cys residues.

13. A nucleic acid comprising a nucleotide sequence encoding the polypeptide of claim 1 .

14. The nucleic acid of claim 13 , wherein the nucleic acid is codon optimized for expression in bacteria.

15. An expression vector comprising the nucleic acid of claim 14 .

16. A host cell comprising the nucleic acid of claim 14 .

17. The host cell of claim 16 , wherein the host cell is bacteria.

18. The host cell of claim 17 , wherein the host cell is an E. coli strain having deletions of genes ilvA and metA.

19. A pharmaceutical formulation comprising the polypeptide of claim 1 in a pharmaceutically acceptable carrier.
