Abstract

The present invention comprises the discovery and development of an effective cryoprotectant composition, without containing skim milk or any other animal-derived compounds, to achieve long-term stability of freeze-dried lactic acid bacteria (LAB), at different temperatures, whereby the retention of viability of the freeze-dried LAB after 6 months of storage, preferably after 9 months of storage, more preferably after 12 months of storage is more than 50%. The invention is in the field of producing freeze dried bacteria, in particular Lactic acid bacteria. More in particular, the invention relates to the use of a novel combination of cryoprotectants for increasing the viability of bacteria after freeze drying, improving the texture of the lyofilized cake for easy grinding and improving the long term stability of the freeze dried bacteria at different temperature conditions. The invention further relates to such freeze dried bacteria for use in food industry or in human or animal health applications. More in particular, the invention relates to the increased viability and long-term storage of recombinant bacteria capable of expressing heterologous proteins or peptides and administered to humans or animals for therapeutic or vaccination purposes.

Claims (41)

1. A composition comprising:

(a) a biomass of freeze dried Lactococcus lactis bacteria; and

(b) a combination of stabilizing compounds consisting essentially of:

(i) a starch hydrolysate selected from a dextran and a dextrin;

(ii) a glutamic acid salt; and

(iii) a polyol selected from sorbitol and mannitol,

wherein the composition is free of animal derived products, and wherein a total amount of the starch hydrolysate, the glutamic acid salt, and the polyol in a mixture comprising the Lactococcus lactis bacteria and the combination of stabilizing compounds, prior to freeze drying, is no more than 12% (w/v).

2. The composition according to claim 1 , wherein prior to freeze drying:

an amount of the starch hydrolysate is no more than 2.5% (w/v);

an amount of the glutamic acid salt is no more than 2.5% (w/v); and

an amount of the polyol is no more than 7% (w/v).

3. The composition according to claim 1 , wherein the starch hydrolysate is a dextran.

4. The composition according to claim 1 , wherein the glutamic acid salt is sodium glutamate.

5. The composition according to claim 1 , wherein the polyol is sorbitol.

6. The composition according to claim 1 , wherein the Lactococcus lactis bacteria are genetically modified.

7. The composition according to claim 1 , wherein the Lactococcus lactis bacteria comprise one or more recombinant nucleic acids which are heterologous to the Lactococcus lactis.

8. The composition according to claim 1 , wherein the Lactococcus lactis bacteria in the composition retain at least 50% viability after 12 months when stored at −20° C. or at 5° C.

9. The composition according to claim 1 , wherein the Lactococcus lactis bacteria in the composition retain at least 80% viability after 12 months when stored at −20° C. or at 5° C.

10. A culture or starter culture composition comprising the freeze dried composition according to claim 1 .

11. A medicament comprising the freeze dried composition according to claim 1 .

12. A food ingredient comprising the freeze dried composition according to claim 1 .

13. A food product or animal feed comprising the culture or the starter culture composition according to claim 11 .

14. A method for freeze drying Lactococcus lactis bacteria comprising:

(a) providing a bacterial biomass comprising Lactococcus lactis bacteria;

(b) mixing the biomass with a combination of stabilizing compounds to form a composition, wherein the combination of stabilizing compounds consists essentially of:

(i) a starch hydrolysate selected from a dextran and a dextrin,

(ii) a glutamic acid salt, and

(iii) a polyol selected from sorbitol and mannitol; and

(c) freeze drying the composition,

wherein a total amount of the starch hydrolysate, the glutamic acid salt, and the polyol is no more than 12% (w/v) in the composition prior to freeze drying, and wherein the composition is free of animal derived products.

15. The method according to claim 14 , wherein the composition prior to freeze drying comprises:

an amount of starch hydrolysate is no more than 2.5% (w/v);

an amount of glutamic acid salt is no more than 2.5% (w/v); and

an amount of polyol is no more than 7% (w/v).

16. The method according to claim 14 , wherein the starch hydrolysate is a dextran.

17. The method according to claim 14 , wherein the glutamic acid salt is sodium glutamate.

18. The method according to claim 14 , wherein the polyol is sorbitol.

19. The method according to claim 14 , wherein the Lactococcus lactis bacteria are genetically modified.

20. The method according to claim 14 , wherein the Lactococcus lactis bacteria comprise one or more recombinant nucleic acids which are heterologous to the Lactococcus lactis.

21. The method according to claim 14 , wherein the Lactococcus lactis bacteria in the freeze-dried composition retain at least 50% viability after 12 months when stored at −20° C. or at 5° C.

22. The method according to claim 14 , wherein the Lactococcus lactis bacteria in the freeze-dried composition retain at least 80% viability after 12 months when stored at −20° C. or at 5° C.
